    Adipokines (Leptin, Adiponectin, Resistin) Differentially Regulate All Hormonal Cell Types in Primary Anterior Pituitary Cell Cultures from Two Primate Species

    Adipose-tissue (AT) is an endocrine organ that dynamically secretes multiple hormones, the adipokines, which regulate key physiological processes. However, adipokines and their receptors are also expressed and regulated in other tissues, including the pituitary, suggesting that locally- and AT-produced adipokines might comprise a regulatory circuit that relevantly modulate pituitary cell-function. Here, we used primary pituitary cell-cultures from two normal nonhuman-primate species [Papio-anubis/Macaca-fascicularis] to determine the impact of different adipokines on the functioning of all anterior-pituitary cell-types. Leptin and resistin stimulated GH-release, a response that was blocked by somatostatin. Conversely, adiponectin decreased GH-release, and inhibited GHRH-, but not ghrelin-stimulated GH-secretion. Furthermore: 1) Leptin stimulated PRL/ACTH/FSH- but not LH/TSH-release; 2) adiponectin stimulated PRL-, inhibited ACTH- and did not alter LH/FSH/TSH-release; and 3) resistin increased ACTH-release and did not alter PRL/LH/FSH/TSH-secretion. These effects were mediated through the activation of common (AC/PKA) and distinct (PLC/PKC, intra-/extra-cellular calcium, PI3K/MAPK/mTOR) signaling-pathways, and by the gene-expression regulation of key receptors/transcriptional-factors involved in the functioning of these pituitary cell-types (e.g. GHRH/ghrelin/somatostatin/insulin/IGF-I-receptors/Pit-1). Finally, we found that primate pituitaries expressed leptin/adiponectin/resistin. Altogether, these and previous data suggest that local-production of adipokines/receptors, in conjunction with circulating adipokine-levels, might comprise a relevant regulatory circuit that contribute to the fine-regulation of pituitary functions

    The Causes and Consequences of Job-Related Stress among Prosecutors

    Despite a growing body of literature documenting work stress among employees in various fields, there is a dearth of research that explicitly assesses the impact of different aspects of prosecutors' working conditions on individual prosecutors and their organizations. Drawing on a sample of prosecutors in a southern state in the U.S., we first used OLS regression to examine which work-related stressors are important in predicting their levels of work stress, job satisfaction, and turnover intention. Using path analysis, we then explored the links between these stressors and a set of proposed mediating and outcome variables. We also conducted unstructured in-depth interviews with a subset of the sample to supplement and further illustrate the observed patterns. While job demands and organizational support play the most important roles in explaining the work stress of prosecutors, they do not have direct impact on the prosecutor's commitment to the job. The roles played by psychological and emotional stressors are found to be negligible. Job-related stressors do not lead to turnover intention directly but indirectly through work stress and job satisfaction with a notable exception of the public/media stressor. This study provides a scientifically-based perspective regarding which working conditions should be addressed to maintain healthy and productive working environments among prosecutors.N

    Swedish Police Officers’ Job Strain, Work-Related Social Support and General Mental Health

    This study investigated the association between psychosocial job characteristics and general mental health among police officers and the extent to which social support at work plays a role in this association. The findings are based on a cross-sectional survey. A written questionnaire was assessed by 714 police officers volunteered to participate in the study. The participants completed a series of validated instruments assessing job demand, control and social support at work (JDCS); general mental health (GHQ); and sociodemographic characteristics. High job strain was associated with low levels of work-related social support. Moreover, poor mental health was associated with low levels of work-related social support, active work and high job strain. The joint effect of high job strain and low levels of work-related social support had a significant effect on poor mental health. Work-related social support buffered job strain to some extent. Workforce health promotion policies should attempt to reduce job strain and emphasise the importance of work-related social support. Knowledge about police officers' general mental health and policymakers' support for police officers may have positive effects on the performance of the police force.</p
