    In vitro Evaluation der lateralen Guttapercha-Kondensationstechnik mit Fingerspreadern verschiedener Spitzengeometrie.

    Ziel: Ziel der Studie war es, den Einfluss von Spreaderform und Aufbereitungstaper bei der lateralen Kondensation auf die Dichtigkeit der Wurzelfüllungen zu untersuchen. Für diese Studie wurde ein handelsüblicher Spreader so modifiziert, dass seine Arbeitsspitze durch Abflachung Pluggerfunktionalität erhält. Material&Methode: Insgesamt n=75 frisch extrahierte menschliche Zähne mit geraden Wurzeln wurden so vorbereitet, dass 75 gerade Zahnwurzeln. Diese Zahnwurzeln wurden randomisiert so auf 5 Gruppen aufgeteilt, mit rotierenden Ni/Ti Instrumenten vom Typ Profile 02/50 und 04/50 aufbereitet und unter Verwendung der lateraler Kondensation mit den konventionellen Spreadern „pointed“ sowie modifizierten Spreadern "flat tip“ gefüllt. Eine negative Kontrollgruppe wurde unter Anwendung der Zentralstifttechnik ohne Sealer abgefüllt. Die Wurzeln wurden bis auf 1 mm oberhalb des Apex mit Nagellack überzogen und anschließend verblindet. Nach Lagerung in 2%-Methylenblau für 4 Tage wurden die Proben in Kaltpolymerisat eingebettet und es wurden sechs serielle Querschnitte im Abstand von 0,8 mm ab dem Apex angefertigt. Die Querschnitte wurden digitalisiert. Bewertet wurden die Penetrationstiefe in mm ab dem Apex und der Penetrationsgrad. Ergebnisse: Alle Proben zeigten eine ausgeprägte Penetration. Lediglich der Parameter Penetrationsgrad konnte für eine Differenzierung zwischen den Prüfgruppen verwendet werden. Es zeigte sich ein signifikanter Unterschied zwischen der Kontrollgruppe und allen Prüfgruppen. Innerhalb der Prüfgruppen, welche als normalverteilt und varianzhomogen befunden wurden, wurde eine zweifaktorielle Varianzanalyse für die Faktoren "Spreader" und "Taper" durchgeführt. Es ergab sich für den Faktor "Spreader" ein signifikanter Unterschied (p=0,030) ebenso wie für den Faktor "Taper" (p=0,006). Wurzelfüllungen welche mit dem modifizierten Spreader angefertigt wurden zeigen demnach eine statistisch signifikant schlechtere Dichtigkeit (gemessen mittels Penetrationsgrad) als solche die mit konventionellem Spreader kondensiert wurden. Eine Aufbereitung auf einen Taper von 0,04 führt zu einer statistisch signifikant besseren Dichtigkeit (gemessen mittels Penetrationsgrad) als eine Aufbereitung auf einen ISO-normkonformen Taper von 0,02. Schlussfolgerung: Es konnte gezeigt werden, dass mit dem gewählten Spreaderdesign kein zusätzlicher Nutzen verbunden ist, ja sogar die Dichtigkeit der resultierenden Wurzelfüllungen abnahm. Es ist also nicht angezeigt, Spreader in der beschriebenen Form klinisch zu verwenden. Weiterhin konnte gezeigt werden, dass die Verwendung eines größeren Aufbereitungstapers auch bei lateraler Kondensation zu dichteren Füllungen führt. Klinisch sollte daher ein entsprechend großer Aufbereitungskonus angestrebt werden

    Nonlinearity of Mechanochemical Motions in Motor Proteins

    The assumption of linear response of protein molecules to thermal noise or structural perturbations, such as ligand binding or detachment, is broadly used in the studies of protein dynamics. Conformational motions in proteins are traditionally analyzed in terms of normal modes and experimental data on thermal fluctuations in such macromolecules is also usually interpreted in terms of the excitation of normal modes. We have chosen two important protein motors - myosin V and kinesin KIF1A - and performed numerical investigations of their conformational relaxation properties within the coarse-grained elastic network approximation. We have found that the linearity assumption is deficient for ligand-induced conformational motions and can even be violated for characteristic thermal fluctuations. The deficiency is particularly pronounced in KIF1A where the normal mode description fails completely in describing functional mechanochemical motions. These results indicate that important assumptions of the theory of protein dynamics may need to be reconsidered. Neither a single normal mode, nor a superposition of such modes yield an approximation of strongly nonlinear dynamics.Comment: 10 pages, 6 figure

    Maternal corticotropin-releasing hormone is associated with LEP DNA methylation at birth and in childhood: an epigenome-wide study in Project Viva

    BackgroundCorticotropin-releasing hormone (CRH) plays a central role in regulating the secretion of cortisol which controls a wide range of biological processes. Fetuses overexposed to cortisol have increased risks of disease in later life. DNA methylation may be the underlying association between prenatal cortisol exposure and health effects. We investigated associations between maternal CRH levels and epigenome-wide DNA methylation of cord blood in offsprings and evaluated whether these associations persisted into mid-childhood.MethodsWe investigated mother-child pairs enrolled in the prospective Project Viva pre-birth cohort. We measured DNA methylation in 257 umbilical cord blood samples using the HumanMethylation450 Bead Chip. We tested associations of maternal CRH concentration with cord blood cells DNA methylation, adjusting the model for maternal age at enrollment, education, maternal race/ethnicity, maternal smoking status, pre-pregnancy body mass index, parity, gestational age at delivery, child sex, and cell-type composition in cord blood. We further examined the persistence of associations between maternal CRH levels and DNA methylation in children's blood cells collected at mid-childhood (n = 239, age: 6.7-10.3 years) additionally adjusting for the children's age at blood drawn.ResultsMaternal CRH levels are associated with DNA methylation variability in cord blood cells at 96 individual CpG sites (False Discovery Rate <0.05). Among the 96 CpG sites, we identified 3 CpGs located near the LEP gene. Regional analyses confirmed the association between maternal CRH and DNA methylation near LEP. Moreover, higher maternal CRH levels were associated with higher blood-cell DNA methylation of the promoter region of LEP in mid-childhood (P < 0.05, β = 0.64, SE = 0.30).ConclusionIn our cohort, maternal CRH was associated with DNA methylation levels in newborns at multiple loci, notably in the LEP gene promoter. The association between maternal CRH and LEP DNA methylation levels persisted into mid-childhood

    Two-Loop Soft Corrections and Resummation of the Thrust Distribution in the Dijet Region

    The thrust distribution in electron-positron annihilation is a classical precision QCD observable. Using renormalization group (RG) evolution in Laplace space, we perform the resummation of logarithmically enhanced corrections in the dijet limit, T1T\to 1 to next-to-next-to-leading logarithmic (NNLL) accuracy. We independently derive the two-loop soft function for the thrust distribution and extract an analytical expression for the NNLL resummation coefficient g3g_3. To combine the resummed expressions with the fixed-order results, we derive the log(R)\log(R)-matching and RR-matching of the NNLL approximation to the fixed-order NNLO distribution.Comment: 50 pages, 12 figures, 1 table.     Deuteron and antideuteron production in Au+Au collisions at sqrt(s_NN)=200 GeV

    The production of deuterons and antideuterons in the transverse momentum range 1.1 < p_T < 4.3 GeV/c at mid-rapidity in Au + Au collisions at sqrt(s_NN)=200 GeV has been studied by the PHENIX experiment at RHIC. A coalescence analysis comparing the deuteron and antideuteron spectra with those of protons and antiprotons, has been performed. The coalescence probability is equal for both deuterons and antideuterons and increases as a function of p_T, which is consistent with an expanding collision zone. Comparing (anti)proton yields p_bar/p = 0.73 +/- 0.01, with (anti)deuteron yields: d_bar/d = 0.47 +/- 0.03, we estimate that n_bar/n = 0.64 +/- 0.04.Comment: 326 authors, 6 pages text, 5 figures, 1 Table.     Single Electrons from Heavy Flavor Decays in p+p Collisions at sqrt(s) = 200 GeV

    The invariant differential cross section for inclusive electron production in p+p collisions at sqrt(s) = 200 GeV has been measured by the PHENIX experiment at the Relativistic Heavy Ion Collider over the transverse momentum range $0.4 <= p_T <= 5.0 GeV/c at midrapidity (eta <= 0.35). The contribution to the inclusive electron spectrum from semileptonic decays of hadrons carrying heavy flavor, i.e. charm quarks or, at high p_T, bottom quarks, is determined via three independent methods. The resulting electron spectrum from heavy flavor decays is compared to recent leading and next-to-leading order perturbative QCD calculations. The total cross section of charm quark-antiquark pair production is determined as sigma_(c c^bar) = 0.92 +/- 0.15 (stat.) +- 0.54 (sys.) mb.Comment: 329 authors, 6 pages text, 3 figures.     Prediction of photoperiodic regulators from quantitative gene circuit models

    Photoperiod sensors allow physiological adaptation to the changing seasons. The external coincidence hypothesis postulates that a light-responsive regulator is modulated by a circadian rhythm. Sufficient data are available to test this quantitatively in plants, though not yet in animals. In Arabidopsis, the clock-regulated genes CONSTANS (CO) and FLAVIN, KELCH, F-BOX (FKF1) and their lightsensitive proteins are thought to form an external coincidence sensor. We use 40 timeseries of molecular data to model the integration of light and timing information by CO, its target gene FLOWERING LOCUS T (FT), and the circadian clock. Among other predictions, the models show that FKF1 activates FT. We demonstrate experimentally that this effect is independent of the known activation of CO by FKF1, thus we locate a major, novel controller of photoperiodism. External coincidence is part of a complex photoperiod sensor: modelling makes this complexity explicit and may thus contribute to crop improvement

    Observation of associated near-side and away-side long-range correlations in √sNN=5.02  TeV proton-lead collisions with the ATLAS detector

    Two-particle correlations in relative azimuthal angle (Δϕ) and pseudorapidity (Δη) are measured in √sNN=5.02  TeV p+Pb collisions using the ATLAS detector at the LHC. The measurements are performed using approximately 1  μb-1 of data as a function of transverse momentum (pT) and the transverse energy (ΣETPb) summed over 3.1<η<4.9 in the direction of the Pb beam. The correlation function, constructed from charged particles, exhibits a long-range (2<|Δη|<5) “near-side” (Δϕ∼0) correlation that grows rapidly with increasing ΣETPb. A long-range “away-side” (Δϕ∼π) correlation, obtained by subtracting the expected contributions from recoiling dijets and other sources estimated using events with small ΣETPb, is found to match the near-side correlation in magnitude, shape (in Δη and Δϕ) and ΣETPb dependence. The resultant Δϕ correlation is approximately symmetric about π/2, and is consistent with a dominant cos⁡2Δϕ modulation for all ΣETPb ranges and particle pT
