Relationship between soil cobalt and vitamin B12 levels in the liver of livestock in Saudi Arabia: role of competing elements in soils
Objective: This study aimed to analyze the agricultural soils from different regions in Saudi Arabia for cobalt and related metals as Cu2+, Ni2+,Cr3++, Zn2+ and Pb2+.Materlais and methods: Liver and muscle tissues of livestock grazing on the selected areas were analyzed for the content of Co and vitamin B12.Results: Our results indicated that the levels of Co in surface soil (0-15 cm) were higher than in sub-surface soil (>15 cm- 45 cm). In contrast, Pb and Zn were higher in sub-surface soil than in surface soil. A significant positive correlation existed between the levels of Co and vitamin B12 in the liver of livestock. However, Co was not detected in muscle tissues while vitamin B12 was present at very low levels in comparison with the levels found in the liver. The results indicated that Zn2+, Pb2+ compete with Co in soil, which eventually affected the levels of vitamin B12 in liver.Conclusion: It was recommended that survey of heavy metals in grazing fields of cattle should consider inclusion of multiple elements that compete with the bioavailability of essential elements in plants and animals for the prevention of deficiency of essential elements such as Co.Keywords: Soil, vitamin B12, cobalt, cattle, uptake, transfe
Bone turnover biomarkers in obese postmenopausal Saudi women with type-ІІ diabetes mellitus
Background: There is a high prevalence of diabetes mellitus type-2 (T2DM) and osteoporosis are problems worldwide. In this study, we evaluated the correlation between T2DM and bone turnover in diabetic obese postmenopausal Saudi women.Subjects and Methods: The present study included total of 65 T2-DM obese postmenopausal Saudi women, (36 uncontrolled, 29 controlled) .The following serum biochemical parameters were evaluated [fasting blood glucose (FBG),total calcium (Ca), phosphorus (Pi), parathyroid hormone (PTH), 1,25-(OH)2Vitamin D3 ,osteocalcin (OC), procollagen (PICP) and cathepsin k (Cath K) ].Results: Serum OC levels were significantly decreased in diabetic obese postmenopausal group compared to their respective healthy group (P < 0.004). PICP and Cath K were significantly elevated in diabetic postmenopausal group compared to the healthy group ( P < 0.024 & 0.001). A significant elevation in 1,25(OH)2 Vitamin D3, Ca and Pi levels in diabetic obese postmenopausal patients group compared to the healthy group. However, a non-significant changes was observed in serum PTH level between different groups.Conclusion: In this study, the changes in the biochemical parameters and bone turnover markers in obese women are strong risk factors for diabetes development that may contribute to osteopenia and osteoporosis. The study showed the strong effect of T2DM on biochemical markers of bone turnover in obese postmenopausal Saudi women.Keyword: Diabetes mellitus type-2, Bone markers, Postmenopausa
Impact of aqueous doash extract on urinary mutagenicity in rats exposed to heterocyclic amines.
Doash (Origanum majorana) is an herbaceous plant found commonly in Saudi Arabia. It is used as a food flavor and a folk remedy to treat a number of diseases. The 2-amino-3-methylimidazo[4,5-f] quinoline (IQ) and 2-amino-1-methyl-6-phenylimidazo[4,5-b]pyridine (PhIP) are the most abundant of the heterocyclic amine carcinogens present in cooked food. In the present study, the potential of doash tea to influence carcinogen metabolism was investigated indirectly using heterocyclic amines as model mutagens, IQ and PhIP. Results obtained showed that doash tea had no influence on body weight in both the studies. Rats were treated with different doses of IQ (1, 3, 5 and 10 mg/kg) or PhIP (1, 5, 10 and 20 mg/kg). The selected dosage was 5 mg/kg for both heterocyclic amines. Results obtained revealed that rats treated with doash tea and given a single dose of the heterocyclic amines, whether for 1 day (short-term) or for 1 month (long term), showed a statistically significant decrease in their excretion of indirect mutagens (IQ or PhIP). Following treatment of the rats with a single oral dose of IQ or PhIP, the highest mutagenic activity determined in the presence of an activation system was excreted in the urine after 24 h, with much lower levels of mutagencity being excreted during subsequent elimination from the body. No mutagenicity was observed in the absence of an activation system that is direct-acting mutagenicity using (IQ and PhIP). Statistical analysis revealed that, in comparison with the control group, the aqueous doash extract significantly reduced the mutagenic response after 24 h. It was concluded that doash extract significantly decreased the excretion of mutagens in comparison with the control group (water only)
