How to create and detect N-dimensional entangled photons with an active phase hologram
The experimental realization of multidimensional quantum states may lead to
unexplored and interesting physics, as well as advanced quantum communication
protocols. The orbital angular momentum of photons is a well suitable discrete
degree of freedom for implementing high-dimensional quantum systems. The
standard method to generate and manipulate such photon modes is to use bulk and
fixed optics. Here the authors demonstrate the utilization of a spatial light
modulator to manipulate the orbital angular momentum of entangled photons
generated in spontaneous parametric downconversion. They show that their setup
allows them to realize photonic entanglement of up to 21 dimensions, which in
principle can be extended to even larger dimensions
Oncogenic K-Ras suppresses IP<sub>3</sub>-dependent Ca<sup>2+</sup> release through remodeling of IP<sub>3</sub>Rs isoform composition and ER luminal Ca<sup>2+</sup> levels in colorectal cancer cell lines
The GTPase Ras is a molecular switch engaged downstream of G-protein coupled receptors and receptor tyrosine inases that controls multiple cell fate-determining signalling athways. Ras signalling is frequently deregulated in cancer underlying associated changes in cell phenotype. Although Ca2+ signalling pathways control some overlapping functions with Ras, and altered Ca2+ signalling pathways are emerging as important players in oncogenic transformation, how Ca2+ signalling is remodelled during transformation and whether it has a causal role remains unclear. We have investigated Ca2+ signalling in two human colorectal cancer cell lines and their isogenic derivatives in which the mutated K-Ras allele (G13D) has been deleted by homologous recombination. We show that agonist-induced Ca2+ release from intracellular stores is enhanced by loss of K-RasG13D through an increase in the ER store content and a modification of IP3R subtype abundance. Consistently, uptake of Ca2+ into mitochondria and sensitivity to apoptosis was enhanced as a result of KRasG13D loss. These results suggest that suppression of Ca2+ signalling is a common response to naturally occurring levels of K-RasG13D that contributes to a survival
advantage during oncogenic transformation
Spatio-temporal malaria transmission patterns in Navrongo demographic surveillance site, northern Ghana
The relationship between entomological measures of malaria transmission intensity and mortality remains uncertain. This is partly because transmission is heterogeneous even within small geographical areas. Studying this relationship requires high resolution, spatially structured, longitudinal entomological data. Geostatistical models that have been used to analyse the spatio-temporal heterogeneity have not considered the uncertainty in both sporozoite rate (SR) and mosquito density data. This study analysed data from Kassena-Nankana districts in northern Ghana to obtain small area estimates of malaria transmission rates allowing for this uncertainty.; Independent Bayesian geostatistical models for sporozoite rate and mosquito density were fitted to produce explicit entomological inoculation rate (EIR) estimates for small areas and short time periods, controlling for environmental factors.; Mosquitoes were trapped from 2,803 unique locations for three years using mainly CDC light traps. Anopheles gambiae constituted 52%, the rest were Anopheles funestus. Mean biting rates for An. funestus and An. gambiae were 32 and 33 respectively. Most bites occurred in September, the wettest month. The sporozoite rates were higher in the dry periods of the last two years compared with the wet period. The annual EIR varied from 1,132 to 157 infective bites. Monthly EIR varied between zero and 388 infective bites. Spatial correlation for SR was lower than that of mosquito densities.; This study confirms the presence of spatio-temporal heterogeneity in malaria transmission within a small geographical area. Spatial variance was stronger than temporal especially in the SR. The estimated EIR will be used in mortality analysis for the area
Circlator: automated circularization of genome assemblies using long sequencing reads
The assembly of DNA sequence data is undergoing a renaissance thanks to emerging technologies capable of producing reads tens of kilobases long. Assembling complete bacterial and small eukaryotic genomes is now possible, but the final step of circularizing sequences remains unsolved. Here we present Circlator, the first tool to automate assembly circularization and produce accurate linear representations of circular sequences. Using Pacific Biosciences and Oxford Nanopore data, Circlator correctly circularized 26 of 27 circularizable sequences, comprising 11 chromosomes and 12 plasmids from bacteria, the apicoplast and mitochondrion of Plasmodium falciparum and a human mitochondrion. Scholarly insight Autumn 2017:a Data wrangler perspective
As the OU is going through several fundamental changes, it is important that strategic decisions made by Faculties and senior management are informed by evidence-based research and insights. One way how Data Wranglers provide insights of longitudinal development and performance of OU modules is the Key Metric Report 2017. A particular new element is that data can now also be unpacked and visualised on a Nation-level. As evidenced by the Nation-level reporting, there are substantial variations of success across the four Nations, and we hope that our interactive dashboards allow OU staff to unpack the underlying data.
The second way Data Wranglers provide insight to Faculties and Units is through the Scholarly insight report series. Building on the previous two reports whereby we reported on substantial variation and inconsistencies in learning designs and assessment practices within qualifications across the OU, in this Scholarly insight Autumn 2017 report we address four big pedagogical questions that were framed and co-constructed together with the Faculties and LTI units. Many Faculties and colleagues have reacted positively on our Scholarly insight Spring 2017 report, whereby for the first time we were able to show empirically that students experienced substantial variations in success within 12 large OU qualifications. As evidenced in our previous report, 55% of variation in students’ success over time was explained by OU institutional factors (i.e., how students were assessed within their respective module; how students were able to effectively transition from one learning design of one module to the next one), rather than students’ characteristics, engagement and behaviour.
We have received several queries and questions from Faculties and Units about how to better understand these students’ journeys, and how qualifications and module designs could be better aligned within their respective qualification(s). As these are complex conceptual and Big Pedagogy questions, in Chapter 1 we continued these complex analyses by looking at the transitional processes of the first two modules that OU students take, and how well aligned these modules and qualification paths are. In Chapter 2, we explored the more fine-grained, qualitative, and lived experiences of 19 students across a range of qualifications to understand how OU grading practices and (in)consistencies of assessment and feedback influenced their affect, behaviour, and cognition. In addition to building on previous topics, we introduced two new Scholarly insights in Chapter 3 and Chapter 4. As the OU is increasingly using learning analytics to support our staff and students, in Chapter 3 we analysed the impact of giving Predictive Learning Analytics to over 500 Associate Lecturers across 31 modules on student retention. Finally, in Chapter 4 we explored the impact of first presentations of new modules on pass rates and satisfaction, whereby we were able to bust another myth that may have profound implications for Student First Transformation.
Working organically in various Faculty sub-group meetings and LTI Units and in a google doc with various key stakeholders in the Faculties , we hope that our Scholarly insights can help to inform our staff, but also spark some ideas how to further improve our module designs and qualification pathways.
