<p>The research goal was to determine the variability in the salivary characteristics, depending on the season, identifying the most important indicators of the salivary characteristics at different degrees of intensity of caries process and the establishment of a correlation between these indicators. Structural characteristics of salivary pools of the students of stomatological faculty have been invented. Object of research. The salivary pools were collected at 8-9 a.m. within 1-1.5 hours after tooth cleaning. The investigation of oral cavity and the biochemical rates were marked twice: in autumn and spring. Results. The tests show, that biochemical characteristics of saliva depend on the caries involvement. It was detected, that caries involvement depends on the region, food characteristics and oral hygiene. It was proved, that saliva has buffers characteristics. Conclusion. These results can be used for explaining the molecular mechanisms of homeostasis disorders in the oral cavity and evaluating the efficiency of preventive and therapeutic measures at early stages of caries.</p

Therapeutic Angiogenesis: Foundations and Practical Application

    Get PDF
Angiogenesis as therapeutic target has emerged since early works by Judah Folkman, yet his “holy grail” was inhibiting vascular growth to block tumor nutrition. However, in modern biomedicine, “therapeutic angiogenesis” became a large field focusing on stimulation of blood vessel growth for ischemia relief to reduce its detrimental effects in the tissues. In this review, we introduce basic principles of tissue vascularization in response to ischemia exploited in this field. An overview of recent status in therapeutic angiogenesis is given with introduction to emerging technologies, including gene therapy, genetic modification of cells ex vivo and tissue engineering

Endothelial and smooth muscle cells derived from human cardiac explants demonstrate angiogenic potential and suitable for design of cell-containing vascular grafts

BACKGROUND: Endothelial and smooth muscle cells are considered promising resources for regenerative medicine and cell replacement therapy. It has been shown that both types of cells are heterogeneous depending on the type of vessels and organs in which they are located. Therefore, isolation of endothelial and smooth muscle cells from tissues relevant to the area of research is necessary for the adequate study of specific pathologies. However, sources of specialized human endothelial and smooth muscle cells are limited, and the search for new sources is still relevant. The main goal of our study is to demonstrate that functional endothelial and smooth muscle cells can be obtained from an available source—post-surgically discarded cardiac tissue from the right atrial appendage and right ventricular myocardium. METHODS: Heterogeneous primary cell cultures were enzymatically isolated from cardiac explants and then grown in specific endothelial and smooth muscle growth media on collagen IV-coated surfaces. The population of endothelial cells was further enriched by immunomagnetic sorting for CD31, and the culture thus obtained was characterized by immunocytochemistry, ultrastructural analysis and in vitro functional tests. The angiogenic potency of the cells was examined by injecting them, along with Matrigel, into immunodeficient mice. Cells were also seeded on characterized polycaprolactone/chitosan membranes with subsequent analysis of cell proliferation and function. RESULTS: Endothelial cells isolated from cardiac explants expressed CD31, VE-cadherin and VEGFR2 and showed typical properties, namely, cytoplasmic Weibel-Palade bodies, metabolism of acetylated low-density lipoproteins, formation of capillary-like structures in Matrigel, and production of extracellular matrix and angiogenic cytokines. Isolated smooth muscle cells expressed extracellular matrix components as well as α-actin and myosin heavy chain. Vascular cells derived from cardiac explants demonstrated the ability to stimulate angiogenesis in vivo. Endothelial cells proliferated most effectively on membranes made of polycaprolactone and chitosan blended in a 25:75 ratio, neutralized by a mixture of alkaline and ethanol. Endothelial and smooth muscle cells retained their functional properties when seeded on the blended membranes. CONCLUSIONS: We established endothelial and smooth muscle cell cultures from human right atrial appendage and right ventricle post-operative explants. The isolated cells revealed angiogenic potential and may be a promising source of patient-specific cells for regenerative medicine. The article examines the history of the development of cybersports journalism, its specifics and features. The object of the research is cybersports journalism, the subject is the development of cybersports journalism in modern media. Research methods – literature analysis method; semantic content analysis; description method. Key conclusions: the phenomenon of cybersports journalism is extremely poorly studied; its specificity is one of the reasons for the low interest in it among researchers. A characteristic feature of cybersports journalism is postanalysis and appeal to the genre of the review. The most popular domestic magazine about video games was «Igromania». The printed magazine was published by Texnomir Publishing House from September 1997 to December 2018 (renamed Igromedia in 2013). Esports journalism is a socially significant activity of people to collect, process and present relevant information about esports through various channels of mass communication (print, radio, television, Internet) using a system of certain genres. At the moment, information about cybersports is provided by online resources: sports.ru , Cybersport.ru

Oligonucleotide Microarrays Identified Potential Regulatory Genes Related to Early Outward Arterial Remodeling Induced by Tissue Plasminogen Activator

Constrictive vascular remodeling limiting blood flow, as well as compensatory outward remodeling, has been observed in many cardiovascular diseases; however, the underlying mechanisms regulating the remodeling response of the vessels remain unclear. Plasminogen activators (PA) are involved in many of the processes of vascular remodeling. We have shown previously that increased levels of tissue-type PA (tPA) contributes to outward vascular remodeling. To elucidate the mechanisms involved in the induction of outward remodeling we characterized changes in the expression profiles of 8799 genes in injured rat carotid arteries 1 and 4 days after recombinant tPA treatment compared to vehicle. Periadventitial tPA significantly increased lumen size and vessel area, encompassed by the external elastic lamina, at both one and 4 days after treatment. Among 41 differentially expressed known genes 1 day after tPA application, five genes were involved in gene transcription, five genes were related to the regulation of vascular tone [for example, thromboxane A2 receptor (D32080) or non-selective-type endothelin receptor (S65355)], and eight genes were identified as participating in vascular innervation [for example, calpain (D14478) or neural cell adhesion molecule L1 (X59149)]. Four days after injury in tPA-treated arteries, four genes, regulating vascular tone, were differentially expressed. Thus, tPA promotes outward arterial remodeling after injury, at least in part, by regulating expression of genes in the vessel wall related to function of the nervous system and vascular tone

Aim. Circulating progenitor endothelial cells (CPCs) play an important role in the regeneration of damaged vascular endothelium. We aimed to study the effect of type 1 diabetes mellitus (DM) on the number of different lines of CPCs.Methods. The number of different lines of CPCs was evaluated by flow cytometry in 45 patients with type 1 DM (mean age: 26,6±6,5 years) of different duration and 14 healthy volunteers.Results. The number of CPCs in the peripheral blood flow of patients with type 1 DM was significanlty reduced compared with that in healthy volunteers and had a reliable inverse correlation with glycated haemoglobin (HbA1c).Conclusions. The number of CPCs in the peripheral blood flow of patients with type 1 DM depended on the level of DM compensation. Of the different lines of CPCs, Lin-CD34+ cells were significantly reduced by 42% (p=0,006), Lin-CD34+c-Kit- cells by 42% (p=0,007) and Lin-CD34+c-Kit+ cells by 41% (p=0,022). Thus, the number of CPCs in the peripheral blood of patients with type 1 DM is reduced with higher levels of HbA1c
