    Physiological constrains on Sverdrup's Critical-Depth-Hypothesis: the influences of dark respiration and sinking

    Discussions on the controls initiating the onset of the phytoplankton spring bloom in particular in the North Atlantic have since Sverdrup been dominated by the role of physical and biological drivers. Undoubtedly, these drivers play an important role in phytoplankton dynamics and thus the onset of the spring bloom. However, they neglect the cells ability to modify vital rates in response to changes in the external environment. In this study, we use a non-hydrostatic convection model coupled to an Individual-Based-Model to simulate changes phytoplankton cells during the transition from winter conditions as driven by convective mixing, and the onset of thermal stratification resulting in the spring bloom. The comparison between a simulation using a standard fixed rate approach in line with the original Sverdrup hypothesis and a simulation parameterized to include variable respiration and sinking rates showed that the latter approach was able to capture the observed phytoplankton concentration during deep convective mixing, the timing and magnitude of the spring bloom as well as simulating realistic physiological rates. In contrast, the model employing fixed rate parameterizations could only replicate field observations when employing unrealistic parameter values. These results highlight the necessity to consider not only the physical and biological external controls determining phytoplankton dynamics but also the cells ability to modify critical physiological rates in response to external constraints. Understanding these adaptive qualities will be of increasing importance in the future as species assemblages and physical controls change with changing climate

    Robotic Arm Manipulator Using Active Control for Sample Acquisition and Transfer, and Passive Mode for Surface Compliance

    A robotic arm that consists of three joints with four degrees of freedom (DOF) has been developed. It can carry an end-effector to acquire and transfer samples by using active control and comply with surface topology in a passive mode during a brief surface contact. The three joints are arranged in such a way that one joint of two DOFs is located at the shoulder, one joint of one DOF is located at the elbow, and one joint of one DOF is located at the wrist. Operationally, three DOFs are moved in the same plane, and the remaining one on the shoulder is moved perpendicular to the other three for better compliance with ground surface and more flexibility of sample handling.     Development of an Airborne Molecular Direct Detection Doppler Lidar for Tropospheric Wind Profiling

    Global measurement of tropospheric winds is a key measurement for understanding atmospheric dynamics and improving numerical weather prediction. Global wind profiles remain a high priority for the operational weather community and also for a variety of research applications including studies of the global hydrologic cycle and transport studies of aerosols and trace species. In addition to space based winds, high altitude airborne Doppler lidar systems flown on research aircraft, UAV's or other advanced sub-orbital platforms would be of great scientific benefit for studying mesoscale dynamics and storm systems such as hurricanes. The Tropospheric Wind Lidar Technology Experiment (TWiLiTE) is a three year program to advance the technology readiness level of the key technologies and subsystems of a molecular direct detection wind lidar system by validating them, at the system level, in an integrated airborne lidar system. The TWiLiTE Doppler lidar system is designed for autonomous operation on the WB57, a high altitude aircraft operated by NASA Johnson. The WE357 is capable of flying well above the midlatitude tropopause so the downward looking lidar will measure complete profiles of the horizontal wind field through the lower stratosphere and the entire troposphere. The completed system will have the capability to profile winds in clear air from the aircraft altitude of 18 km to the surface with 250 m vertical resolution and < 3 mis velocity accuracy. Progress in technology development and status of the instrument design will be presented

    Francisella tularensis Schu S4 lipopolysaccharide core sugar and o-antigen mutants are attenuated in a mouse model of tularemia

    The virulence factors mediating Francisella pathogenesis are being investigated, with an emphasis on understanding how the organism evades innate immunity mechanisms. Francisella tularensis produces a lipopolysaccharide (LPS) that is essentially inert and a polysaccharide capsule that helps the organism to evade detection by components of innate immunity. Using an F. tularensis Schu S4 mutant library, we identified strains that are disrupted for capsule and O-antigen production. These serum-sensitive strains lack both capsule production and O-antigen laddering. Analysis of the predicted protein sequences for the disrupted genes (FTT1236 and FTT1238c) revealed similarity to those for waa (rfa) biosynthetic genes in other bacteria. Mass spectrometry further revealed that these proteins are involved in LPS core sugar biosynthesis and the ligation of O antigen to the LPS core sugars. The 50% lethal dose (LD(50)) values of these strains are increased 100- to 1,000-fold for mice. Histopathology revealed that the immune response to the F. tularensis mutant strains was significantly different from that observed with wild-type-infected mice. The lung tissue from mutant-infected mice had widespread necrotic debris, but the spleens lacked necrosis and displayed neutrophilia. In contrast, the lungs of wild-type-infected mice had nominal necrosis, but the spleens had widespread necrosis. These data indicate that murine death caused by wild-type strains occurs by a mechanism different from that by which the mutant strains kill mice. Mice immunized with these mutant strains displayed >10-fold protective effects against virulent type A F. tularensis challenge

    The mTORC1 inhibitor everolimus prevents and treats Eμ-Myc lymphoma by restoring oncogene-induced senescence

    MYC deregulation is common in human cancer. IG-MYC translocations that are modeled in EμMyc mice occur in almost all cases of Burkitt lymphoma as well as in other B-cell lymphoproliferative disorders. Deregulated expression of MYC results in increased mTOR complex 1 (mTORC1) signaling. As tumors with mTORC1 activation are sensitive to mTORC1 inhibition, we used everolimus, a potent and specific mTORC1 inhibitor, to test the requirement for mTORC1 in the initiation and maintenance of EμMyc lymphoma. Everolimus selectively cleared premalignant B cells from the bone marrow and spleen, restored a normal pattern of B-cell differentiation, and strongly protected against lymphoma development. Established EμMyc lymphoma also regressed after everolimus therapy. Therapeutic response correlated with a cellular senescence phenotype and induction of p53 activity. Therefore, mTORC1-dependent evasion of senescence is critical for cellular transformation and tumor maintenance by MYC in B lymphocytes
