    A Study of Participation Rate, Transparency and Accountability of Special Autonomy Funds Management in Jayapura

    The aim of this study is to identify and review financial planning and management issues from Papua's Special Autonomy Funds in the fields of education and health over a period of 5 years (2008-2012) in Jayapura and to assess what issues of public concern in that planning and management. The observed object of study is the input, output and outcome of the implementation of the planning and budgeting of Special Autonomy funds related to the services for indigenous people in the education and health sectors in Jayapura. The population in this study is the local work units (SKPD) that manage the funds in both sectors and legislature in the government of Jayapura. The sampling techniques are non-multistage random sampling, non-random and purposive sampling. Data collection is through (1) Questionnaire. (2) Interviews (3) Study of Literature (4) Focus Group Discussion. All the collected date was analysed through the descriptive statistics and case studies. The results show that the medical staff at the health centres (Puskesmas dan Pustu), teachers and principals at the education centres as well as society consider that that level of participation, transparency and accountability in the management of public funds is less satisfactory for indigenous Papuans. Planning through village and district’s development planning meetings (Musrenbang) do not get involved health and educational institutions. The community is more involved in the planning process of the Strategic Plans of Village Development (Respek) Funds than the use of special autonomy funds; therefore the information about spending of Respek funds is easier obtained than the government spending on the special autonomy fund. In the compilation of the Proposed Definitive Plan, Development Planning Agency (Bapeda) need to get involved all SKPDs as fund users and the discussion of the Plan needs more consistency in funds allocation of the priority sectors as indicated in the Law 21 of 2001

    Expression of a novel gene, gluP, is essential for normal Bacillus subtilis cell division and contributes to glucose export

    BACKGROUND: The Bacillus subtilis glucokinase operon was predicted to be comprised of the genes, yqgP (now named gluP), yqgQ, and glcK. We have previously established a role for glcK in glucose metabolism. In the absence of enzymes that phosphorylate glucose, such as GlcK and/or enzyme II(Glc), accumulated cytoplasmic glucose can be transported out of the cell. Genes within the glucokinase operon were not previously known to play a role in glucose transport. Here we describe the expression of gluP and its function in glucose transport. RESULTS: We found that transcription of the glucokinase operon was regulated, putatively, by two promoters: σ(A )and σ(H). Putative σ(A )and σ(H)-recognition sites were located upstream of and within gluP, respectively. Transcriptional glucokinase operon – lacZ fusions and Northern blotting were used to analyze the expression of gluP. GluP was predicted to be an integral membrane protein. Moreover, the prediction of GluP structure revealed interesting signatures: a rhomboid domain and two tetracopeptide repeat (TPR) motifs. Microscopic analysis showed that GluP minus cells were unable to divide completely, resulting in a filamentous phenotype. The cells were grown in either rich or minimal medium. We found GluP may be involved in glucose transport. [(14)C]-glucose uptake by the GluP minus strain was slightly less than in the wild type. On the other hand, trehalose-derived glucose in the growth medium of the GluP minus strain was detected in very low amounts. Experimental controls comprised of single or multiple genes mutations within the glucose transporting phosphotransferase system. CONCLUSIONS: gluP seems to be regulated only by a putative σ(A)-dependent promoter. The glucose uptake and export assays suggest that GluP is important for glucose export and may act as an exporter. This also supports the role of the glucokinase operon in glucose utilization

    Studi Dampak Covid 19 Terhadap Perekonomian Provinsi Papua 2020

    Kajian ini bertujuan untuk mendeskripsikan dampak COVID-19 terhadap kinerja ekonomi kesempatan kerja dan harga pada 9 bahan pokok serta terhadap pemanfatan tekhnolog dalam proses Belajar mengajar. Lokasi kajian di Provinsi Papua dengan wilayah sampel kota Jayapura, dengan alasan, kota Jayapura merupakan pusat aktivitas social dan ekonomi di provinsi Papua, dan penduduknya sangat padat secara rill. Data yang digunakan data primer dan dekunder. Teknik pengumpulan data melalui studi pustaka dan survey lapang. Dari kajian ini ditemukan bahwa Dampak Covid 19 terhadap pertumbuhan ekonomi Provinsi Papua pada triwulan I tahun 2020, mencapai 1,48 persen (yoy), pada triwulan II-2020, mencapai pertumbuhan sebesar 4,52 persen, namun pada triwukan III tahun 2020 terjadi kontraksi sebesar 2,62 persen, lebih kecil dibandingkan dengan pertumbuhan secara nasional terjadi kontraksi sebesar 3,49 persen. Tingkat Pengangguran Terbuka (TPT) pada Februari 2020 sebesar 66.296 orang atau meningkat 7,13 persen dibanding Februari 2019 hanya 61.885 orang (meningkat 4.411 orang). Dampak Covid 19 terhadap pemutusan hubungan kerja di Papua sebanyak 2.602 orang yang dirumahkan, dan ini tersebar di beberapa kota besar seperti Kota Jayapura sebanyak 1.452 orang, Kabupaten Jayapura sebanyak 465 orang, Keerom sebanyak 79 orang dan Kabupaten Merauke sebanyak 66 orang. Hasil Survey menunjukkan sebanyak 53.10 persen responden menyatakan bahwa harga 9 bahan kebutuhan pokok mengalami peningkatan di masa pandei covi’19. Dari responden pelajar dan mahasiswa yang disurvey 95 persen responden menyatakan mereka menggunakan fasilitas internet untuk belajar/mengajar di era pandemic Covid 19 ini. Sisanya sebesar 5 persen menyatakan “tidak pernah”. Aplikasi yang sering digunakan dalam proses pembelajaran 61,3 persen menggunakan WhatsApp dan 55,4 persen menggunakan aplikasi zoom, sisanya menggunakan lainnya. Dari survey ditemukan bahwa 23,4 persen responden adalah guru/dosen yang tidak pernah menggunakan vasilitas internet sebagai media pembelajaran. Kendala yang dihadapi guru/dosen : tidak tahu atau lainnya dan tidak memiliki akses internet

    Bacillus subtilis GlcK activity requires cysteines within a motif that discriminates microbial glucokinases into two lineages

    BACKGROUND: Bacillus subtilis glucokinase (GlcK) (GenBank NP_390365) is an ATP-dependent kinase that phosphorylates glucose to glucose 6-phosphate. The GlcK protein has very low sequence identity (13.7%) to the Escherichia coli glucokinase (Glk) (GenBank P46880) and some other glucokinases (EC 2.7.1.2), yet glucose is merely its substrate. Our lab has previously isolated and characterized the glcK gene. RESULTS: Microbial glucokinases can be grouped into two different lineages. One of the lineages contains three conserved cysteine (C) residues in a CXCGX(2)GCXE motif. This motif is also present in the B. subtilis GlcK. The GlcK protein occurs in both monomer and homodimer. Each GlcK monomer has six cysteines. All cysteine residues have been mutated, one-by-one, into alanine (A). The in vivo GlcK enzymatic activity was assayed by functional complementation in E. coli UE26 (ptsG ptsM glk). Mutation of the three motif-specific residues led to an inactive enzyme. The other mutated forms retained, or in one case (GlcK(C321A)) even gained, activity. The fluorescence spectra of the GlcK(C321A )showed a red shift and enhanced fluorescence intensity compare to the wild type's. CONCLUSIONS: Our results emphasize the necessity of cysteines within the CXCGX(2)GCXE motif for GlcK activity. On the other hand, the C321A mutation led to higher GlcK(C321A )enzymatic activity with respect to the wild type's, suggesting more adequate glucose phosphorylation

    Lignin Nanoparticles Comparative Study for their Emulsifying, Antibacterial and Antioxidant Properties

    Converting lignin, the second most natural abundant polymer on earth, into lignin nanoparticles (LNPs) form has potential applications. LNPs’ emulsifying, antibacterial and antioxidant properties of three different lignins, softwood kraft lignoboost (LB), hardwood birch (BB) and alkali protobind 1000 (PB) were evaluated in a comparative study at different pH and concentrations. Lower ionic strength of 5 millimole (mM) citric acid (CA) and pH of 7 were found to be the most optimum conditions for emulsion formation. Comparison among the two homogenization techniques revealed that microfluidizer is favored in case of BB- and PB-LNPs based emulsions, while ultrasonication is optimum in case of LB-LNPs based emulsions. LNPs’ concentrations were found directly in proportion with their emulsifying activity to stabilize emulsions. LNPs based rapeseed oil (RO)/water emulsions showed slightly better yield than LNPs based hexadecane (HD)/water emulsions in terms of particle size distribution (PSD) in PB-LNPs and LB-LNPs based samples while the opposite in case BB-LNPs emulsions. As an emulsifier, PB-LNPs showed the highest effect versus LP- LNPs least effective. In terms of antioxidant activity, BB-LNPs showed the highest effect represented by 83 % of 2,2-diphenyl-1-picrylhydrazyl (DPPH) inhibition while LB-LNPs’ DPPH inhibition was the lowest by 67 at 0.5 w/w% LNPs concentration. Microbiologically, LNPs exhibited strong inhibition against gram positive Staphylococcus aureus by achieving 3 logs of reduction; however, found less effective against gram negative Escherichia coli (E. coli) without major reduction. Metode yang digunakan dalam penelitian ini yaitu penelitian tindakan kelas. Subjek penelitian yaitu siswa kelas XI SM Angkasa dengan jumlah 15 Orang. Data yang diperoleh melalui tes, observasi dan dokumentasi. hasil penelitian yang telah dilakukan selama siklus sebagai berikut: Hasil belajar Menggiring bola dalam permainan sepak bola melalui Model pembelajaran Kooperatif tife team Games Tournamen(TGT) menunjukan peningkatan .Hal ini dibuktikan dengan hasil analisis data menggiring bola selama dua siklus,yaitu siklus 118,14% atau 15 subjek penelitian Dengan demikian dapat disimpulkan bahwa model pembelajaran kooperatif Tife Team Games Tournamen(TGT) efektif untuk membelajarkan menggiring dalam permainan sepakbola pada siswa kelas XI SMS Angkasa Lanud Pattimura Ambon.
