    CCharPPI web server: computational characterization of protein–protein interactions from structure

    The atomic structures of protein–protein interactions are central to understanding their role in biological systems, and a wide variety of biophysical functions and potentials have been developed for their characterization and the construction of predictive models. These tools are scattered across a multitude of stand-alone programs, and are often available only as model parameters requiring reimplementation. This acts as a significant barrier to their widespread adoption. CCharPPI integrates many of these tools into a single web server.     The Sagittarius Dwarf spheroidal Galaxy Survey (SDGS) II: The stellar content and constraints on the star formation history

    A detailed study of the Star Formation History of the Sgr dSph galaxy is performed through the analysis of the data from the Sagittarius Dwarf Galaxy Survey (SDGS; Bellazzini, Ferraro & Buonanno 1999). Accurate statistical decontamination of the SDGS Color - Magnitude diagrams allow us to obtain many useful constraints on the age and metal content of the Sgr stellar populations in three different region of the galaxy. A coarse metallicity distribution of Sgr stars is derived, ranging from [Fe/H]~ -2.0 to [Fe/H]~ -0.7, the upper limit being somewhat higher in the central region of the galaxy. A qualitative global fit to all the observed CMD features is attempted, and a general scheme for the Star Formation History of the Sgr is derived. According to this scheme, star formation began at very early time from a low metal content Inter Stellar Medium and lasted for several Gyr, coupled with progressive chemical enrichment. The Star Formation Rate (SFR) had a peak from 8 to 10 gyr ago when the mean metallicity was in the range -1.3<= [Fe/H] <= -0.7.     Frequency shifts of photoassociative spectra of ultracold metastable Helium atoms : a new measurement of the s-wave scattering length

    We observe light-induced frequency shifts in one-color photoassociative spectra of magnetically trapped 4^4He^* atoms in the metastable 23S12^3S_1 state. A pair of ultracold spin-polarized 23S12^3S_1 helium atoms is excited into a molecular bound state in the purely long range 0u+0_u^+ potential connected to the 23S123P02^3S_1 - 2^3P_0 asymptote. The shift arises from the optical coupling of the molecular excited bound state with the scattering states and the bound states of two colliding 23S12^3S_1 atoms. We measure the frequency-shifts for several ro-vibrational levels in the 0u+0^+_u potential and find a linear dependence on the photoassociation laser intensity. Comparison with a theoretical analysis provides a good indication for the s-wave scattering length aa of the quintet (5Σg+^5\Sigma_g^+) potential, a=7.2±0.6a=7.2\pm 0.6 nm, which is significantly lower than most previous results obtained by non-spectroscopic methods.Comment: 7 pages, 4 figure

    Radiographic Outcomes of Adult Spinal Deformity Correction : A Critical Analysis of Variability and Failures Across Deformity Patterns

    Study Design: Multicenter, prospective, consecutive, surgical case series from the International Spine Study Group. Objectives: To evaluate the effectiveness of surgical treatment in restoring spinopelvic (SP) alignment. Summary of Background Data: Pain and disability in the setting of adult spinal deformity have been correlated with global coronal alignment (GCA), sagittal vertical axis (SVA), pelvic incidence/lumbar lordosis mismatch (PI-LL), and pelvic tilt (PT). One of the main goals of surgery for adult spinal deformity is to correct these parameters to restore harmonious SP alignment. Methods: Inclusion criteria were operative patients (age greater than 18 years) with baseline (BL) and 1-year full-length X-rays. Thoracic and thoracolumbar Cobb angle and previous mentioned parameters were calculated. Each parameter at BL and 1 year was categorized as either pathological or normal. Pathologic limits were: Cobb greater than 30 , GCA greater than 40 mm, SVA greater than 40 mm, PI-LL greater than 10 , and PT greater than 20 . According to thresholds, corrected or worsened alignment groups of patients were identified and overall radiographic effectiveness of procedure was evaluated by combining the results from the coronal and sagittal planes. Fondation Paristech, ISS

    Corticosterone Regulates Both Naturally Occurring and Cocaine‐Induced Dopamine Signaling by Selectively Decreasing Dopamine Uptake

    Stressful and aversive events promote maladaptive reward‐seeking behaviors such as drug addiction by acting, in part, on the mesolimbic dopamine system. Using animal models, data from our laboratory and others show that stress and cocaine can interact to produce a synergistic effect on reward circuitry. This effect is also observed when the stress hormone corticosterone is administered directly into the nucleus accumbens (NAc), indicating that glucocorticoids act locally in dopamine terminal regions to enhance cocaine\u27s effects on dopamine signaling. However, prior studies in behaving animals have not provided mechanistic insight. Using fast‐scan cyclic voltammetry, we examined the effect of systemic corticosterone on spontaneous dopamine release events (transients) in the NAc core and shell in behaving rats. A physiologically relevant systemic injection of corticosterone (2 mg/kg i.p.) induced an increase in dopamine transient amplitude and duration (both voltammetric measures sensitive to decreases in dopamine clearance), but had no effect on the frequency of transient release events. This effect was compounded by cocaine (2.5 mg/kg i.p.). However, a second experiment indicated that the same injection of corticosterone had no detectable effect on the dopaminergic encoding of a palatable natural reward (saccharin). Taken together, these results suggest that corticosterone interferes with naturally occurring dopamine uptake locally, and this effect is a critical determinant of dopamine concentration specifically in situations in which the dopamine transporter is pharmacologically blocked by cocaine

    Review of optimism, creativity and spirituality in tourism research

    Purpose - Optimism, creativity, and spirituality are important variables in Positive Psychology that can lead to better lives. The authors believe these are also fundamental concepts that can bring a new understanding of the tourism experiences, particularly in the current trend of the 'experience economy'. The study aimed to explore optimism, creativity, and spirituality from a Positive Psychology perspective on tourism settings to deepen the understanding of the state of the art and develop ideas for improvement of tourism research experiences Design - An extensive systematic literature review was developed anchored on PRISMA (2009) guidelines. The search engine Online Knowledge Library (B-On) was used and the search terms 'Optimism', 'Creativity' and 'Spirituality' crossed individually with 'Tourism', 'Travel' and 'Visitor'. In-depth analysis considering the chosen inclusion/exclusion criteria were undertaken concluding with a sample of 31 references. Findings - Overall, results showed a lack of optimism studies on tourism, leading to an urgent need to endeavor in the challenge of developing research. Creativity in tourism is mainly used as a background for the place and its culture and considers organizational, structural and product innovation development. Further, tourism can be seen as a spiritual activity seeking personal enrichment and fulfillment. Originality of the research - This study considers three variables not usually applied to tourism contexts as is clear by the low number of references found.     Morris Water Maze Learning in Two Rat Strains Increases the Expression of the Polysialylated Form of the Neural Cell Adhesion Molecule in the Dentate Gyrus But Has No Effect on Hippocampal Neurogenesis

    In the current study, the authors investigated whether Morris water maze learning induces alterations in hippocampal neurogenesis or neural cell adhesion molecule (NCAM) polysialylation in the dentate gyrus. Two frequently used rat strains, Wistar and Sprague–Dawley, were trained in the spatial or the nonspatial version of the water maze. Both training paradigms did not have an effect on survival of newly formed cells that were labeled 7–9 days prior to the training or on progenitor proliferation in the subgranular zone. However, the granule cell layer of the spatially trained rats contained significantly more positive cells of the polysialylated form of the NCAM. These data demonstrate that Morris water maze learning causes plastic change in the dentate gyrus without affecting hippocampal neurogenesis.

    Corticosterone Acts in the Nucleus Accumbens to Enhance Dopamine Signaling and Potentiate Reinstatement of Cocaine Seeking

    Stressful life events are important contributors to relapse in recovering cocaine addicts, but the mechanisms by which they influence motivational systems are poorly understood. Studies suggest that stress may “set the stage” for relapse by increasing the sensitivity of brain reward circuits to drug-associated stimuli. We examined the effects of stress and corticosterone on behavioral and neurochemical responses of rats to a cocaine prime after cocaine self-administration and extinction. Exposure of rats to acute electric footshock stress did not by itself reinstate drug-seeking behavior but potentiated reinstatement in response to a subthreshold dose of cocaine. This effect of stress was not observed in adrenalectomized animals, and was reproduced in nonstressed animals by administration of corticosterone at a dose that reproduced stress-induced plasma levels. Pretreatment with the glucocorticoid receptor antagonist RU38486 did not block the corticosterone effect. Corticosterone potentiated cocaine-induced increases in extracellular dopamine in the nucleus accumbens (NAc), and pharmacological blockade of NAc dopamine receptors blocked corticosterone-induced potentiation of reinstatement. Intra-accumbens administration of corticosterone reproduced the behavioral effects of stress and systemic corticosterone. Corticosterone treatment acutely decreased NAc dopamine clearance measured by fast-scan cyclic voltammetry, suggesting that inhibition of uptake2-mediated dopamine clearance may underlie corticosterone effects. Consistent with this hypothesis, intra-accumbens administration of the uptake2 inhibitor normetanephrine potentiated cocaine-induced reinstatement. Expression of organic cation transporter 3, a corticosterone-sensitive uptake2 transporter, was detected on NAc neurons. These findings reveal a novel mechanism by which stress hormones can rapidly regulate dopamine signaling and contribute to the impact of stress on drug intake
