    Illinois Digital Scholarship: Preserving and Accessing the Digital Past, Present, and Future

    Since the University's establishment in 1867, its scholarly output has been issued primarily in print, and the University Library and Archives have been readily able to collect, preserve, and to provide access to that output. Today, technological, economic, political and social forces are buffeting all means of scholarly communication. Scholars, academic institutions and publishers are engaged in debate about the impact of digital scholarship and open access publishing on the promotion and tenure process. The upsurge in digital scholarship affects many aspects of the academic enterprise, including how we record, evaluate, preserve, organize and disseminate scholarly work. The result has left the Library with no ready means by which to archive digitally produced publications, reports, presentations, and learning objects, much of which cannot be adequately represented in print form. In this incredibly fluid environment of digital scholarship, the critical question of how we will collect, preserve, and manage access to this important part of the University scholarly record demands a rational and forward-looking plan - one that includes perspectives from diverse scholarly disciplines, incorporates significant research breakthroughs in information science and computer science, and makes effective projections for future integration within the Library and computing services as a part of the campus infrastructure.Prepared jointly by the University of Illinois Library and CITES at the University of Illinois at Urbana-Champaig

    Proposal for an IMLS Collection Registry and Metadata Repository

    The University of Illinois at Urbana-Champaign proposes to design, implement, and research a collection-level registry and item-level metadata repository service that will aggregate information about digital collections and items of digital content created using funds from Institute of Museum and Library Services (IMLS) National Leadership Grants. This work will be a collaboration by the University Library and the Graduate School of Library and Information Science. All extant digital collections initiated or augmented under IMLS aegis from 1998 through September 30, 2005 will be included in the proposed collection registry. Item-level metadata will be harvested from collections making such content available using the Open Archives Initiative Protocol for Metadata Harvesting (OAI PMH). As part of this work, project personnel, in cooperation with IMLS staff and grantees, will define and document appropriate metadata schemas, help create and maintain collection-level metadata records, assist in implementing OAI compliant metadata provider services for dissemination of item-level metadata records, and research potential benefits and issues associated with these activities. The immediate outcomes of this work will be the practical demonstration of technologies that have the potential to enhance the visibility of IMLS funded online exhibits and digital library collections and improve discoverability of items contained in these resources. Experience gained and research conducted during this project will make clearer both the costs and the potential benefits associated with such services. Metadata provider and harvesting service implementations will be appropriately instrumented (e.g., customized anonymous transaction logs, online questionnaires for targeted user groups, performance monitors). At the conclusion of this project we will submit a final report that discusses tasks performed and lessons learned, presents business plans for sustaining registry and repository services, enumerates and summarizes potential benefits of these services, and makes recommendations regarding future implementations of these and related intermediary and end user interoperability services by IMLS projects.unpublishednot peer reviewe

    Chimeric carrier proteins for targeted delivery of tumor antigens to professional antigen presenting cells

    Tumor-specific T lymphocytes can be regarded as a highly effective mechanism for tumor rejection. A substantial number of T-cell defined tumor antigens including mutated oncoproteins and differentiation antigens have been identified. However, while most spontaneous tumors appear to be antigenic, few are immunogenic. Activation of tumor-specific cytotoxic T cells (CTL) requires presentation of tumor antigens by professional antigen presenting cells (APCs) via MHC I molecules. Due to their crucial role in T-cell activation, APCs are being exploited for active cancer immunotherapy. Present experimental strategies include the incubation of dendritic cells with synthetic, tumor specific peptides to achieve uptake of tumor antigens and presentation in the context of MHC molecules. Alternatively, gene therapeutic approaches are aimed at the endogenous expression of tumor antigens in APCs upon transfer of suitable vector constructs. Our strategy for the presentation of tumor antigens by APCs is based on the intracellular delivery of tumor antigens as part of a fusion protein specifically targeted to APC cell surface receptors. We have constructed prototype molecules that contain a soluble fragment of CTLA-4 for cell binding via interaction with B7 molecules, genetically fused to a protein fragment derived from the tumor-associated antigen ErbB2. To improve uptake and direct the antigenic determinant preferentially to the MHC class I pathway, in one of these protein vaccines also the translocation domain of the bacterial Pseudomonas exotoxin A has been included. In the parental toxin this protein domain facilitates escape from the endosomal compartment to the cytosol upon receptor mediated endocytosis. Here we have investigated the in vitro cell binding activity of such reagents and their antitumoral activity in immunocompetent murine model systems. Specific binding to B7 molecules and uptake of bacterially expressed protein vaccines could be demonstrated. Ex vivo restimulation with an ErbB2-derived peptide of splenocytes from Balb/c mice injected with the fusion proteins resulted in enhanced IFN-gamma production by T cells. Protective and therapeutic effects of ErbB2 protein vaccines were also investigated. Vaccinated animals were protected against subsequent challenge with syngeneic ErbB2 expressing tumor cells. Likewise, s.c. injection of ErbB2 protein vaccines in the vicinity of established tumors resulted in tumor rejection and long lasting protection indicating that immunological memory was induced. Our results suggest that chimeric proteins combining a tumor antigen and specific recognition of APCs in a single molecule are suitable for targeted delivery of antigens to professional APCs and might become valuable tools for cancer immunotherapy

    Risk-adapted FDG-PET/CT-based follow-up in patients with diffuse large B-cell lymphoma after first-line therapy

    Background: The purpose of this study was to evaluate the impact of 2-[fluorine-18]fluoro-2-deoxy-D-glucose-positron emission tomography/computed tomography (FDG-PET/CT) during follow-up of patients with diffuse large B-cell lymphoma (DLBCL) being in complete remission or unconfirmed complete remission after first-line therapy. Patients and methods: DLBCL patients receiving FDG-PET/CT during follow-up were analyzed retrospectively. Confirmatory biopsy was mandatory in cases of suspected disease recurrence. Results: Seventy-five patients were analyzed and 23 (30%) had disease recurrence. The positive predictive value (PPV) of FDG-PET/CT was 0.85. Patients >60 years [P = 0.036, hazard ratio (HR) = 3.82, 95% confidence interval (CI) 1.02-7.77] and patients with symptoms indicative of a relapse (P = 0.015; HR = 4.1; 95% CI 1.20-14.03) had a significantly higher risk for relapse. A risk score on the basis of signs of relapse, age >60 years, or a combination of these factors identified patients at high risk for recurrence (P = 0.041). Conclusions: FDG-PET/CT detects recurrent DLBCL after first-line therapy with high PPV. However, it should not be used routinely and if only in selected high-risk patients to reduce radiation burden and costs. On the basis of our retrospective data, FDG-PET/CT during follow-up is indicated for patients 60 years with and without clinical signs of relaps

    Hodgkin's lymphoma in remission after first-line therapy: which patients need FDG-PET/CT for follow-up?

    Background: The purpose of the study was to evaluate the impact of 2-[fluorine-18]fluoro-2-deoxy-D-glucose-positron emission tomography (FDG-PET)/computed tomography (CT) during follow-up of patients with Hodgkin's lymphoma. Patients and methods: Patients in complete remission or an unconfirmed complete remission after first-line therapy who received FDG-PET/CT during their follow-up were analyzed retrospectively. Confirmatory biopsy was mandatory in case of recurrence. Results: Overall, 134 patients were analyzed. Forty-two (31.3%) patients had a recurrence. The positive predictive value of FDG-PET/CT was 0.98. Single-factor analysis identified morphological residual mass [P = 0.0005, hazard ratio (HR) 3.4, 95% confidence interval (CI) 1.7-6.6] and symptoms (P 24 months). Conclusions: Asymptomatic patients without morphological residues and an early stage of disease do not need a routine FDG-PET/CT for follow-up. Asymptomatic patients with morphological residues should receive routine follow-up FDG-PET/CT for the first 24 months. Only patients with advanced initial stage do need a routine follow-up FDG-PET/CT beyond 24 month

    Reimagining fitness practice in schools: an experimental study on self-perceptions and motivational outcomes in youth

    BACKGROUND: Physical fitness testing in physical education (PE) is mandatory in several US states, based on the promise that testing will assist students in establishing lifelong habits of regular physical activity. However, in actuality, the influence of fitness practice for testing, as commonly implemented in schools, on motivational outcomes remains largely unknown. Modifying the experience of fitness practice testing by translating psychological theory into practice may represent untapped potential in the effort to promote physical activity in childhood. Thus, the purpose of the present study is to examine the effect of different structures of fitness practice (traditional versus needs supporting lessons) on affect and motivational outcomes, as well as the moderating role of BMI and self-perceptions. METHODS: Using a within-subject experimental design, the present study measured pre-post changes on two 20 to 30-minute PE lessons of fitness practice (traditional versus novel): one simulated traditional testing (using standard FITNESSGRAM instructions) whereas the other ( novel ) emphasized positive peer interactions and alternative ways of delivering the same tasks. The dependent variables were positive affect (Feeling Scale), arousal (Felt Arousal Scale), body related emotions (Body Esteem Scale), enjoyment (PACES), intention, and need satisfaction (Activity Feeling States Scale). The sample consisted of 148 students in 4th 5th and 6th grade (119 normal-weight, 29 overweight; 77 female), recruited from two schools in Central Iowa. Physical activity during both PE lessons was measured by accelerometers (Sensewear Armbands), and BMI records were obtained from the schools. RESULTS: The present study found that affective responses and motivation differed overall between the two fitness practice lessons, even though the volume of physical activity was the same. This effect was partially moderated by BMI and physical self-perceptions. Children reported significantly higher levels of affect at the conclusion of the non-traditional fitness practice, as well as increased levels of enjoyment, arousal, perceived competence and autonomy, and were more likely to choose the non-traditional lesson again. BMI played a significant moderating role in arousal, social competence in affect, and physical self-perceptions in enjoyment, with beneficial scores for the non-traditional lesson. Results did not show significant differences between the two lessons on perceived relatedness. CONCLUSION: Overall, the novel lesson was more enjoyable, resulting in a more attractive option for fitness practice for children. The present findings suggest that an alternative approach to traditional fitness practice might be beneficial for all children, as well as those with low physical self-perceptions and social competence. Those benefits can be seen in increases in affect and motivational outcomes. An approach like this may lead to higher levels of physical activity in the future. These results highlight the potential impact of translating extant psychological research into practice in PE settings
