    Studies of Relativistic Jets in Active Galactic Nuclei with SKA

    Relativistic jets in active galactic nuclei (AGN) are among the most powerful astrophysical objects discovered to date. Indeed, jetted AGN studies have been considered a prominent science case for SKA, and were included in several different chapters of the previous SKA Science Book (Carilli & Rawlings 2004). Most of the fundamental questions about the physics of relativistic jets still remain unanswered, and await high-sensitivity radio instruments such as SKA to solve them. These questions will be addressed specially through analysis of the massive data sets arising from the deep, all-sky surveys (both total and polarimetric flux) from SKA1. Wide-field very-long-baseline-interferometric survey observations involving SKA1 will serve as a unique tool for distinguishing between extragalactic relativistic jets and star forming galaxies via brightness temperature measurements. Subsequent SKA1 studies of relativistic jets at different resolutions will allow for unprecedented cosmological studies of AGN jets up to the epoch of re-ionization, enabling detailed characterization of the jet composition, magnetic field, particle populations, and plasma properties on all scales. SKA will enable us to study the dependence of jet power and star formation on other properties of the AGN system. SKA1 will enable such studies for large samples of jets, while VLBI observations involving SKA1 will provide the sensitivity for pc-scale imaging, and SKA2 (with its extraordinary sensitivity and dynamic range) will allow us for the first time to resolve and model the weakest radio structures in the most powerful radio-loud AGN.Comment: 19 pages, 4 figures; to appear as part of 'Cosmic Magnetism' in Proceedings 'Advancing Astrophysics with the SKA (AASKA14)', PoS(AASKA14_093

    Rapid detection of Mycobacterium tuberculosis by recombinase polymerase amplification.

    Improved access to effective tests for diagnosing tuberculosis (TB) has been designated a public health priority by the World Health Organisation. In high burden TB countries nucleic acid based TB tests have been restricted to centralised laboratories and specialised research settings. Requirements such as a constant electrical supply, air conditioning and skilled, computer literate operators prevent implementation of such tests in many settings. Isothermal DNA amplification technologies permit the use of simpler, less energy intensive detection platforms more suited to low resource settings that allow the accurate diagnosis of a disease within a short timeframe. Recombinase Polymerase Amplification (RPA) is a rapid, low temperature isothermal DNA amplification reaction. We report here RPA-based detection of Mycobacterium tuberculosis complex (MTC) DNA in <20 minutes at 39 °C. Assays for two MTC specific targets were investigated, IS6110 and IS1081. When testing purified MTC genomic DNA, limits of detection of 6.25 fg (IS6110) and 20 fg (IS1081)were consistently achieved. When testing a convenience sample of pulmonary specimens from suspected TB patients, RPA demonstrated superior accuracy to indirect fluorescence microscopy. Compared to culture, sensitivities for the IS1081 RPA and microscopy were 91.4% (95%CI: 85, 97.9) and 86.1% (95%CI: 78.1, 94.1) respectively (n = 71). Specificities were 100% and 88.6% (95% CI: 80.8, 96.1) respectively. For the IS6110 RPA and microscopy sensitivities of 87.5% (95%CI: 81.7, 93.2) and 70.8% (95%CI: 62.9, 78.7) were obtained (n = 90). Specificities were 95.4 (95% CI: 92.3,98.1) and 88% (95% CI: 83.6, 92.4) respectively. The superior specificity of RPA for detecting tuberculosis was due to the reduced ability of fluorescence microscopy to distinguish Mtb complex from other acid fast bacteria. The rapid nature of the RPA assay and its low energy requirement compared to other amplification technologies suggest RPA-based TB assays could be of use for integration into a point-of-care test for use in resource constrained settings

    Geoarchaeology has a key role to play in the human-climate-environment debate, including research into the controls and variability of water availability. However its interdisciplinarity can cause complications as well as advantages and leads to confusion as to what researchers should be investigating and how they should be reporting their findings. In this paper I investigate Geoarchaeology, and its place in 21st century science, as well as investigating its role in reconstructing past water availability and some of the challenges involved in ensuring these investigations are undertaken in a robust manner that will enhance academic knowledge and debate

    How accurately do adult patients report their absence seizures?

    We depend upon self-reporting to determine seizure frequency for epilepsy management decisions, but people often misreport their seizures. Here, we determined misreporting rates in adults with absence seizures, undergoing inpatient video-EEG telemetry (VET) or outpatient ambulatory electroencephalography (aEEG). Under-reporting rates were based on VET data, where behavior could be assessed, whilst over-reporting was assessed using both VET and aEEG. Forty-two patients (31 female and 11 males, median age 28.5 years) and 759 reported absence seizures were included in this study. Overall, only 24% of the 759 reported seizures had an associated EEG correlate, indicating a high over-reporting rate, which occurred in 57% of patients. Age, sex, time of epilepsy, VET versus aEEG, epilepsy syndrome or medication were not significant predictors of over-reporting. In the VET group in which we could assess both over- and under-reporting (22 patients), only 2 patients correctly reported their seizures, and patients were predominantly over-reporters or under reporters, not both. Only 26% of 423 absence seizures were reported. Use of zonisamide or valproate was associated with under-reporting, possibly through an impact on attention. These findings indicate that self-reported AS are a poor measure to use for treatment decisions due to both over- and under-reporting

    Understanding the non-professional needs of early career doctors: An interview-based study

    Speciality colleges and health services are often well attuned to professional factors, but non-professional needs are less acknowledged and are the focus of this study. This likely relates to limited research about the non-professional needs of early career doctors. This study aimed to describe the non-professional needs of doctors in their early postgraduate career, including how they intersect with career and training experiences. Semi-structured interviews were conducted with 32 male and female medical graduates working across all Australian states and territories, spanning a variety of speciality areas and early career stages. Participants were asked about their career journey to date including non-professional factors related to their experiences. This study identified important non-professional needs, that strongly interplayed with career and training experiences, including: children's education; partner's career needs; family stability; major life stages; proximity to the extended family; and spending time with immediate family. Results suggested clear gender differences, with female doctor's needs orientated to partner work and carer responsibilities, while male doctor's needs were oriented to spending time with family and meeting the family's needs. Non-professional needs should be considered as legitimate needs within health service employment and speciality training arrangements enabling early career doctors to realise their full potential

    Representational predicaments at three Hong Kong sites

    Representational predicaments arise when a job incumbent believes that attributions and images assumed by dominant authorities unfavourably ignore, or disproportionately and unfavourably emphasize, aspects of the incumbent\u27s own work and social identity. This is likely to happen when the incumbent does not have a close relationship with a dominant authority, and when power asymmetries give the former relatively little control over which aspects of their work and social identity are made visible or invisible to the latter. We draw on critical incident interviews from three organizations to illustrate a typology of six types of representational predicament: invasive spotlighting, idiosyncratic spotlighting, embedded background work, paradoxical social visibility, standardization of work processes, and standardization of work outputs. We analyse responses to representational predicaments according to whether they entailed exit, voice, loyalty, or neglect. Incumbents tended to respond with loyalty if they felt able and willing to accommodate their work behaviour and/or social identity to the dominant representations, and if there were sufficient compensatory factors, such as intrinsic rewards from the work or solidarity with colleagues. Exit or neglect appeared to reflect the belief that it was impossible to accommodate. Power asymmetries appeared to deter voice. Individual employees with a close and cordial working relationship with a member of a dominant authority group, or who were relationally networked to one, appeared not to experience representational predicaments

    Vancomycin AUC/MIC ratio and 30-day mortality in patients with Staphylococcus aureus bacteremia

    A ratio of the vancomycin area under the concentration-time curve to the MIC (AUC/MIC) of ≥ 400 has been associated with clinical success when treating Staphylococcus aureus pneumonia, and this target was recommended by recently published vancomycin therapeutic monitoring consensus guidelines for treating all serious S. aureus infections. Here, vancomycin serum trough levels and vancomycin AUC/MIC were evaluated in a "real-world" context by following a cohort of 182 patients with S. aureus bacteremia (SAB) and analyzing these parameters within the critical first 96 h of vancomycin therapy. The median vancomycin trough level at this time point was 19.5 mg/liter. There was a significant difference in vancomycin AUC/MIC when using broth microdilution (BMD) compared with Etest MIC (medians of 436.1 and 271.5, respectively; P373, derived using classification and regression tree analysis, was associated with reduced mortality (P=0.043) and remained significant in a multivariable model. This study demonstrated that we obtained vancomycin trough levels in the target therapeutic range early during the course of therapy and that obtaining a higher vancomycin AUC/MIC (in this case, >373) within 96 h was associated with reduced mortality. The MIC test method has a significant impact on vancomycin AUC/MIC estimation. Clinicians should be aware that the current target AUC/MIC of ≥400 was derived using the reference BMD method, so adjustments to this target need to be made when calculating AUC/MIC ratio using other MIC testing methods.     Towards a graphene transparent conducting electrode for perovskite/silicon tandem solar cells

    Indium-based transparent conducting electrodes (TCEs) are a major limiting factor in perovskite/silicon tandem cell scalability, while also limiting maximum cell efficiencies. In this work, we propose a novel TCE based on electrostatically doped graphene monolayers to circumvent these challenges. The electrode is enabled by a thin film dielectric that is charged and interfaced to a graphene film, optimally exploiting electrostatic doping. The field effect mechanism allows the modulation of charge carriers in monolayer graphene as a function of charge concentration in the dielectric thin film. Electrostatic charge was deposited on SiO2 membranes, and graphene transferred onto them exhibited a reduction in sheet resistance because of the induced charge carriers. We show a reduction in sheet resistance of graphene by 60% in just 3 min of dielectric charging, without impacting the transmission of light through the film stack. Hall effect measurements indicated that the mobility of the films was not significantly degraded. The deposition of negative electrostatic charge reversed this effect, allowing for precise tunability of charge concentration from n- to p-type. We develop a model to determine the required sheet resistance of a graphene TCE with 97% transmittance in a perovskite/silicon tandem cell. As the technique here reported does not impact transmittance, a graphene TCE with a sheet resistance below 50 Ω/□ could enable efficiencies up to 44%, presenting a promising alternative to indium-based TCEs
