    Polypeptide-grafted macroporous polyHIPE by surface-initiated N-Carboxyanhydride (NCA) polymerization as a platform for bioconjugation

    A new class of functional macroporous monoliths from polymerized high internal phase emulsion (polyHIPE) with tunable surface functional groups was developed by direct polypeptide surface grafting. In the first step, amino-functional polyHIPEs were obtained by the addition of 4-vinylbenzyl or 4-vinylbenzylphthalimide to the styrenic emulsion and thermal radical polymerization. The obtained monoliths present the expected open-cell morphology and a high surface area. The incorporated amino group was successfully utilized to initiate the ring-opening polymer- ization of benzyl-L-glutamate N-carboxyanhydride (BLG NCA) and benzyloxycarbonyl-L-lysine (Lys(Z)) NCA, which resulted in a dense homogeneous coating of polypeptides throughout the internal polyHIPE surfaces as confirmed by SEM and FTIR analysis. The amount of polypeptide grafted to the polyHIPE surfaces could be modulated by varying the initial ratio of amino acid NCA to amino-functional polyHIPE. Subsequent removal of the polypeptide protecting groups yielded highly functional polyHIPE-g-poly(glutamic acid) and polyHIPE-g- poly(lysine). Both types of polypeptide-grafted monoliths responded to pH by changes in their hydrohilicity. The possibility to use the high density of function (−COOH or −NH2) for secondary reaction was demonstrated by the successful bioconjugation of enhanced green fluorescent protein (eGFP) and fluorescein isocyanate (FITC) on the polymer 3D-scaffold surface. The amount of eGFP and FITC conjugated to the polypeptide-grafted polyHIPE was significantly higher than to the amino- functional polyHIPE, signifying the advantage of polypeptide grafting to achieve highly functional polyHIPEs

    What Explains the Redistribution Achieved by the Italian Personal Income Tax? Evidence from Administrative Data

    We analyze the Italian personal income tax (PIT) in the light of the different tools available to the government to achieve income redistribution. Wefocus in particular on three mechanisms: marginal tax rates, deductions, and tax credits. Exploiting an extended version of the standard Pfahler decomposition, we estimate the contribution of each of these three tools to the overall redistributive effect of the PIT using administrative data on more than 1.3 million individual tax returns. Our estimates suggest that more than half of the total PIT redistributive effect is due to the two most important tax credits (the tax credit for employment and the tax credit for retirement income), while the marginal rates schedule contribution is about 40 percent. On the contrary, most of the itemized expenditures do not show any sizable impact on redistribution

    Good or Bad? Understanding the Effects Over Time of Multigrading on Child Achievement

    Multigrading represents the practice of mixing children of different ages in the same classroom. This paper examines the effect of attending a multigrade class in Grade 2 on students’ academic achievement in Grades 2, 5, and 8, respectively, considering Italy as a case study. To address the issue of endogeneity of multigrading (and class size), we adopt an IV identification strategy based on a law that disciplines class composition. We show that multigrading has a positive (16 percent of a standard deviation) short-term effect on academic achievements. However, this effect diminishes over time and becomes negative (-10 percent of a standard deviation) if students spend several years in a multigrade class. Mechanism analysis indicates the fundamental role of teachers and suggests that the negative long-term effect of multigrading is not statistically different from zero when multigrade classes are taught by more experienced teachers. These findings, based on longitudinal data, reconcile contrasting results in the literature, which are based on cross-sectional data and on the short-term effects of multigrading

    The availability of cheap Wi-Fi internet connections has encouraged schools to adopt Web 2.0 platforms for teaching, with the intention of stimulating students’ academic achievement and participation in school. Moreover, during the recent explosion of the SARS-CoV-2 crisis that forced many countries to close schools (as well as offices and factories), the widespread diffusion of these applications kept school systems going. Despite their widespread use as teaching tools, the effect of adopting Web 2.0 platforms on students’ performance has never been rigorously tested. We fill this gap in the literature by analyzing the impact of using Twitter as a teaching tool on high school students’ literature skills. Based on a large-scale, randomized controlled trial that involved 70 schools and about 1,500 students, we find that using Twitter to teach literature has an overall negative effect on students’ average achievement, reducing standardized test scores by about 25 percent of a standard deviation. The negative effect is stronger on students who usually perform better

    Functional reconstruction of the glenoid fossa utilizing a pedicled temporal osteomuscular flap

    Current techniques in management of end stage pathology of the temporomandibular joint (TMJ) include the use of alloplastic joint reconstruction. A polyethylene glenoid fossa prosthesis is a necessity of this treatment as it provides a stable platform for function of the metal alloy condylar head. Additionally, the fossa prosthesis limits superior and posterior movement of the reconstructed joint which prevents complications such as migration of the condylar prosthesis into the middle cranial fossa and ear, ankylosis, and pain. When a pathologic process affects the glenoid fossa alone, alloplastic joint reconstruction becomes a less desirable treatment option. Lack of osseous structure along the temporal bone and zygomatic arch can impact the surgeon\u27s ability to fixate a glenoid fossa prosthesis. Additionally, resection of an uninvolved condylar head in situations where there is no advanced pathology would provide a functional solution, but may be overly aggressive and potentially unnecessary. The following is our experience with utilizing a pedicled temporal osteomuscular flap to reconstruct an acquired defect of the glenoid fossa in a 42-year-old male with a diffuse-type tenosynovial giant cell tumor. In this case the mandibular condyle was not affected by the pathology

    COVID-19 Pandemic: Brief Overview of the Consequences on Family Informal Caregiving

    Background and aim of the work: The COVID-19 pandemic has deeply affected the quality of people’s social life, strongly impacting family dynamics, too, not only in the harshest periods of the pandemic but also afterwards. Pandemic-related measures led to a ‘stay-at-home’ approach that increased the mental and physical burdens of family caregivers, irrespective of whether they were living together with the person they were caring for or not. In this paper, we provide an overview of the impact that the COVID-19 pandemic had on family relationships and dynamics, as well as on family caregivers’ mental burden, and outline how this developed. Methods: We collected relevant info by searching the PubMed/Medline database with appropriate keywords. The search was performed up to 28 February 2023. This paper is reported in line with PRISMA guidance. Results: Given the recent onset of the issue, the number of relevant papers was limited. However, the additional burden that the pandemic has caused worldwide to informal caregivers clearly emerges. Conclusions: The worldwide impact of the pandemic on informal caregiving is assessed, and recommendations on how the issue can be handled are briefly sketched, too

    Il contributo analizza l’evoluzione del ruolo della religione nei Peace Studies, da ‘elemento integrato’ e marginale a variabile autonoma e strategica nei processi di analisi, gestione e trasformazione dei conflitti. Muovendo dal paradigma del ‘conflitto costruttivo’, si evidenzia il potenziale ambivalente della religione, capace tanto di alimentare dinamiche di radicalizzazione e polarizzazione quanto di favorire percorsi di riconciliazione. Attraverso un approccio multidisciplinare, si esamina il ruolo delle retoriche religiose, intese come dispositivi comunicativi interni alle organizzazioni, nella loro funzione propedeutica al prosieguo del conflitto e alla risoluzione di problematiche di varia natura; successivamente, si propone una riflessione sugli strumenti di ‘religious diplomacy’, evidenziando e valorizzando come il dialogo interreligioso e l’alfabetizzazione religiosa possano rafforzare i processi di peacebuilding. In questa prospettiva, la variabile religiosa emerge come elemento chiave per comprendere e trasformare le dinamiche di conflitto, contribuendo alla costruzione di una pace inclusiva, stabile e duratura.The article explores the evolving role of religion within Peace Studies, highlighting its shift from a marginal and ‘integrated presence’ to an autonomous and strategic variable in the analysis, management and transformation of conflicts. Adopting the paradigm of ‘constructive conflict’, it explores the ambivalent role of religion, which may serve either to intensify dynamics of radicalization and polarization or to foster reconciliation and social cohesion. Through a multidisciplinary approach, this study investigates the role of religious rhetoric as an internal communicative device within organizations, functioning both as a catalyst for conflicts and as a means of managing different challenges; furthermore, the paper reflects on the tools of ‘religious diplomacy’, emphasizing how interreligious dialogue and religious literacy can reinforce peacebuilding efforts. In this perspective, the religious variable emerges as a key factor for interpreting and transforming conflict dynamics, ultimately contributing to the establishment of an inclusive and lasting peace

    This paper studies the effect of multigrading—mixing children of different ages in the same classroom—on students’ short- versus long-term academic achievement in Italy. We cope with the endogeneity of multigrading (and class size) through an instrumental variable identification strategy based on a law that disciplines class composition. By relying on longitudinal data that follow a cohort of Italian students over their compulsory school career, we show that multigrading has a positive short-term effect on achievements. This effect fades away over time to become negative in the long run if students spend several years in a multigrade class. The analysis of mechanisms points to the fundamental role of teachers and suggests that no negative long-term effect arises when multigrade classes are taught by more experienced and motivated teachers. These results reconcile contrasting findings in the literature based on cross-sectional data and a short-term focus

    Estimating velocity distribution and flood discharge at river bridges using entropy theory – insights from computational fluid dynamics flow fields

    Estimating the flow velocity and discharge in rivers is of particular interest for monitoring, modeling, and research purposes. Instruments for measuring water level and surface velocity are generally mounted on bridge decks, and this poses a challenge because the bridge structure, with piers and abutments, can perturb the flow field. The current research aims to investigate the applicability of entropy theory to estimate the velocity distribution and the discharge in the vicinity of river bridges. For this purpose, a computational fluid dynamics (CFD) model is used to obtain three-dimensional flow fields along a stretch of the Paglia River (central Italy), where a historical multi-arch bridge strongly affects flood flows. The input data for the entropy model include the cross-sectional bathymetry and the surface velocity provided by the numerical simulations. A total of 12 samples, including three different flow conditions at four cross-sections, one upstream and three downstream of the bridge, are considered. It is found that the entropy model can be reliably applied upstream of the bridge, also when forced with a single (i.e., the maximum) value of the surface velocity, with errors on total discharge below 13 % in the considered case. By contrast, downstream of the bridge, the wakes generated by the bridge piers strongly affect the velocity distribution, both in the spanwise and in the vertical directions and for very long distances. Here, notwithstanding the complex and multimodal spanwise distribution of flow velocity, the entropy model estimates the discharge with error lower than 8 % if forced with the river-wide distribution of the surface velocity. The present study has important implications for the optimal positioning of sensors and suggests the potential of using CFD modeling and entropy theory jointly to foster greater knowledge of river systems.</p

    Effects of anti-IL5 biological treatments on blood IgE levels in severe asthmatic patients: A real-life multicentre study (BIONIGE)

    Background: Mepolizumab and benralizumab are clinically effective biological treatments for severe eosinophilic asthmatic patients by hampering eosinophilic inflammation. The effects of these compound on the immunoglobulin (Ig)E T2 component are virtually unknown. Objectives: To evaluate the change in total IgE levels at 4&nbsp;±&nbsp;2&nbsp;months after initiation of the mepolizumab (primary outcome) or benralizumab. When available, the changes of blood inflammatory cell counts, lung function and asthma control test (ACT) were also assessed and correlated with changes in total IgE levels. Methods: Observational, retrospective, multicentre, cohort study. Severe eosinophilic atopic asthmatic patients treated with mepolizumab or benralizumab were included in the analysis. Results: Three-month treatment (on average) with mepolizumab (n&nbsp;=&nbsp;104) or benralizumab (n&nbsp;=&nbsp;82) resulted in significantly higher reduction of blood eosinophil and basophil levels in patients treated with benralizumab compared to mepolizumab. Mepolizumab did not significantly modified the levels of blood total IgE during the study period, whereas benralizumab significantly reduced (−35%, p&nbsp;&lt;&nbsp;0.001) total blood IgE levels. In patients treated with benralizumab the reduction of blood total Ig-E levels correlated with the reduction of blood basophils (but not eosinophils) and weakly with the improvement of asthma control. Conclusion: Benralizumab but not mepolizumab, treatment led to a significant reduction of circulating IgE level.
