The Generation of Successive Unmarked Mutations and Chromosomal Insertion of Heterologous Genes in Actinobacillus pleuropneumoniae Using Natural Transformation
We have developed a simple method of generating scarless, unmarked mutations in Actinobacillus pleuropneumoniae by exploiting the ability of this bacterium to undergo natural transformation, and with no need to introduce plasmids encoding recombinases or resolvases. This method involves two successive rounds of natural transformation using linear DNA: the first introduces a cassette carrying cat (which allows selection by chloramphenicol) and sacB (which allows counter-selection using sucrose) flanked by sequences to either side of the target gene; the second transformation utilises the flanking sequences ligated directly to each other in order to remove the cat-sacB cassette. In order to ensure efficient uptake of the target DNA during transformation, A. pleuropneumoniae uptake sequences are added into the constructs used in both rounds of transformation. This method can be used to generate multiple successive deletions and can also be used to introduce targeted point mutations or insertions of heterologous genes into the A. pleuropneumoniae chromosome for development of live attenuated vaccine strains. So far, we have applied this method to highly transformable isolates of serovars 8 (MIDG2331), which is the most prevalent in the UK, and 15 (HS143). By screening clinical isolates of other serovars, it should be possible to identify other amenable strains
Resonance Patterns in a Stadium-shaped Microcavity
We investigate resonance patterns in a stadium-shaped microcavity around
, where is the refractive index, the vacuum
wavenumber, and the radius of the circular part of the cavity. We find that
the patterns of high resonances can be classified, even though the
classical dynamics of the stadium system is chaotic. The patterns of the high
resonances are consistent with the ray dynamical consideration, and appears
as the stationary lasing modes with low pumping rate in the nonlinear dynamical

Factors governing macrozoobenthic assemblages in perennial springs in north-western Switzerland
Springs are important freshwater habitats that provide refuge for many rare species. In this study, the fauna and abiotic parameters of 20 perennial springs in north-western Switzerland were investigated. Correlation of abiotic and macrozoobenthos data showed that physicochemical parameters had little impact on macrozoobenthic composition, whereas specific substrate parameters strongly influenced the composition of the macrofauna. Surprisingly, nonmetric multidimensional scaling did not reveal a grouping of springs with similar substrate composition or macrozoobenthic assemblages. However, discharge was identified as the factor significantly determining substrate and the composition of macroinvertebrate assemblages. Social roles and aging from a life-span perspective
Este trabalho investigou os papéis sociais e as tarefas evolutivas desempenhados por adultos. O local escolhido para investigação foi um assentamento de famílias de baixa renda do Distrito Federal criado em 1989. Utilizou-se um questionário contendo 17 questões abertas e 15 questões fechadas, preenchido pela primeira autora durante uma visita domiciliar. Participaram 98 respondentes (73 F e 25 M), sendo 51 entre 50 e 59 anos e 47 a partir de 60 anos. Os resultados apontaram que este grupo é heterogêneo e que seus papéis sociais são influenciados pelas variáveis demográficas (idade, sexo, escolaridade, ocupação, naturalidade e estado civil) e também pelas variáveis relativas à moradia atual. Concluiu-se também que as expectativas sociais, o suporte social e a escolarização são fatores de suma importância para oferecer recursos para a otimização e compensação necessárias a um envelhecimento bem sucedido. __________________________________________________________________________________________________________ ABSTRACTThis study investigated the social roles and developmental tasks of adults. The study took place in a settlement of low-income families, created in The Federal District in 1989. Data were collected through a questionnaire composed of 17 open and 15 closed questions, administered by the first author during a home visit. There were 98 respondents, 73 female and 25 male, being 51 between 50 to 59 years old and 47 elders above the age of 60. The result indicated that this group is heterogeneous and that its social roles are influenced by the demographic variables - age, sex, educational level, work, place of the birth and marital status, as well for the relative variables to current residence. The data allow the conclusion that social expectations, social support and the educational level are important resources for the optimization and necessary compensation to successful aging
Average ground-state energy of finite Fermi systems
Semiclassical theories like the Thomas-Fermi and Wigner-Kirkwood methods give
a good description of the smooth average part of the total energy of a Fermi
gas in some external potential when the chemical potential is varied. However,
in systems with a fixed number of particles N, these methods overbind the
actual average of the quantum energy as N is varied. We describe a theory that
accounts for this effect. Numerical illustrations are discussed for fermions
trapped in a harmonic oscillator potential and in a hard wall cavity, and for
self-consistent calculations of atomic nuclei. In the latter case, the
influence of deformations on the average behavior of the energy is also

Discrete Symmetries in the Weyl Expansion for Quantum Billiards
We consider two and three-dimensional quantum billiards with discrete
symmetries. We derive the first terms of the Weyl expansion for the level
density projected onto the irreducible representations of the symmetry group.
As an illustration the method is applied to the icosahedral billiard. The paper

Semiclassical theory for spatial density oscillations in fermionic systems
We investigate the particle and kinetic-energy densities for a system of
fermions bound in a local (mean-field) potential V(\bfr). We generalize a
recently developed semiclassical theory [J. Roccia and M. Brack, Phys. Rev.\
Lett. {\bf 100}, 200408 (2008)], in which the densities are calculated in terms
of the closed orbits of the corresponding classical system, to
dimensions. We regularize the semiclassical results for the U(1) symmetry
breaking occurring for spherical systems at and near the classical
turning points where the Friedel oscillations are predominant and well
reproduced by the shortest orbit going from to the closest turning point
and back. For systems with spherical symmetry, we show that there exist two
types of oscillations which can be attributed to radial and non-radial orbits,
respectively. The semiclassical theory is tested against exact
quantum-mechanical calculations for a variety of model potentials. We find a
very good overall numerical agreement between semiclassical and exact numerical
densities even for moderate particle numbers . Using a "local virial
theorem", shown to be valid (except for a small region around the classical
turning points) for arbitrary local potentials, we can prove that the
Thomas-Fermi functional reproduces the oscillations in

Families of spherical caps: spectra and ray limit
We consider a family of surfaces of revolution ranging between a disc and a
hemisphere, that is spherical caps. For this family, we study the spectral
density in the ray limit and arrive at a trace formula with geodesic polygons
describing the spectral fluctuations. When the caps approach the hemisphere the
spectrum becomes equally spaced and highly degenerate whereas the derived trace
formula breaks down. We discuss its divergence and also derive a different
trace formula for this hemispherical case. We next turn to perturbative
corrections in the wave number where the work in the literature is done for
either flat domains or curved without boundaries. In the present case, we
calculate the leading correction explicitly and incorporate it into the
semiclassical expression for the fluctuating part of the spectral density. To
the best of our knowledge, this is the first calculation of such perturbative
corrections in the case of curvature and boundary.Comment: 28 pages, 7 figure
Exploring differential item functioning in the SF-36 by demographic, clinical, psychological and social factors in an osteoarthritis population
The SF-36 is a very commonly used generic measure of health outcome in osteoarthritis (OA). An important, but frequently overlooked, aspect of validating health outcome measures is to establish if items work in the same way across subgroup of a population. That is, if respondents have the same 'true' level of outcome, does the item give the same score in different subgroups or is it biased towards one subgroup or another. Differential item functioning (DIF) can identify items that may be biased for one group or another and has been applied to measuring patient reported outcomes. Items may show DIF for different conditions and between cultures, however the SF-36 has not been specifically examined in an osteoarthritis population nor in a UK population. Hence, the aim of the study was to apply the DIF method to the SF-36 for a UK OA population. The sample comprised a community sample of 763 people with OA who participated in the Somerset and Avon Survey of Health. The SF-36 was explored for DIF with respect to demographic, social, clinical and psychological factors. Well developed ordinal regression models were used to identify DIF items. Results: DIF items were found by age (6 items), employment status (6 items), social class (2 items), mood (2 items), hip v knee (2 items), social deprivation (1 item) and body mass index (1 item). Although the impact of the DIF items rarely had a significant effect on the conclusions of group comparisons, in most cases there was a significant change in effect size. Overall, the SF-36 performed well with only a small number of DIF items identified, a reassuring finding in view of the frequent use of the SF-36 in OA. Nevertheless, where DIF items were identified it would be advisable to analyse data taking account of DIF items, especially when age effects are the focus of interest
Quantum chaos in a deformable billiard: Applications to quantum dots
We perform a detailed numerical study of energy-level and wavefunction
statistics of a deformable quantum billiard focusing on properties relevant to
semiconductor quantum dots. We consider the family of Robnik billiards
generated by simple conformal maps of the unit disk; the shape of this family
of billiards may be varied continuously at fixed area by tuning the parameters
of the map. The classical dynamics of these billiards is well-understood and
this allows us to study the quantum properties of subfamilies which span the
transition from integrability to chaos as well as families at approximately
constant degree of chaoticity (Kolmogorov entropy). In the regime of hard chaos
we find that the statistical properties of interest are well-described by
random-matrix theory and completely insensitive to the particular shape of the
dot. However in the nearly-integrable regime non-universal behavior is found.
Specifically, the level-width distribution is well-described by the predicted
distribution both in the presence and absence of magnetic flux when
the system is fully chaotic; however it departs substantially from this
behavior in the mixed regime. The chaotic behavior corroborates the previously
predicted behavior of the peak-height distribution for deformed quantum dots.
We also investigate the energy-level correlation functions which are found to
agree well with the behavior calculated for quasi-zero-dimensional disordered
