    The role of Stewartson and Ekman layers in turbulent rotating Rayleigh-B\'enard convection

    When the classical Rayleigh-B\'enard (RB) system is rotated about its vertical axis roughly three regimes can be identified. In regime I (weak rotation) the large scale circulation (LSC) is the dominant feature of the flow. In regime II (moderate rotation) the LSC is replaced by vertically aligned vortices. Regime III (strong rotation) is characterized by suppression of the vertical velocity fluctuations. Using results from experiments and direct numerical simulations of RB convection for a cell with a diameter-to-height aspect ratio equal to one at Ra108109Ra \sim 10^8-10^9 (Pr=46Pr=4-6) and 01/Ro250 \lesssim 1/Ro \lesssim 25 we identified the characteristics of the azimuthal temperature profiles at the sidewall in the different regimes. In regime I the azimuthal wall temperature profile shows a cosine shape and a vertical temperature gradient due to plumes that travel with the LSC close to the sidewall. In regime II and III this cosine profile disappears, but the vertical wall temperature gradient is still observed. It turns out that the vertical wall temperature gradient in regimes II and III has a different origin than that observed in regime I. It is caused by boundary layer dynamics characteristic for rotating flows, which drives a secondary flow that transports hot fluid up the sidewall in the lower part of the container and cold fluid downwards along the sidewall in the top part.Comment: 21 pages, 12 figure

    Compensation in resting metabolism for experimentally increased activity

    To study zebra finch allocation of energy to day and night at two different workloads, we assessed the daily energy turnover from: (1) metabolizable energy of the food, and (2) doubly-labeled water. In both experiments we imposed two levels of activity on captive zebra finches (Taeniopygia guttata), by applying different computer-controlled workload schedules. A low workload required 20 hops, and a high workload 40 hops to obtain 10 s access to food. In experiment 1, we further measured nocturnal energy expenditure by overnight oxygen consumption. From experiment 2 we derived an estimate of the costs of hopping activity, from inter-individual association of daily amount of hopping and daily energy expenditure. Surprisingly, the daily energy budget was, on average, reduced slightly when birds were subjected to a high workload. Since hopping activity was 50% higher during the high workload than during the low workload, the birds apparently compensated, even over-compensated, for the increased energetic demands of activity. Nocturnal energy expenditure was indeed reduced for the high workload, which was largely due to a reduction in resting metabolic rate. Economizing on energy was more than could have been accomplished by a reduction in mass alone, and we discuss the occurrence and potential mechanisms of physiological compensation. The amount of energy saved during the night did account for part of the total amount of energy saved. We surmise that the strategy of energetic compensation observed during the night was extended into the inactive hours of the day

    2D and 3D similarity landscape analysis identifies PARP as a novel off-target for the drug Vatalanib

    Background Searching for two-dimensional (2D) structural similarities is a useful tool to identify new active compounds in drug-discovery programs. However, as 2D similarity measures neglect important structural and functional features, similarity by 2D might be underestimated. In the present study, we used combined 2D and three-dimensional (3D) similarity comparisons to reveal possible new functions and/or side-effects of known bioactive compounds. Results We utilised more than 10,000 compounds from the SuperTarget database with known inhibition values for twelve different anti-cancer targets. We performed all-against-all comparisons resulting in 2D similarity landscapes. Among the regions with low 2D similarity scores are inhibitors of vascular endothelial growth factor receptor (VEGFR) and inhibitors of poly ADP-ribose polymerase (PARP). To demonstrate that 3D landscape comparison can identify similarities, which are untraceable in 2D similarity comparisons, we analysed this region in more detail. This 3D analysis showed the unexpected structural similarity between inhibitors of VEGFR and inhibitors of PARP. Among the VEGFR inhibitors that show similarities to PARP inhibitors was Vatalanib, an oral “multi-targeted” small molecule protein kinase inhibitor being studied in phase-III clinical trials in cancer therapy. An in silico docking simulation and an in vitro HT universal colorimetric PARP assay confirmed that the VEGFR inhibitor Vatalanib exhibits off-target activity as a PARP inhibitor, broadening its mode of action. Conclusion In contrast to the 2D-similarity search, the 3D-similarity landscape comparison identifies new functions and side effects of the known VEGFR inhibitor Vatalanib
