Advanced SBAS-DInSAR technique for controlling large civil infrastructures: an application to the Genzano di Lucania dam
Monitoring surface deformation on dams is commonly carried out by in situ geodetic surveying, which is time consuming and characterized by some limitations in space coverage and frequency. More recently microwave satellite-based technologies, such as advanced-DInSAR (Differential Synthetic Aperture Radar Interferometry), have allowed the integration and improvement of the observation capabilities of ground-based methods thanks to their effectiveness in collecting displacement measurements on many non-destructive control points, corresponding to radar reflecting targets. The availability of such a large number of points of measurement, which are distributed along the whole structure and are characterized by millimetric accuracy on displacement rates, can be profitably adopted for the calibration of numerical models. These models are implemented to simulate the structural behaviour of a dam under conditions of stress thus improving the ability to maintain safety standards. In this work, after having analysed how advanced DInSAR can effectively enhance the results from traditional monitoring systems that provide comparable accuracy measurements on a limited number of points, an FEM model of the Genzano di Lucania earth dam is developed and calibrated. DInSAR deformation time series for monitoring urban areas: The impact of the second generation SAR systems
We investigate the capability improvement of the DInSAR techniques to map deformation phenomena affecting urban areas, by performing a comparative analysis of the deformation time series retrieved by applying the full resolution Small BAseline Subset (SBAS) DInSAR technique to selected sequences of SAR data acquired by the ENVISAT, RADARSAT-1 and COSMO-SkyMed (CSK) SAR data. The presented study, focused on the city of Napoli (Italy), allows us to quantify the dramatic increase of the DInSAR coherent pixel density achieved by exploiting the high resolution X-Band CSK SAR images with respect to the RADARSAT-1 and ENVISAT products, respectively; this permits us to analyze nearly all the structures located within the investigated urbanized area and, in many cases, also portions of a same building. © 2012 IEEE
Testicular degeneration and infertility following arbovirus infection
Arboviruses can cause a variety of clinical signs including febrile illness, arthritis, encephalitis and hemorrhagic fever. The recent Zika epidemic highlighted the possibility that arboviruses may also negatively affect the male reproductive tract. In this study, we focused on bluetongue virus (BTV), the causative agent of bluetongue and one of the major arboviruses of ruminants. We show that rams that recovered from bluetongue displayed signs of testicular degeneration and azoospermia up to 100 days after the initial infection. Importantly, testicular degeneration was induced in rams experimentally infected with either a high (BTV-1IT2006) or low (BTV-1IT2013) virulence strain of BTV. Rams infected with the low virulent BTV strain displayed testicular lesions in the absence of other major clinical signs. Testicular lesions in BTV-infected rams were due to viral replication in the endothelial cells of the peritubular areas of the testes, resulting in stimulation of a type-I IFN response, reduction of testosterone biosynthesis by Leydig cells, and destruction of Sertoli cells and the blood-testis barrier in more severe cases. Hence, BTV induces testicular degeneration and disruption of spermatogenesis by replicating solely in the endothelial cells of the peritubular areas unlike other gonadotropic viruses. This study shows that a naturally occurring arboviral disease can cause testicular degeneration and affect male fertility at least temporarily
Treatment with embryonic stem-like cells into osteochondral defects in sheep femoral condyles
Background
Articular cartilage has poor intrinsic capacity for regeneration because of its avascularity and
very slow cellular turnover. Defects deriving from trauma or joint disease tend to be repaired with fibrocartilage rather than hyaline cartilage. Consequent degenerative processes are related to the width and depth of the defect. Since mesenchymal stem cells (MSCs) deriving from patients affected by osteoarthritis have a lower proliferative and chondrogenic activity, the systemic or local delivery of heterologous cells may enhance regeneration or inhibit the
progressive loss of joint tissue. Embryonic stem cells (ESCs) are very promising, since they can self-renew for prolonged periods without differentiation and can differentiate into tissues from all the 3 germ layers. To date only a few experiments have used ESCs for the study of
the cartilage regeneration in animal models and most of them used laboratory animals. Sheep, due to their anatomical, physiological and immunological similarity to humans, represent a valid model for translational studies. This experiment aimed to evaluate if the local delivery
of male sheep embryonic stem-like (ES-like) cells into osteochondral defects in the femoral condyles of adult sheep can enhance the regeneration of articular cartilage. Twenty-two ewes were divided into 5 groups (1, 2, 6, 12 and 24 months after surgery). Newly formed tissue was evaluated by macroscopic, histological, immunohistochemical (collagen type II) and fluorescent in situ hybridization (FISH) assays.
Results
Regenerated tissue was ultimately evaluated on 17 sheep. Samples engrafted with ES-like cells had significantly better histologic evidence of regeneration with respect to empty defects, used as controls, at all time periods.
Conclusions
Histological assessments demonstrated that the local delivery of ES-like cells into osteochondral defects in sheep femoral condyles enhances the regeneration of the articular hyaline cartilage, without signs of immune rejection or teratoma for 24 months after
engraftment.</br

a therapeutic strategy for cystic fibrosis
The inhibition of ENaC may have therapeutic potential in CF airways by
reducing sodium hyperabsorption, restoring lung epithelial surface fluid
levels, airway hydration and mucociliary function. The challenge has been to
deliver siRNA to the lung with sufficient efficacy for a sustained therapeutic
effect. We have developed a self-assembling nanocomplex formulation for siRNA
delivery to the airways that consists of a liposome (DOTMA/DOPE; L), an
epithelial targeting peptide (P) and siRNA (R). LPR formulations were assessed
for their ability to silence expression of the transcript of the gene encoding
the α-subunit of the sodium channel ENaC in cell lines and primary epithelial
cells, in submerged cultures or grown in air-liquid interface conditions.
LPRs, containing 50 nM or 100 nM siRNA, showed high levels of silencing,
particularly in primary airway epithelial cells. When nebulised these
nanocomplexes still retained their biophysical properties and transfection
efficiencies. The silencing ability was determined at protein level by
confocal microscopy and western blotting. In vivo data demonstrated that these
nanoparticles had the ability to silence expression of the α-ENaC subunit
gene. In conclusion, these findings show that LPRs can modulate the activity
of ENaC and this approach might be promising as co-adjuvant therapy for cystic
fibrosis
Sheep embryonic stem-like cells engrafted into sheep femoral condyle osteochondral defects: 4-year follow up
Deciphering Variability of PKD1 and PKD2 in an Italian Cohort of 643 Patients with Autosomal Dominant Polycystic Kidney Disease (ADPKD)
Autosomal Dominant Polycystic Kidney Disease (ADPKD) is the most common hereditary kidney disease. We analysed PKD1 and PKD2, in a large cohort of 440 unrelated Italian patients with ADPKD and 203 relatives by direct sequencing and MLPA. Molecular and detailed phenotypic data have been collected and submitted to the PKD1/PKD2 LOVD database. This is the first large retrospective study in Italian patients, describing 701 variants, 249 (35.5%) already associated with ADPKD and 452 (64.5%) novel. According to the criteria adopted, the overall detection rate was 80% (352/440). Novel variants with uncertain significance were found in 14% of patients. Among patients with pathogenic variants, in 301 (85.5%) the disease is associated with PKD1, 196 (55.7%) truncating, 81 (23%) non truncating, 24 (6.8%) IF indels, and in 51 (14.5%) with PKD2. Our results outline the high allelic heterogeneity of variants, complicated by the presence of variants of uncertain significance as well as of multiple variants in the same subject. Classification of novel variants may be particularly cumbersome having an important impact on the genetic counselling. Our study confirms the importance to improve the assessment of variant pathogenicity for ADPKD; to this point databasing of both clinical and molecular data is crucial
