Impact of temperature on the pullout of reinforcing geotextiles from unsaturated silt
This study investigates the thermal soil-geosynthetic interaction mechanisms of reinforcing geotextiles confined in compacted silt that may be encountered when using mechanically-stabilized earth (MSE) walls as geothermal heat sinks. A thermo-mechanical geosynthetic pullout device was used that incorporates standard components for geosynthetic pullout or creep testing but also heating elements at the top and bottom of the soil box to apply boundary temperatures and dielectric sensors embedded in the soil layer to monitor distributions in temperature and volumetric water content. Two test series were performed: the first involves monotonic pullout of woven polypropylene geotextiles after reaching steady-state conditions under different boundary temperatures without a seating load, and the second involves monotonic pullout of woven polyethylene-terephthalate geotextiles after reaching steady-state conditions under different boundary temperatures with a seating pullout load. The results indicate that the pullout resistance of both geotextiles decreased with increasing temperature. Although heating led to drying of the unsaturated silt layers as expected, measurements from the second test series indicate accumulation of water at the silt-geotextile interface. An effective stress analysis considering thermal softening of soils indicates that the increase in effective saturation at the silt-geotextile interface was the cause of the decrease in pullout resistance with heating

Assessment of approaches to obtain ebullition pressures for organophilic clay blankets
The objective of this study is to compare two experimental approaches to characterize the ebullition pressure (or air-entry suction) of initially water-saturated organophilic clay blankets. The first is an indirect approach using the water-retention curve (WRC) and the second is a direct approach using ebullition experiments. The WRC along with the hydraulic conductivity of organophilic clay blankets in saturated and unsaturated conditions were measured using a flexible-wall permeameter with suction-saturation control. This device was also adapted to measure the ebullition pressure and the air permeability. The comparison of the experimental approaches was performed on organophilic clay blanket specimens in different initial conditions (unrinsed and rinsed to remove loose fines) under high and low effective confining stresses (20 and 5 kPa). The indirect estimates of air-entry suction from the WRC were similar to those obtained from the ebullition tests. This good agreement between the two approaches may add flexibility to the development of design specifications for capping systems. The hydraulic properties were found to be sensitive to rinsing and effective stress, with greater hydraulic conductivity and air permeability for the rinsed specimen due to the removal of fines, and greater air-entry suctions for specimens under higher effective stress
Dual requirement of cytokine and activation receptor triggering for cytotoxic control of murine cytomegalovirus by NK cells
Natural killer (NK) cells play a critical role in controlling murine cytomegalovirus (MCMV) and can mediate both cytokine production and direct cytotoxicity. The NK cell activation receptor, Ly49H, is responsible for genetic resistance to MCMV in C57BL/6 mice. Recognition of the viral m157 protein by Ly49H is sufficient for effective control of MCMV infection. Additionally, during the host response to infection, distinct immune and non-immune cells elaborate a variety of pleiotropic cytokines which have the potential to impact viral pathogenesis, NK cells, and other immune functions, both directly and indirectly. While the effects of various immune deficiencies have been examined for general antiviral phenotypes, their direct effects on Ly49H-dependent MCMV control are poorly understood. To specifically interrogate Ly49H-dependent functions, herein we employed an in vivo viral competition approach to show Ly49H-dependent MCMV control is specifically mediated through cytotoxicity but not IFNγ production. Whereas m157 induced Ly49H-dependent degranulation, efficient cytotoxicity also required either IL-12 or type I interferon (IFN-I) which acted directly on NK cells to produce granzyme B. These studies demonstrate that both of these distinct NK cell-intrinsic mechanisms are integrated for optimal viral control by NK cells
The provision of NHS health checks in a community setting: an ethnographic account
Background: The UK National Health Service Health Checks programme aims to reduce avoidable cardiovascular deaths, disability and health inequalities in England. However, due to the reported lower uptake of screening in specific black and minority ethnic communities who are recognised as being more at risk of cardiovascular disease, there are concerns that NHS Health Checks may increase inequalities in health. This study aimed to examine the feasibility and acceptability of community outreach NHS Health Checks targeted at the Afro-Caribbean community.
Methods: This paper reports findings from an ethnographic study including direct observation of four outreach events in four different community venues in inner-city Bristol, England and follow up semi-structured interviews with attendees (n = 16) and staff (n = 4). Interviews and field notes were transcribed, anonymized and analysed thematically using a process of constant comparison.
Results: Analysis revealed the value of community assets (community engagement workers, churches, and community centres) to publicise the event and engage community members. People were motivated to attend for preventative reasons, often prompted by familial experience of cardiovascular disease. Attendees valued outreach NHS Health Checks, reinforcing or prompting some to make healthy lifestyle changes. The NHS Health Check provided an opportunity for attendees to raise other health concerns with health staff and to discuss their test results with peers. For some participants, the communication of test results, risk and lifestyle information was confusing and unwelcome. The findings additionally highlight the need to ensure community venues are fit for purpose in terms of assuring confidentiality.
Conclusions: Outreach events provide evidence of how local health partnerships (family practice staff and health trainers) and community assets, including informal networks, can enhance the delivery of outreach NHS Health Checks and in promoting the health of targeted communities. To deliver NHS Health Checks effectively, the location and timing of events needs to be carefully considered and staff need to be provided with the appropriate training to ensure patients are supported and enabled to make lifestyle changes
Quantifying soil hydrology to explain the development of vegetation at an ex-arable wetland restoration site
Wetland restoration frequently sets well-defined vegetation targets, but where restoration occurs on highly degraded land such targets are not practical and setting looser targets may be more appropriate. Where this more ‘open-ended’ approach to restoration is adopted, surveillance methods that can track developing wetland habitats need to be established. Water regime and soil structure are known to influence the distribution and composition of developing wetland vegetation, and may be quantified using Sum Exceedence Values (SEV), calculated using the position of the water table and knowledge of soil stress thresholds. Use of SEV to explain patterns in naturally colonizing vegetation on restored, ex-arable land was tested at Wicken Fen (UK). Analysis of values from ten locations showed that soil structure was highly heterogeneous. Five locations had shallow aeration stress thresholds and so had the potential to support diverse wetland assemblages. Deep aeration stress thresholds at other locations precluded the establishment of a diverse wetland flora, but identified areas where species-poor wetland assemblages may develop. SEV was found to be a useful tool for the surveillance of sites where restoration targets are not specified in detail at the outset and may help predict likely habitat outcomes at sites using an open-ended restoration approach

An in vitro approach to study effects of prebiotics and probiotics on the faecal microbiota and selected immune parameters relevant to the elderly
The aging process leads to alterations of gut microbiota and modifications to the immune response, such changes may be associated with increased disease risk. Prebiotics and probiotics can modulate microbiome changes induced by aging; however, their effects have not been directly compared. The aim of this study was to use anaerobic batch culture fermenters to assess the impact of various fermentable carbohydrates and microorganisms on the gut microbiota and selected immune markers. Elderly volunteers were used as donors for these experiments to enable relevance to an aging population. The impact of fermentation supernatants on immune markers relevant to the elderly were assessed in vitro. Levels of IL-1β, IL-6, IL-8, IL-10 and TNF-α in peripheral blood mononuclear cell culture supernatants were measured using flow cytometry. Trans-galactooligosaccharides (B-GOS) and inulin both stimulated bifidobacteria compared to other treatments (p<0.05). Fermentation supernatants taken from faecal batch cultures supplemented with B-GOS, inulin, B. bifidum, L. acidophilus and Ba. coagulans inhibited LPS induced TNF-α (p<0.05). IL-10 production, induced by LPS, was enhanced by fermentation supernatants from faecal batch cultures supplemented with B-GOS, inulin, B. bifidum, L. acidophilus, Ba. coagulans and Bac. thetaiotaomicron (p<0.05). To conclude, prebiotics and probiotics could lead to potentially beneficial effects to host health by targeting specific bacterial groups, increasing saccharolytic fermentation and decreasing inflammation associated with aging. Compared to probiotics, prebiotics led to greater microbiota modulation at the genus level within the fermenters
