    Disruption of aminergic signalling reveals novel compounds with distinct inhibitory effects on mosquito reproduction, locomotor function and survival

    Insecticide resistance amongst disease vectors is a growing problem and novel compounds are needed. Biogenic amines are important for neurotransmission and we have recently shown a potential role for these in mosquito fertility. Here, we dissected the relative contribution of different aminergic signalling pathways to biological processes essential for vectorial capacity such as fertility, locomotion and survival by injecting agonists and antagonists and showed that octopaminergic/tyraminergic signalling is essential for oviposition and hatching rate. We show that egg melanisation is regulated by adrenergic signalling, whose disruption causes premature melanisation specifically through the action of tyramine. In addition to this, co-injection of tyramine with DOPA, the precursor of melanin, had a strong cumulative negative effect on mosquito locomotion and survival. Dopaminergic and serotonergic antagonists such as amitriptyline and citalopram recapitulate this effect. Together these results reveal potential new target sites for the development of future mosquito sterilants and insecticides

    A Chandra Observation of the Nearby Lenticular Galaxy NGC 5102: Where are the X-ray Binaries?

    We present results from a 34 ks Chandra/ACIS-S observation of the nearby (d=3.1 Mpc) lenticular galaxy NGC 5102, previously shown to have an unusually low X-ray luminosity. We detect eleven X-ray point sources within the the D25D_{25} optical boundary of the galaxy (93% of the light), one third to one half of which are likely to be background AGN. One source is coincident with the optical nucleus and may be a low-luminosity AGN. Only two sources with an X-ray luminosity greater than 1037^{37} ergs s1^{-1} in the 0.5-5.0 keV band were detected, one of which is statistically likely to be a background AGN. We expected to detect 6 such luminous sources if the XRB population scales linearly with optical magnitude of the host galaxy. NGC 5102 has an unusually low number of XRBs. NGC 5102 is unusually blue for its morphological type, and has undergone at least two recent bursts of star formation. We present the results of optical/UV spectral synthesis analysis and demonstrate that a significant fraction (>>50%) of the stars in this galaxy are comparatively young (<3×109<3\times10^9 years old). If the lack of X-ray binaries is related to the relative youth of most of the stars, this would support models of LMXB formation and evolution that require wide binaries to shed angular momentum on a timescale of Gyrs. We find that NGC 5102 has an unusually low specific frequency of globular clusters (SNS_N\sim0.4), which could also explain the lack of LMXBs. We also detect diffuse X-ray emission in the central \sim1 kpc of the galaxy. This hot gas is most likely a superbubble created by multiple supernovae of massive stars born during the most recent star burst, and is driving the shock into the ISM which was inferred from optical observations.Comment: 33 pages, 7 figures, 6 tables - Accepted for publication in the Astrophysical Journa

    HD148937: a multiwavelength study of the third Galactic member of the Of?p class

    Three Galactic O-type stars belong to the rare class of Of?p objects: HD108, HD191612, and HD148937. The first two stars show a wealth of phenomena, including magnetic fields and strong X-ray emission, light variability, and dramatic periodic spectral variability. We present here the first detailed optical and X-ray study of the third Galactic Of?p star, HD148937. Spectroscopic monitoring has revealed low-level variability in the Balmer and HeII4686 lines, but constancy at HeI and CIII4650. The Ha line exhibits profile variations at a possible periodicity of ~7d. Model atmosphere fits yield T_{eff}=41000+-2000K, log(g)=4.0+-0.1, Mdot_{sph}<~ 10^{-7}Msol/yr and a surabondance of nitrogen by a factor of four. At X-ray wavelengths, HD148937 resembles HD108 and HD191612 in having a thermal spectrum dominated by a relatively cool component (kT=0.2keV), broad lines (>1700km/s), and an order-of-magnitude overluminosity compared to normal O stars (log [L_X^unabs/L_BOL] ~ -6).Comment: accepted by AJ; 15p, 15fig available in jp

    The Central Singularity in Spherical Collapse

    The gravitational strength of the central singularity in spherically symmetric space-times is investigated. Necessary conditions for the singularity to be gravitationally weak are derived and it is shown that these are violated in a wide variety of circumstances. These conditions allow conclusions to be drawn about the nature of the singularity without having to integrate the geodesic equations. In particular, any geodesic with a non-zero amount of angular momentum which impinges on the singularity terminates in a strong curvature singularity.Comment: 17 pages; revised and corrected with improved result

    Deep subcutaneous application of poly-L-lactic acid as a filler for facial lipoatrophy in HIV-infected patients

    Introduction: Facial lipoatrophy is a crucial problem of HIV-infected patients undergoing highly active antiretroviral therapy (HAART). Poly-L-lactic acid (PLA), provided as New-Fill(R)/Sculptra(TM), is known as one possible treatment option. In 2004 PLA was approved by the FDA as Sculptra(TM) for the treatment of lipoatrophy of the face in HIV-infected patients. While the first trials demonstrated relevant efficacy, this was to some extent linked to unwanted effects. As the depth of injection was considered relevant in this context, the application modalities of the preparation were changed. The preparation was to be injected more deeply into subcutaneous tissue, after increased dilution. Material and Methods: To test this approach we performed a pilot study following the new recommendations in 14 patients. Results: While the efficacy turned out to be about the same, tolerability was markedly improved. The increase in facial dermal thickness was particularly obvious in those patients who had suffered from lipoatrophy for a comparatively small period of time. Conclusion: With the new recommendations to dilute PLA powder and to inject it into the deeper subcutaneous tissue nodule formation is a minor problem. However, good treatment results can only be achieved if lipoatrophy is not too intense; treatment intervals should be about 2 - 3 weeks. Copyright (C) 2005 S. Karger AG, Basel
