Glutathione increase by the n-butanoyl glutathione derivative (GSH-C4) inhibits viral replication and induces a predominant Th1 immune profile in old mice infected with influenza virus
During aging, glutathione (GSH) content declines and the immune system undergoes a
deficiency in the induction of Th1 response. Reduced secretion of Th1 cytokines, which is
associated with GSH depletion, could weaken the host defenses against viral infections.
We first evaluated the concentration of GSH and cysteine in organs of old mice; then, the
effect of the administration of the N-butanoyl GSH derivative (GSH-C4) on the response of
aged mice infected with influenza A PR8/H1N1 virus was studied through the determination
of GSH concentration in organs, lung viral titer, IgA and IgG1/IgG2a production and
Th1/Th2 cytokine profile.
Old mice had lower GSH than young mice in organs. Also the gene expression of
endoplasmic reticulum (ER) stress markers involved in GSH metabolism and folding of
proteins, i.e. Nrf2 and PDI, was reduced. Following infection, GSH content remained low
and neither infection nor GSH-C4 treatment affected Nrf2 expression. In contrast, PDI
expression was upregulated during infection and appeared counterbalanced by GSH-C4.
Moreover, the treatment with GSH-C4 increased GSH content in organs, reduced viral
replication and induced a predominant Th1 response.
In conclusion, GSH-C4 treatment could be used in the elderly to contrast influenza virus
infection by inducing immune response, in particular the Th1 profile
Efficiency of transient transformation in tobacco protoplasts is independent of plasmid amount
An optimized protocol for the
transient transformation of tobacco protoplasts mediated
by polyethylene\u2013glycol (PEG) is here described. As expected, the quantitative
b\u2013glucuronidase (Gus) activity driven by pCaMVGus
was dependent on the amount of plasmid used.
Nevertheless, we demonstrate by an immunodetection
method that transformation efficiency did not depend on
the amount of plasmid used but on the limitation imposed
by cell competence. In fact, we obtained the same
percentage of transformed cells (about 60%) using a wide
range of plasmid concentrations (0.1\u201310 mg per test).
Finally, we show that, when we used two plasmid types in
a mixture at a concentration ranging from 0.1 to 10 mg for
each, all transformed cells expressed proteins encoded by
both plasmids. Transient expression and co-transformation
experiments are routinely used methods and, probably,
the major results from this work were assumed by
many researchers in this field, but our data experimentally
support this assumption
Structural, electronic, and magnetic characteristics of Np_2Co_(17)
A previously unknown neptunium-transition-metal binary compound Np_2Co_(17) has been synthesized and characterized by means of powder x-ray diffraction, ^(237)Np Mössbauer spectroscopy, superconducting-quantum-interference-device magnetometry, and x-ray magnetic circular dichroism (XMCD). The compound crystallizes in a Th_2Ni_(17)-type hexagonal structure with room-temperature lattice parameters α=8.3107(1) Å and c=8.1058(1) Å. Magnetization curves indicate the occurrence of ferromagnetic order below T_C>350 K. Mössbauer spectra suggest a Np^(3+) oxidation state and give an ordered moment of μ_(Np)=1.57(4) μ_B and μ_(Np)=1.63(4) μ_B for the Np atoms located, respectively, at the 2b and 2d crystallographic positions of the P6_3/mmc space group. Combining these values with a sum-rule analysis of the XMCD spectra measured at the neptunium M_(4,5) absorption edges, one obtains the spin and orbital contributions to the site-averaged Np moment [μ_S=−1.88(9) μ_B, μ_L=3.48(9) μ_B]. The ratio between the expectation value of the magnetic-dipole moment and the spin magnetic moment (m_(md)/μS=+1.36) is positive as predicted for localized 5f electrons and lies between the values calculated in intermediate-coupling (IC) and jj approximations. The expectation value of the angular part of the spin-orbit-interaction operator is in excellent agreement with the IC estimate. The ordered moment averaged over the four inequivalent Co sites, as obtained from the saturation value of the magnetization, is μ_(Co)≃1.6 μ_B. The experimental results are discussed against the predictions of first-principles electronic-structure calculations based on the spin-polarized local-spin-density approximation plus the Hubbard interaction

The geometrical nature of optical resonances : from a sphere to fused dimer nanoparticles
We study the electromagnetic response of smooth gold nanoparticles with shapes varying from a single sphere to two ellipsoids joined smoothly at their vertices. We show that the plasmonic resonance visible in the extinction and absorption cross sections shifts to longer wavelengths and eventually disappears as the mid-plane waist of the composite particle becomes narrower. This process corresponds to an increase of the numbers of internal and scattering modes that are mainly confined to the surface and coupled to the incident field. These modes strongly affect the near field, and therefore are of great importance in surface spectroscopy, but are almost undetectable in the far field
AzTEC 1.1 mm Observations of the MBM12 Molecular Cloud
We present 1.1 mm observations of the dust continuum emission from the MBM12
high-latitude molecular cloud observed with the Astronomical Thermal Emission
Camera (AzTEC) mounted on the James Clerk Maxwell Telescope on Mauna Kea,
Hawaii. We surveyed a 6.34 deg centered on MBM12, making this the largest
area that has ever been surveyed in this region with submillimeter and
millimeter telescopes. Eight secure individual sources were detected with a
signal-to-noise ratio of over 4.4. These eight AzTEC sources can be considered
to be real astronomical objects compared to the other candidates based on
calculations of the false detection rate. The distribution of the detected 1.1
mm sources or compact 1.1 mm peaks is spatially anti-correlated with that of
the 100 micronm emission and the CO emission. We detected the 1.1 mm
dust continuum emitting sources associated with two classical T Tauri stars,
LkHalpha262 and LkHalpha264. Observations of spectral energy distributions
(SEDs) indicate that LkHalpha262 is likely to be Class II (pre-main-sequence
star), but there are also indications that it could be a late Class I
(protostar). A flared disk and a bipolar cavity in the models of Class I
sources lead to more complicated SEDs. From the present AzTEC observations of
the MBM12 region, it appears that other sources detected with AzTEC are likely
to be extragalactic and located behind MBM12. Some of these have radio
counterparts and their star formation rates are derived from a fit of the SEDs
to the photometric evolution of galaxies in which the effects of a dusty

A new way of valorizing biomaterials: the use of sunflower protein for 1 a-tocopherol microencapsulation
Biopolymer based microparticles were efficiently prepared from sunflower protein (SP) wall material and a-tocopherol (T) active core using a spray-drying technique. Protein enzymatic hydrolysis and/or N-acylation were carried out to make some structural modifications to the vegetable protein. Native and hydrolyzed SP were characterized by Asymmetrical Flow Field-Flow Fractionation (AsFlFFF). Results of AsFlFFF confirmed that size of proteinic macromolecules was influenced by degree of hydrolysis. The effect of protein modifications and the influence of wall/core ratio on both emulsions and microparticle properties were evaluated. Concerning emulsion properties, enzymatic hydrolysis involved a decrease in viscosity, whereas acylation did not significantly affect emulsion droplet size and viscosity. Microparticles obtained with hydrolyzed SP wall material showed lower retention efficiency (RE) than native SP microparticles (62-80% and 93% respectively). Conversely, acylation of both hydrolyzed SP and native SP allowed a higher RE to be reached (up to 100%). Increasing T concentration increased emulsion viscosity, emulsion droplet size, microparticle size, and enhanced RE. These results demonstrated the feasibility of high loaded (up to 79.2% T) microparticles

Differential epigenetic reprogramming in response to specific endocrine therapies promotes cholesterol biosynthesis and cellular invasion
Endocrine therapies target the activation of the oestrogen receptor alpha (ERα) via distinct mechanisms, but it is not clear whether breast cancer cells can adapt to treatment using drug-specific mechanisms. Here we demonstrate that resistance emerges via drug-specific epigenetic reprogramming. Resistant cells display a spectrum of phenotypical changes with invasive phenotypes evolving in lines resistant to the aromatase inhibitor (AI). Orthogonal genomics analysis of reprogrammed regulatory regions identifies individual drug-induced epigenetic states involving large topologically associating domains (TADs) and the activation of super-enhancers. AI-resistant cells activate endogenous cholesterol biosynthesis (CB) through stable epigenetic activation in vitro and in vivo. Mechanistically, CB sparks the constitutive activation of oestrogen receptors alpha (ERα) in AI-resistant cells, partly via the biosynthesis of 27-hydroxycholesterol. By targeting CB using statins, ERα binding is reduced and cell invasion is prevented. Epigenomic-led stratification can predict resistance to AI in a subset of ERα-positive patients
MBM 12: young protoplanetary discs at high galactic latitude
(abridged) We present Spitzer infrared observations to constrain disc and
dust evolution in young T Tauri stars in MBM 12, a star-forming cloud at high
latitude with an age of 2 Myr and a distance of 275 pc. The region contains 12
T Tauri systems, with primary spectral types between K3 and M6; 5 are weak-line
and the rest classical T Tauri stars. We first use MIPS and literature
photometry to compile spectral energy distributions for each of the 12 members
in MBM 12, and derive their IR excesses. The IRS spectra are analysed with the
newly developed two-layer temperature distribution (TLTD) spectral
decomposition method. For the 7 T Tauri stars with a detected IR excess, we
analyse their solid-state features to derive dust properties such as
mass-averaged grain size, composition and crystallinity. We find a spatial
gradient in the forsterite to enstatite range, with more enstatite present in
the warmer regions. The fact that we see a radial dependence of the dust
properties indicates that radial mixing is not very efficient in the discs of
these young T Tauri stars. The SED analysis shows that the discs in MBM 12, in
general, undergo rapid inner disc clearing, while the binary sources have
faster discevolution. The dust grains seem to evolve independently from the
stellar properties, but are mildly related to disc properties such as flaring
