Cooled propylene glycol as a pragmatic choice for preservation of DNA from remote field-collected Diptera for next-generation sequence analysis
Next-generation sequencing (NGS)-based methods can now be applied to large population-scale studies, but this demands very high-quality DNA. For specimens collected from remote field locations, DNA degradation can be a problem, requiring logistically challenging preservation techniques. Simpler preservation techniques are therefore required. Prior to collection of exotic fruit fly (Tephritidae) species, a number of readily available preservatives with storage at either 4°C or room temperature were trialed here to determine the DNA quality for three locally available Diptera species,Fannia canicularis(L.),Musca domesticaL., and Lucilia sericataMeigen. Considerable variation was observed between the different preservatives, species, and temperatures, but several preservatives at 4°C were favored. Chilled propylene glycol was subsequently used for the storage and carriage of Australian field-collectedBactrocera fruit fly specimens to New Zealand. When processed up to 20 d later, DNA fragments of ∼10-20 kb were obtained for successful genotyping by sequencing analysis. This protocol is therefore recommended as a logistically simple and safe approach for distant collection of dipteran samples for NGS population genomic studies

Orthosis reduces breast pain and mechanical forces through natural and augmented breast tissue in women lying prone
BACKGROUND: Breast implant displacement or rupture can cause aesthetic problems and serious medical complications. Activities with prone positioning and loading of the anterior chest wall, such as massage, chiropractic or osteopathic therapies may increase the risk of implant failure and can also cause discomfort in women with natural breast tissue. Here we test the effectiveness of a newly developed orthosis on pain, mechanical pressure and displacement of breast tissue in women with cosmetic augmentation, post-mastectomy reconstruction, lactating or natural breast tissue. METHODS: Thirty-two females volunteers, aged 25–56 years with augmented, reconstructed, natural or lactating breast tissue and cup sizes B-F, participated in this open-label clinical trial. We measured pain perception, peak pressure, maximum force, and breast tissue displacement using different sizes of the orthosis compared to no orthosis. Different densities of the orthosis were also tested in a subgroup of women (n = 7). Pain perception was rated using a validated 11-point visual-analogue scale. Peak pressure and maximum force were assessed using a bilateral set of capacitance-pliance® sensor strips whilst participants were load bearing in a prone position, and breast displacement was measured by magnetic-resonance-imaging. RESULTS: The orthosis significantly reduced pain, breast displacement and mechanical pressures in women with natural and augmented breast tissue in prone position. Greater relief of pain and greater reduction in mechanical forces were found with increased size and density of the orthosis. Use of the orthosis improved overall comfort by 64-100%, lowered peak pressure by up to 85% and maximum force by up to 96%. Medio-lateral displacement of breast tissue was reduced by 16%, resulting in a 51% desirable increase of breast tissue height. CONCLUSION: Our study demonstrated that the newly developed orthosis significantly reduced pain, mechanical pressure and breast tissue displacement in women with augmented and natural breast tissue when lying prone. Our findings are of clinical significance, potentially reducing the risk of complication from prone activities in women with breast augmentation or reconstruction, as well as improving comfort whilst undergoing prone procedures. Closed-Loop Targeted Memory Reactivation during Sleep Improves Spatial Navigation
Sounds associated with newly learned information that are replayed during non-rapid eye movement (NREM) sleep can improve recall in simple tasks. The mechanism for this improvement is presumed to be reactivation of the newly learned memory during sleep when consolidation takes place. We have developed an EEG-based closed-loop system to precisely deliver sensory stimulation at the time of down-state to up-state transitions during NREM sleep. Here, we demonstrate that applying this technology to participants performing a realistic navigation task in virtual reality results in a significant improvement in navigation efficiency after sleep that is accompanied by increases in the spectral power especially in the fast (12\u201315 Hz) sleep spindle band. Our results show promise for the application of sleep-based interventions to drive improvement in real-world tasks
Comparison of candidate architectures for future distributed propulsion aircraft
Turbine engine driven distributed electrical aircraft power systems (also referred to as Turboelectric Distributed Propulsion (TeDP)) are proposed for providing thrust for future aircraft with superconducting components operating at 77K in order for performance and emissions targets to be met. The proposal of such systems presents a radical change from current state-of-the-art aero-electrical power systems. Central to the development of such power systems are architecture design trades which must consider system functionality and performance, system robustness and fault ride-through capability, in addition to the balance between mass and efficiency. This paper presents a quantitative comparison of the three potential candidate architectures for TeDP electrical networks. This analysis provides the foundations for establishing the feasibility of these different architectures subject to design and operational constraints. The findings of this paper conclude that a purely AC synchronous network performs best in terms of mass and efficiency, but similar levels of functionality and controllability to an architecture with electrical decoupling via DC cannot readily be achieved. If power electronic converters with cryocoolers are found to be necessary for functionality and controllability purposes, then studies show that a significant increase in the efficiency of solid state switching components is necessary to achieve specified aircraft performance targets
Architecture, Voltage and Components for a Turboelectric Distributed Propulsion Electric Grid
The development of a wholly superconducting turboelectric distributed propulsion system presents hide unique opportunities for the aerospace industry. However, this transition from normally conducting systems to superconducting systems significantly increases the equipment complexity necessary to manage the electrical power systems. Due to the low technology readiness level (TRL) nature of all components and systems, current Turboelectric Distributed Propulsion (TeDP) technology developments are driven by an ambiguous set of system-level electrical integration standards for an airborne microgrid system (Figure 1). While multiple decades' worth of advancements are still required for concept realization, current system-level studies are necessary to focus the technology development, target specific technological shortcomings, and enable accurate prediction of concept feasibility and viability. An understanding of the performance sensitivity to operating voltages and an early definition of advantageous voltage regulation standards for unconventional airborne microgrids will allow for more accurate targeting of technology development. Propulsive power-rated microgrid systems necessitate the introduction of new aircraft distribution system voltage standards. All protection, distribution, control, power conversion, generation, and cryocooling equipment are affected by voltage regulation standards. Information on the desired operating voltage and voltage regulation is required to determine nominal and maximum currents for sizing distribution and fault isolation equipment, developing machine topologies and machine controls, and the physical attributes of all component shielding and insulation. Partnership, high performance work systems and organizational effectiveness
Using data gathered from 132 organizations operating in Ireland, we examined the impact of high performance work systems (HPWS) and partnership on firm-level performance. Our results reveal that HPWS and partnership practices are positively associated with labour productivity, workplace innovation and negatively associated with voluntary turnover. More specifically, both HPWS and partnership are positively associated with labour productivity and employee retention, and the positive relationship between partnership and workplace innovation is mediated by HPWS
