Methods of Protein Characterization
AppA is a protein in Rhodobacter sphaeroides that has been the topic of debate among scientists for the past several years with regards to the structure of the protein. It has been known that AppA has an effect on the activity of PpsR, which controls the gene expression of photosystems. There are two conflicting experimental structures (2IYG and 1YRX) of the protein, both of which claim to be taken in the dark phase (meaning when there is no light shining on the protein). The debate is about whether some slight differences in the structures represent the shift from the dark state to the light-induced state. It is known that in the light state, AppA releases PpsR and undergoes a dimerization event with itself. We observed the interactions within an experimental structure of the AppA dimer and compared them to the 2IYG and 1YRX structures. Most scientists have focused on the positioning of Trp104 to explain the functional changes of AppA upon light excitation, but our analysis of experimental structures suggests that the dimerization could play a larger role in the functional mechanism of AppA

Survey of traffic volume is one of the simplest methods to obtain traffic data in order to better understand optimalisation andefficiency so that it can minimize vehicle traffic congestion problems on the highway. The method used is based on descriptive andanalytical methods, which is done is to classify the vehicles in classes manually by counting the number of vehicles per time unitbased on class - class. The purpose of the volume of traffic surveys carried out in the classified Narogong Cullinan Road, Bogorregency during the period August, 2011, results that can be found is the degree of saturation of the highway is still in an acceptablelevel. Analysis of traffic flow at the study site is still under the limit congestion. It is suggested that to reduce the traffic density, thetype of heavy vehicles such as out of the factory operates around the study site between the hours of 10:00 pm to 3:00 am
Developing Formulas for Quick Calculation of Polyhedron Volume in Spatial Geometry: Application to Vietnam
In the age of globalization, an effective leadership skill is the ability for quick calculation of work-related problems. From an economic perspective, fast computation often provides a competitive advantage in business, where speed, efficiency and accuracy are required. Quick calculation techniques are a central problem in modern mathematics because it shortens the time for solving technical problems. The purpose of the paper is to provide an explanation that will lead to a quick solution to a volume problem. Specifically, some convenient formulas are provided for quick calculation of the volume of the common polyhedron, together with a number of multiple-choice questions with IATA software to practice. Based on the evaluation results, reliable multiple-choice questions are used for an empirical study in Can Tho City, Vietnam on the effectiveness of the formulas for quick calculation of the polyhedron volume in spatial geometry. Statistical analysis shows that quick formulas help students to complete lessons at a higher rate, thereby contributing to improvements in the effectiveness of teaching geometry, especially the volume of the Polyhedron

Wicked problems in a post-truth political economy: a dilemma for knowledge translation
The discipline of knowledge translation (KT) emerged as a way of systematically understanding and addressing the challenges of applying health and medical research in practice. In light of ongoing and emerging critique of KT from the medical humanities and social sciences disciplines, KT researchers have become increasingly aware of the complexity of the translational process, particularly the significance of culture, tradition and values in how scientific evidence is understood and received, and thus increasingly receptive to pluralistic notions of knowledge. Hence, there is now an emerging view of KT as a highly complex, dynamic, and integrated sociological phenomenon, which neither assumes nor creates knowledge hierarchies and neither prescribes nor privileges scientific evidence. Such a view, however, does not guarantee that scientific evidence will be applied in practice and thus poses a significant dilemma for KT regarding its status as a scientific and practice-oriented discipline, particularly within the current sociopolitical climate. Therefore, in response to the ongoing and emerging critique of KT, we argue that KT must provide scope for relevant scientific evidence to occupy an appropriate position of epistemic primacy in public discourse. Such a view is not intended to uphold the privileged status of science nor affirm the “scientific logos” per se. It is proffered as a counterbalance to powerful social, cultural, political and market forces that are able to challenge scientific evidence and promote disinformation to the detriment of democratic outcomes and the public good

Transcriptomic cytoarchitecture reveals principles of human neocortex organization.
Variation in cytoarchitecture is the basis for the histological definition of cortical areas. We used single cell transcriptomics and performed cellular characterization of the human cortex to better understand cortical areal specialization. Single-nucleus RNA-sequencing of 8 areas spanning cortical structural variation showed a highly consistent cellular makeup for 24 cell subclasses. However, proportions of excitatory neuron subclasses varied substantially, likely reflecting differences in connectivity across primary sensorimotor and association cortices. Laminar organization of astrocytes and oligodendrocytes also differed across areas. Primary visual cortex showed characteristic organization with major changes in the excitatory to inhibitory neuron ratio, expansion of layer 4 excitatory neurons, and specialized inhibitory neurons. These results lay the groundwork for a refined cellular and molecular characterization of human cortical cytoarchitecture and areal specialization
Predictive markers for the early prognosis of dengue severity: A systematic review and meta-analysis
BACKGROUND: Predictive markers represent a solution for the proactive management of severe dengue. Despite the low mortality rate resulting from severe cases, dengue requires constant examination and round-the-clock nursing care due to the unpredictable progression of complications, posing a burden on clinical triage and material resources. Accordingly, identifying markers that allow for predicting disease prognosis from the initial diagnosis is needed. Given the improved pathogenesis understanding, myriad candidates have been proposed to be associated with severe dengue progression. Thus, we aim to review the relationship between the available biomarkers and severe dengue. METHODOLOGY: We performed a systematic review and meta-analysis to compare the differences in host data collected within 72 hours of fever onset amongst the different disease severity levels. We searched nine bibliographic databases without restrictive criteria of language and publication date. We assessed risk of bias and graded robustness of evidence using NHLBI quality assessments and GRADE, respectively. This study protocol is registered in PROSPERO (CRD42018104495). PRINCIPAL FINDINGS: Of 4000 records found, 40 studies for qualitative synthesis, 19 for meta-analysis. We identified 108 host and viral markers collected within 72 hours of fever onset from 6160 laboratory-confirmed dengue cases, including hematopoietic parameters, biochemical substances, clinical symptoms, immune mediators, viral particles, and host genes. Overall, inconsistent case classifications explained substantial heterogeneity, and meta-analyses lacked statistical power. Still, moderate-certainty evidence indicated significantly lower platelet counts (SMD -0.65, 95% CI -0.97 to -0.32) and higher AST levels (SMD 0.87, 95% CI 0.36 to 1.38) in severe cases when compared to non-severe dengue during this time window. CONCLUSION: The findings suggest that alterations of platelet count and AST level—in the first 72 hours of fever onset—are independent markers predicting the development of severe dengue
