    Successive translocation into and out of the mitochondrial matrix

    We investigated the import and sorting pathways of cytochrome b2 and cytochrome c1, which are functionally located in the intermembrane space of mitochondria. Both proteins are synthesized on cytoplasmic ribosomes as larger precursors and are processed in mitochondria in two steps upon import. The precursors are first translocated across both mitochondrial membranes via contact sites into the matrix. Processing by the matrix peptidase leads to intermediate-sized forms, which are subsequently redirected across the inner membrane. The second proteolytic processing occurs in the intermembrane space. We conclude that the hydrophobic stretches in the presequences of the intermediate-sized forms do not stop transfer across the inner membrane, but rather act as transport signals to direct export from the matrix into the intermembrane space

    Mitochondrial precursor proteins are imported through a hydrophilic membrane environment

    We have analyzed how translocation intermediates of imported mitochondrial precursor proteins, which span contact sites, interact with the mitochondrial membranes. F1-ATPase subunit β(F1β) was trapped at contact sites by importing it into Neurospora mitochondria in the presence of low levels of nucleoside triphosphates. This F1β translocation intermediate could be extracted from the membranes by treatment with protein denaturants such as alkaline pH or urea. By performing import at low temperatures, the ADP/ATP carrier was accumulated in contact sites of Neurospora mitochondria and cytochrome b2 in contact sites of yeast mitochondria. These translocation intermediates were also extractable from the membranes at alkaline pH. Thus, translocation of precursor proteins across mitochondrial membranes seems to occur through an environment which is accessible to aqueous perturbants. We propose that proteinaceous structures are essential components of a translocation apparatus present in contact sites

    Modeling The Fate of Sumatran Elephants in Bukit Tigapuluh Indonesia: Research Needs & Implications for Population Management

    The critically endangered Sumatran elephant persists in mainly small and isolated populations that may require intensive management to be viable in the long term. Population Viability Analysis (PVA) provides the opportunity to evaluate conservation strategies and objectives prior to implementation, which can be very valuable for site managers by supporting their decision making process. This study applies PVA to a local population of Sumatran elephants roaming the Bukit Tigapuluh landscape, Sumatra, with the main goal to explore the impact of pre-selected conservation measures and population scenarios on both population growth rate and extinction probability. Sensitivity testing revealed considerable parameter uncertainties that should be addressed by targeted research projects in order to improve the predictive power of the baseline population model. Analisis Viabilitas Populasi (Population Viability Analysis, PVA) berpeluang untuk digunakan sebagai sarana evaluasi atas tujuan dan strategi konservasi yang disusun sebelum implementasi, yang akan sangat bermanfaat bagi pengelola kawasan guna mendukung pengambilan keputusan. Studi ini menggunakan PVA pada populasi lokal gajah Sumatera yang menjelajahi lanskap Bukit Tigapuluh, Sumatera, dengan tujuan utama mengeksplorasi dampak atas skenario upaya konservasi dan populasi terpilih terhadap laju pertumbuhan populasi dan probabilitas kepunahan. Uji sensitivitas menunjukkan adanya ketidakpastian atas sejumlah parameter pokok yang seharusnya diteliti untuk meningkatkan kekuatan prediksi atas baseline model populasi. Mengingat kerusakan habitat yang lebih parah dapat dicegah, untuk itu upaya penangkalan pembunuhan ilegal merupakan prioritas tertinggi di antara upaya-upaya konservasi yang sudah diuji dan menjadi prasyarat wajib untuk menjawab masalah kemungkinan dampak perkawinan sedarah (inbreeding depression) seperti translokasi gajah atau membangun koridor konservasi.

    Implanted Radio Telemetry in Orangutan Reintroduction and Post-release Monitoring and its Application in Other Ape Species

    Designed as a new method to facilitate the reintroduction and post-release monitoring of orangutans and other apes, implanted radio-telemetry (IRT) was developed and first deployed in 2009. Since that time, it has been necessary to collate and review information on its uptake and general efficacy to inform its ongoing development and that of other emerging tracking technologies. We present here technical specifications and the surgical procedure used to implant miniaturized radio transmitters, as well as a formal testing procedure for measuring detectable transmission distances of implanted devices. Feedback from IRT practitioners (veterinarians and field managers) was gathered through questionnaires and is also presented. To date, IRT has been used in at least 250 individual animals (mainly orangutans) from four species of ape in both Asia and Africa. Median surgical and wound healing times were 30 min and 15 days, respectively, with implants needing to be removed on at least 36 separate occasions. Confirmed failures within the first year of operation were 18.1%, while longer distances were reported from positions of higher elevation relative to the focal animal. IRT has been a transformational technology in facilitating the relocation of apes after their release, resulting in much larger amounts of post-release data collection than ever before. It is crucial however, that implant casings are strengthened to prevent the requirement for recapture and removal surgeries, especially for gradually adapting apes. As with all emerging technological solutions, IRT carries with it inherent risk, especially so due to the requirement for subcutaneous implantation. These risks must, however, be balanced with the realities of releasing an animal with no means of relocation, as has historically been, and is still, the case with orangutans and gorillas
