Validating a Method for Turbocharging Single Cylinder Four Stroke Engines

This paper presents a method for turbocharging single cylinder four stroke internal combustion engines, an experimental setup used to test this method, and the results from this experiment. A turbocharged engine has better fuel economy, cost efficiency, and power density than an equivalently sized, naturally aspirated engine. Most multi-cylinder diesel engines are turbocharged for this reason. However, due to the timing mismatch between the exhaust stroke (when the turbocharger is powered) and the intake stroke (when the engine intakes air), turbocharging is not used in commercial single cylinder engines. Single cylinder engines are ubiquitous in developing world offgrid power applications such as tractors, generators, and water pumps due to their low cost. Turbocharging these engines could give users a lower cost and more fuel efficient engine. The proposed solution is to add an air capacitor, in the form of a large volume intake manifold, between the turbocharger compressor and the engine intake to smooth out the flow. This research builds on a previous theoretical study where the turbocharger, capacitor, and engine system were modeled analytically. In order to validate the theoretical model, an experimental setup was created around a single cylinder four stroke diesel engine. A typical developing world engine was chosen and was fitted with a turbocharger. A series of sensors were added to this engine to measure pressure, temperature, and power output. Our tests showed that a turbocharger and air capacitor could be successfully fitted to a single cylinder engine to increase intake air density by forty-three percent and peak power output by twenty-nine percent.Massachusetts Institute of Technology. Method for Turbocharging Single Cylinder Four Stroke Engines

This paper presents a feasibility study of a method for turbocharging single cylinder, four-stroke internal combustion engines. Turbocharging is not conventionally used with single cylinder engines because of the timing mismatch between when the turbo is powered, during the exhaust stroke, and when it can deliver air to the cylinder, during the intake stroke. The proposed solution involves an air capacitor on the intake side of the engine between the turbocharger and intake valves. The capacitor acts as a buffer and would be implemented as a new style of intake manifold with a larger volume than traditional systems. In order for the air capacitor to be practical, it needs to be sized large enough to maintain the turbocharger pressure during the intake stroke, cause minimal turbo lag, and significantly increase the density of the intake air. By creating multiple flow models of air through the turbocharged engine system, we found that the optimal size air capacitor is between four and five times the engine capacity. For a capacitor sized for a one-liter engine, the lag time was found to be approximately two seconds, which would be acceptable for slowly accelerating applications such as tractors, or steady state applications such as generators. The density increase that can be achieved in the capacitor, compared to air at standard ambient temperature and pressure, was found to vary between fifty percent for adiabatic compression and no heat transfer from the capacitor, to eighty percent for perfect heat transfer. These increases in density are proportional to, to first order, the anticipated power increases that could be realized with a turbocharger and air capacitor system applied to a single cylinder, four-stroke engine.Massachusetts Institute of Technology. This paper draws on lessons from a UK case study in the management of diffuse microbial pollution from grassland farm systems in the Taw catchment, south west England. We report on the development and preliminary testing of a field-scale faecal indicator organism risk indexing tool (FIORIT). This tool aims to prioritise those fields most vulnerable in terms of their risk of contributing FIOs to water. FIORIT risk indices were related to recorded microbial water quality parameters (faecal coliforms [FC] and intestinal enterococci [IE]) to provide a concurrent on-farm evaluation of the tool. There was a significant upward trend in Log[FC] and Log[IE] values with FIORIT risk score classification (r2 =0.87 and 0.70, respectively and P<0.01 for both FIOs). The FIORIT was then applied to 162 representative grassland fields through different seasons for ten farms in the case study catchment to determine the distribution of on-farm spatial and temporal risk. The high risk fields made up only a small proportion (1%, 2%, 2% and 3% for winter, spring, summer and autumn, respectively) of the total number of fields assessed (and less than 10% of the total area), but the likelihood of the hydrological connection of high FIO source areas to receiving watercourses makes them a priority for mitigation efforts. The FIORIT provides a preliminary and evolving mechanism through which we can combine risk assessment with risk communication to end-users and provides a framework for prioritising future empirical research. Continued testing of FIORIT across different geographical areas under both low and high flow conditions is now needed to initiate its long term development into a robust indexing tool

Analyzing the Effect of Air Capacitor Turbocharging Single Cylinder Engines on Fuel Economy and Emissions Through Modeling and Experimentation

Turbocharging can provide a cost effective means for increasing the power output and fuel economy of an internal combustion engine. A turbocharger added to an internal combustion engine consists of a coupled turbine and compressor. Currently, turbocharging is common in multi-cylinder engines, but it is not commonly used on single-cylinder engines due to the phase mismatch between the exhaust stroke (when the turbocharger is powered) and the intake stroke (when the engine intakes the compressed air). The proposed method adds an air capacitor, an additional volume in series with the intake manifold, between the turbocharger compressor and the engine intake, to buffer the output from the turbocharger compressor and deliver pressurized air during the intake stroke. This research builds on previous work where it was shown experimentally that a power gain of 29% was achievable and that analytically a power gain of 40-60% was possible using a turbocharger and air capacitor system. The goal of this study is to further analyze the commercial viability of this technology by analyzing the effect of air capacitor turbocharging on emissions, fuel economy, and power density. An experiment was built and conducted that looked at how air capacitor sizing affected emissions, fuel economy, and the equivalence ratio. The experimental data was then used to calibrate a computational model built in Ricardo Wave. Finally this model was used to evaluate strategies to further improve the performance of a single cylinder diesel turbocharged engine with an air capacitor.Massachusetts Institute of Technology. BACKGROUND: Prior animal and human studies of prenatal exposure to solvents including tetrachloroethylene (PCE) have shown increases in the risk of certain congenital anomalies among exposed offspring. OBJECTIVES: This retrospective cohort study examined whether PCE contamination of public drinking water supplies in Massachusetts influenced the occurrence of congenital anomalies among children whose mothers were exposed around the time of conception. METHODS: The study included 1,658 children whose mothers were exposed to PCE-contaminated drinking water and a comparable group of 2,999 children of unexposed mothers. Mothers completed a self-administered questionnaire to gather information on all of their prior births, including the presence of anomalies, residential histories and confounding variables. PCE exposure was estimated using EPANET water distribution system modeling software that incorporated a fate and transport model. RESULTS: Children whose mothers had high exposure levels around the time of conception had an increased risk of congenital anomalies. The adjusted odds ratio of all anomalies combined among children with prenatal exposure in the uppermost quartile was 1.5 (95% CI: 0.9, 2.5). No meaningful increases in the risk were seen for lower exposure levels. Increases were also observed in the risk of neural tube defects (OR: 3.5, 95% CI: 0.8, 14.0) and oral clefts (OR 3.2, 95% CI: 0.7, 15.0) among offspring with any prenatal exposure. CONCLUSION: The results of this study suggest that the risk of certain congenital anomalies is increased among the offspring of women who were exposed to PCE-contaminated drinking water around the time of conception. Because these results are limited by the small number of children with congenital anomalies that were based on maternal reports, a follow-up investigation should be conducted with a larger number of affected children who are identified by independent records.National Institute of Environmental Health (5 P42 ES007381); National Institutes of Healt

Posttranscriptional regulation of PARG mRNA by HuR facilitates DNA repair and resistance to PARP inhibitors

The majority of pancreatic ductal adenocarcinomas (PDAC) rely on the mRNA stability factor HuR (ELAV-L1) to drive cancer growth and progression. Here, we show that CRISPR-Cas9–mediated silencing of the HuR locus increases the relative sensitivity of PDAC cells to PARP inhibitors (PARPi). PDAC cells treated with PARPi stimulated translocation of HuR from the nucleus to the cytoplasm, specifically promoting stabilization of a new target, poly (ADP-ribose) glycohydrolase (PARG) mRNA, by binding a unique sequence embedded in its 30 untranslated region. HuR-dependent upregulation of PARG expression facilitated DNA repair via hydrolysis of polyADP-ribose on related repair proteins. Accordingly, strategies to inhibit HuR directly promoted DNA damage accumulation, inefficient PAR removal, and persistent PARP-1 residency on chromatin (PARP-1 trapping). Immunoprecipitation assays demonstrated that the PARP-1 protein binds and posttranslationally modifies HuR in PARPi-treated PDAC cells. In a mouse xenograft model of human PDAC, PARPi monotherapy combined with targeted silencing of HuR significantly reduced tumor growth compared with PARPi therapy alone. Our results highlight the HuR–PARG axis as an opportunity to enhance PARPi-based therapies. ©2017 AACR

We describe the pedagogy behind the MIT Beaver Works Summer Institute Robotics Program, a new high-school STEM program in robotics. The program utilizes state-of-the-art sensors and embedded computers for mobile robotics. These components are carried on an exciting 1/10-scale race-car platform. The program has three salient, distinguishing features: (i) it focuses on robotics software systems: the students design and build robotics software towards real-world applications, without being distracted by hardware issues; (ii) it champions project-based learning: the students learn through weekly project assignments and a final course challenge; (iii) the learning is implemented in a collaborative fashion: the students learn the basics of collaboration and technical communication in lectures, and they work in teams to design and implement their software systems. The program was offered as a four-week residential program at MIT in the summer of 2016. In this paper, we provide the details of this new program, its teaching objectives, and its results. We also briefly discuss future directions and opportunities
