    Occult Cushing\u27s Syndrome Presenting with Osteoporosis

    Osteoporosis is a frequent complication both of endogenous hypercortisolism and of long-term treatment with corticosteroids, but only rarely is it the major clinical feature with the more characteristic features absent or minimally present. In the two patients presented, hypercortisolism was uncovered only during routine evaluation of osteoporosis. This presentation is probably due to slow progression of the disease and is often associated with so-called black adenoma of the adrenal gland. Secondary causes should be sought in all patients with seemingly senile or postmenopausal osteoporosis

    Rational Design of a Biocatalyst Based on Immobilized CALB onto Nanostructured SiO2

    The adsorption of the lipase B from Candida antarctica (CALB) over nanostructured SiO2 (Ns SiO2 from now on) with and without the addition of polyols (sorbitol and glycerol) was investigated. The isotherms of adsorption made it possible to establish that the maximum dispersion limit was 0.029 µmol of protein per surface area unit of Ns SiO2 (29.4 mg per 100 mg of support), which was reached in 30 min of exposure. The studies through SDS-PAGE of the immobilization solutions and infrared spectroscopy of the prepared solids determined that CALB (from a commercial extract) is selectively adsorbed, and its secondary structure distribution is thus modified. Its biocatalytic activity was corroborated through the kinetic resolution of rac-ibuprofen. Conversions of up to 70% and 52% enantiomeric excess toward S-ibuprofen in 24 h of reaction at 45 °C were achieved. The biocatalytic performance increased with the increase in protein loading until it leveled off at 0.021 µmol.m−2, reaching 0.6 µmol.min−1. The biocatalyst containing the lipase at the maximum dispersion limit and co-adsorbed polyols presented the best catalytic performance in the kinetic resolution of rac-ibuprofen, an improved thermal resistance (up to 70 °C), and stability under long-term storage (more than 2 years).Fil: Llerena Suster, Carlos Rafael.     Structural biology of STAT3 and its implications for anticancer therapies development

    Transcription factors are proteins able to bind DNA and induce the transcription of specific genes. Consequently, they play a pivotal role in multiple cellular pathways and are frequently over-expressed or dysregulated in cancer. Here, we will focus on a specific “signal transducer and activator of transcription” (STAT3) factor that is involved in several pathologies, including cancer. For long time, the mechanism by which STAT3 exerts its cellular functions has been summarized by a three steps process: (1) Protein phosphorylation by specific kinases, (2) dimerization promoted by phosphorylation, (3) activation of gene expression by the phosphorylated dimer. Consequently, most of the inhibitors reported in literature aimed at blocking phosphorylation and dimerization. However, recent observations reopened the debate and the entire functional mechanism has been revisited stimulating the scientific community to pursue new inhibition strategies. In particular, the dimerization of the unphosphorylated species has been experimentally demonstrated and specific roles proposed also for these dimers. Despite difficulties in the expression and purification of the full length STAT3, structural biology investigations allowed the determination of atomistic structures of STAT3 dimers and several protein domains. Starting from this information, computational methods have been used both to improve the understanding of the STAT3 functional mechanism and to design new inhibitors to be used as anticancer drugs. In this review, we will focus on the contribution of structural biology to understand the roles of STAT3, to design new inhibitors and to suggest new strategies of pharmacological intervention

    Enhanced photocatalytic degradation of RO16 dye using Ag modified ZnO nanopowders prepared by the solvothermal method

    In this work, Zn(CH3 COO)(2) 2 H-2 O with AgNO3 content from 0 to 6 mol% was solvothermally treated at 120 C for 18 h in the presence of poly(vinyl pyrrolidone), ethylene glycol and sodium hydroxide. The structural , microstructural and photocatalytic properties of the unmodified and Ag modified ZnO powders have been investigated by the XRPD, FESEM, TEM, UV-vis, Raman and BET techniques. The Ag modified samples consist of ZnO nanocrystals and metallic Ag on the surface. The average crystallite size of all samples was about 20 nm. The FESEM revealed the uniformity in size and approximately spherical shape of ZnO nanopar-ticles. The BET data suggest that all prepared samples are mesoporous. All prepared samples showed higher photocatalytic efficiency in the degradation of the Reactive Orange 16 (RO16) azo dye than the commercial ZnO. In addition, Ag modified ZnO powders, especially those with 1.5 and 0.75 mol% of Ag, were more efficient than the unmodified one

    Point cloud subjective evaluation methodology based on reconstructed surfaces

    Point clouds have been gaining importance as a solution to the problem of efficient representation of 3D geometric and visual information. They are commonly represented by large amounts of data, and compression schemes are important for their manipulation transmission and storing. However, the selection of appropriate compression schemes requires effective quality evaluation. In this work a subjective quality evaluation of point clouds using a surface representation is analyzed. Using a set of point cloud data objects encoded with the popular octree pruning method with different qualities, a subjective evaluation was designed. The point cloud geometry was presented to observers in the form of a movie showing the 3D Poisson reconstructed surface without textural information with the point of view changing in time. Subjective evaluations were performed in three different laboratories. Scores obtained from each test were correlated and no statistical differences were observed. Scores were also correlated with previous subjective tests and a good correlation was obtained when compared with mesh rendering in 2D monitors. Moreover, the results were correlated with state of the art point cloud objective metrics revealing poor correlation. Likewise, the correlation with a subjective test using a different representation of the point cloud data also showed poor correlation. These results suggest the need for more reliable objective quality metrics and further studies on adequate point cloud data representations

    Peak bone mineral density in Vietnamese women

    While the prevalence of osteoporosis and risk factors for low bone mineral density (BMD) has been well documented in Caucasian populations, there is a lack of data from Asia. This work was designed to clarify to what extent osteoporosis could be regarded as a major public health problem in Vietnam. Furthermore, to elucidate the prevalence of certain risk factors, such as vitamin D deficiency and other determinants of bone mass as a basis to indentify high-risk individuals among the Vietnamese women and men. The clinical studies were designed as cross-sectional investigations using a multistage sampling scheme. Within the setting of northern Vietnam (latitude 21°N), districts were selected to represent urban and rural areas. In total 612 healthy women and 222 men aged 13-83 years were investigated. BMD was measured at the lumbar spine, femoral neck and total hip in all qualified subjects with dual energy X-ray absortiometry. Serum concentrations of 25(OH)D, parathyroid hormone, estrogen and testosterone were quantified by electrochemiluminescence immunoassay. Data on clinical history and lifestyle were collected by individual face-to-face interviews. Reference values for peak BMD were defined. These data allowed the calculation of T-scores and thus for the first time, an accurate identification of osteoporosis in a Vietnamese population. As determined at the femoral neck, the prevalence of osteoporosis was 17-23% in women and 9% in men. The results clearly suggest that osteoporosis is an important public health problem. Postmenopausal women living in urban areas experienced osteoporosis more than rural residents. Serum levels of 25(OH)D and estrogen were significantly associated with bone mass in both women and men. The prevalence of vitamin D deficiency (<20 ng/mL) was very high, 30% in women and 16% in men. An experimental study on the isoflavone content of different soymilk preparations was performed by HPLC (high pressure liquid chromatography). Values of isoflavones were very low, around 60-80 mg/L, and there were only 10-20% of bioactive aglycones. This is far below the reported threshold levels to exert significant effects on bone. In the future these data will be useful as a valuable reference base to diagnose osteoporosis and for the clinical management of its consequences. The high prevalence of vitamin D deficiency should raise the awareness of potentially important health issues such as osteoporosis but also about other serious diseases within the Vietnamese society

    Sugar-added beverages consumption among kindergarten children of Crete: effects on nutritional status and risk of obesity

    <p>Abstract</p> <p>Objective</p> <p>To assess the intake of sugar-added beverages such as soft drinks and commercially available fruit juices in kindergarten children, and to examine its association with obesity indices, physical activity levels and dietary habits.</p> <p>Methods</p> <p>A total of 856 children aged 4–7 years living in Crete, Greece in 2004–5 were included in this cross-sectional study. Nutrient and food intake was assessed with the use of 3-day weighed food records. Body measurements were used in order to assess BMI and waist circumference, and moderate-to-vigorous physical activity was calculated with the use of a questionnaire.</p> <p>Results</p> <p>Approximately 59.8% of all children consumed sugar-added beverages on a daily basis. High intake of sugar-added beverages (> 250 g/day) was associated with low intakes of calcium (p < 0.001), vitamin A and E (p < 0.010), fruits and vegetables (p = 0.007), and milk and yogurt (p = 0.048). Compared to non or low consumers, high consumers of sugar-added beverages (> 250 g/day) had higher BMI levels and two times greater risk of being overweight and/or obese (OR:2.35, p = 0.023).</p> <p>Conclusion</p> <p>High intake of sugar-added beverages in kindergarten children is associated with poor eating habits and inadequate nutrient intake, as well as increased risk for developing childhood obesity.</p
