    Motivic invariants of p-adic fields

    We provide a complete analysis of the motivic Adams spectral sequences converging to the bigraded coefficients of the 2-complete algebraic Johnson-Wilson spectra BPGL over p-adic fields. These spectra interpolate between integral motivic cohomology (n=0), a connective version of algebraic K-theory (n=1), and the algebraic Brown-Peterson spectrum. We deduce that, over p-adic fields, the 2-complete BPGL split over 2-complete BPGL, implying that the slice spectral sequence for BPGL collapses. This is the first in a series of two papers investigating motivic invariants of p-adic fields, and it lays the groundwork for an understanding of the motivic Adams-Novikov spectral sequence over such base fields.Comment: 19 pages, 3 figures.     Spectroscopic techniques and the conservation of artists’ acrylic emulsion paints

    Artists’ acrylic emulsion paints are used in many contexts such as paintings, murals, sculptures, works on paper and mixed media; and are forming increasing proportions of modern and contemporary art collections. Although acrylic emulsion paints have been the focus of museum-led research over the past decade, the impact of artists’ technique and conservation treatment on the upper-most surface of these paints remains essentially unexplored ; This paper summarises previous studies using vibrational (FTIR) spectroscopy and presents initial assessments of paint surfaces using X-ray spectroscopies (XPS and NEXAFS) aimed at characterising artists’ acrylic paint film surfaces after natural ageing and wet surface cleaning treatment. Both techniques were found to be well suited for surface-sensitive investigations of the organic materials associated with artists’ acrylic paints, including explorations into: (A) cleaning system residues, (B) surfactant extraction from paint surfaces, (C) the identification of migrated surfactant, and (D) monitoring pigment changes at the paint/air interface of paint films ; It has been shown is that these X-ray spectroscopic techniques can be used for the analysis of almost purely organic materials in a way that complements mass spectroscopic techniques, FTIR and XRF. This investigation forms part of broader, currently ongoing, multi-technique investigation into the properties of artists’ acrylic paints and development of conservation treatments for works-of-art made with these materials

    Motivic Brown-Peterson invariants of the rationals

    Fix the base field Q of rational numbers and let BP denote the family of motivic truncated Brown-Peterson spectra over Q. We employ a "local-to-global" philosophy in order to compute the motivic Adams spectral sequence converging to the bi-graded homotopy groups of BP.     Variation of turbulent burning rate of methane, methanol, and iso-octane air mixtures with equivalence ratio at elevated pressure

    Turbulent burning velocities for premixed methane, methanol, and iso-octane/air mixtures have been experimentally determined for an rms turbulent velocity of 2 m/s and pressure of 0.5 MPa for a wide range of equivalence ratios. Turbulent burning velocity data were derived using high-speed schlieren photography and transient pressure recording; measurements were processed to yield a turbulent mass rate burning velocity, utr. The consistency between the values derived using the two techniques, for all fuels for both fuel-lean and fuel-rich mixtures, was good. Laminar burning measurements were made at the same pressure, temperature, and equivalence ratios as the turbulent cases and laminar burning velocities and Markstein numbers were determined. The equivalence ratio (φ) for peak turbulent burning velocity proved not always coincident with that for laminar burning velocity for the same fuel; for isooctane, the turbulent burning velocity unexpectedly remained high over the range φ = 1 to 2. The ratio of turbulent to laminar burning velocity proved remarkably high for very rich iso-octane/air and lean methane/air mixtures

    Endogenous transforming growth factor β1 suppresses inflammation and promotes survival in adult CNS

    Transforming growth factor β1 (TGFβ1) is a pleiotropic cytokine with potent neurotrophic and immunosuppressive properties that is upregulated after injury, but also expressed in the normal nervous system. In the current study, we examined the regulation of TGFβ1 and the effects of TGFβ1 deletion on cellular response in the uninjured adult brain and in the injured and regenerating facial motor nucleus. To avoid lethal autoimmune inflammation within 3 weeks after birth in TGFβ1-deficient mice, this study was performed on a T- and B-cell-deficient RAG2-/- background. Compared with wild-type siblings, homozygous deletion of TGFβ1 resulted in an extensive inflammatory response in otherwise uninjured brain parenchyma. Astrocytes increased in GFAP and CD44 immunoreactivity; microglia showed proliferative activity, expression of phagocytosis-associated markers [αXβ2, B7.2, and MHC1 (major histocompatibility complex type 1)], and reduced branching. Ultrastructural analysis revealed focal blockade of axonal transport, perinodal damming of axonal organelles, focal demyelination, and myelin debris in granule-rich, phagocytic microglia. After facial axotomy, absence of TGFβ1 led to a fourfold increase in neuronal cell death (52 vs 13%), decreased central axonal sprouting, and significant delay in functional recovery. It also interfered with the microglial response, resulting in a diminished expression of early activation markers [ICAM1 (intercellular adhesion molecule 1), α6β1, and αMβ2] and reduced proliferation. In line with axonal and glial findings in the otherwise uninjured CNS, absence of endogenous TGFβ1 also caused an ∼10% reduction in the number of normal motoneurons, pointing to an ongoing and potent trophic role of this anti-inflammatory cytokine in the normal as well as in the injured brain.     Gene expression and splicing alterations analyzed by high throughput RNA sequencing of chronic lymphocytic leukemia specimens.

    BackgroundTo determine differentially expressed and spliced RNA transcripts in chronic lymphocytic leukemia specimens a high throughput RNA-sequencing (HTS RNA-seq) analysis was performed.MethodsTen CLL specimens and five normal peripheral blood CD19+ B cells were analyzed by HTS RNA-seq. The library preparation was performed with Illumina TrueSeq RNA kit and analyzed by Illumina HiSeq 2000 sequencing system.ResultsAn average of 48.5 million reads for B cells, and 50.6 million reads for CLL specimens were obtained with 10396 and 10448 assembled transcripts for normal B cells and primary CLL specimens respectively. With the Cuffdiff analysis, 2091 differentially expressed genes (DEG) between B cells and CLL specimens based on FPKM (fragments per kilobase of transcript per million reads and false discovery rate, FDR q < 0.05, fold change >2) were identified. Expression of selected DEGs (n = 32) with up regulated and down regulated expression in CLL from RNA-seq data were also analyzed by qRT-PCR in a test cohort of CLL specimens. Even though there was a variation in fold expression of DEG genes between RNA-seq and qRT-PCR; more than 90 % of analyzed genes were validated by qRT-PCR analysis. Analysis of RNA-seq data for splicing alterations in CLL and B cells was performed by Multivariate Analysis of Transcript Splicing (MATS analysis). Skipped exon was the most frequent splicing alteration in CLL specimens with 128 significant events (P-value <0.05, minimum inclusion level difference >0.1).ConclusionThe RNA-seq analysis of CLL specimens identifies novel DEG and alternatively spliced genes that are potential prognostic markers and therapeutic targets. High level of validation by qRT-PCR for a number of DEG genes supports the accuracy of this analysis. Global comparison of transcriptomes of B cells, IGVH non-mutated CLL (U-CLL) and mutated CLL specimens (M-CLL) with multidimensional scaling analysis was able to segregate CLL and B cell transcriptomes but the M-CLL and U-CLL transcriptomes were indistinguishable. The analysis of HTS RNA-seq data to identify alternative splicing events and other genetic abnormalities specific to CLL is an added advantage of RNA-seq that is not feasible with other genome wide analysis

    Degradation studies of hydrophilic, partially degradable and bioactive cements (HDBCs) incorporating chemically modified starch

    The degradation rate in Hydrophilic, Degradable and Bioactive Cements (HDBCs) containing starch/cellulose acetate blends (SCA) is still low. In order to increase degradation, higher amounts of starch are required to exceed the percolation threshold. In this work, gelatinization, acetylation and methacrylation of corn starch were performed and assessed as candidates to replace SCA in HDBCs. Formulations containing methacrylated starch were prepared with different molar ratios of 2-hydroxyethyl methacrylate and methyl methacrylate in the liquid component and the amount of residual monomer released into water was evaluated. The concentration of reducing sugars, percentage of weight loss and morphologic analyses after degradation all confirmed increased degradation of HDBC with alpha-amylase, with the appearance of pores and voids from enzymatic action. Methacrylated starch therefore is a better alternative to be used as the solid component of HDBC then SCA, since it leads to the formation of cements with a lower release of toxic monomers and more prone to hydrolytic degradation while keeping the other advantages of HDBCs.The authors acknowledge to Foundation for Science and Technology (FCT), who supported this study through funds from project Concept2Cement (POCTI/CTM/60735/2004)

    Model structures on finite total orders

    We initiate the study of model structures on (categories induced by) lattice posets, a subject we dub homotopical combinatorics. In the case of a finite total order [n][n], we enumerate all model structures, exhibiting a rich combinatorial structure encoded by Shapiro's Catalan triangle.
