Symmetric Rotating Wave Approximation for the Generalized Single-Mode Spin-Boson System
The single-mode spin-boson model exhibits behavior not included in the
rotating wave approximation (RWA) in the ultra and deep-strong coupling
regimes, where counter-rotating contributions become important. We introduce a
symmetric rotating wave approximation that treats rotating and counter-rotating
terms equally, preserves the invariances of the Hamiltonian with respect to its
parameters, and reproduces several qualitative features of the spin-boson
spectrum not present in the original rotating wave approximation both
off-resonance and at deep strong coupling. The symmetric rotating wave
approximation allows for the treatment of certain ultra and deep-strong
coupling regimes with similar accuracy and mathematical simplicity as does the
RWA in the weak coupling regime. Additionally, we symmetrize the generalized
form of the rotating wave approximation to obtain the same qualitative
correspondence with the addition of improved quantitative agreement with the
exact numerical results. The method is readily extended to higher accuracy if
needed. Finally, we introduce the two-photon parity operator for the two-photon
Rabi Hamiltonian and obtain its generalized symmetric rotating wave
approximation. The existence of this operator reveals a parity symmetry similar
to that in the Rabi Hamiltonian as well as another symmetry that is unique to
the two-photon case, providing insight into the mathematical structure of the
two-photon spectrum, significantly simplifying the numerics, and revealing some
interesting dynamical properties.Comment: 11 pages, 5 figure
Tn6188 - A Novel Transposon in Listeria monocytogenes Responsible for Tolerance to Benzalkonium Chloride
peer-reviewedControlling the food-borne pathogen Listeria (L.) monocytogenes is of great importance from a food safety perspective, and thus for human health. The consequences of failures in this regard have been exemplified by recent large listeriosis outbreaks in the USA and Europe. It is thus particularly notable that tolerance to quaternary ammonium compounds such as benzalkonium chloride (BC) has been observed in many L. monocytogenes strains. However, the molecular determinants and mechanisms of BC tolerance of L. monocytogenes are still largely unknown. Here we describe Tn6188, a novel transposon in L. monocytogenes conferring tolerance to BC. Tn6188 is related to Tn554 from Staphylococcus (S.) aureus and other Tn554-like transposons such as Tn558, Tn559 and Tn5406 found in various Firmicutes. Tn6188 comprises 5117 bp, is integrated chromosomally within the radC gene and consists of three transposase genes (tnpABC) as well as genes encoding a putative transcriptional regulator and QacH, a small multidrug resistance protein family (SMR) transporter putatively associated with export of BC that shows high amino acid identity to Smr/QacC from S. aureus and to EmrE from Escherichia coli. We screened 91 L. monocytogenes strains for the presence of Tn6188 by PCR and found Tn6188 in 10 of the analyzed strains. These isolates were from food and food processing environments and predominantly from serovar 1/2a. L. monocytogenes strains harboring Tn6188 had significantly higher BC minimum inhibitory concentrations (MICs) (28.5 ± 4.7 mg/l) than strains without Tn6188 (14 ± 3.2 mg/l). Using quantitative reverse transcriptase PCR we could show a significant increase in qacH expression in the presence of BC. QacH deletion mutants were generated in two L. monocytogenes strains and growth analysis revealed that ΔqacH strains had lower BC MICs than wildtype strains. In conclusion, our results provide evidence that Tn6188 is responsible for BC tolerance in various L. monocytogenes strains.This work was supported by a grant from the Austrian Science Fund (FWF, http://www.fwf.ac.at/) to SSE (grant no. Incommensurate magnetic ordering in Cu2Te2O5X2 (X=Cl, Br) studied by single crystal neutron diffraction
Polarized and unpolarized neutron diffraction studies have been carried out
on single crystals of the coupled spin tetrahedra systems Cu2Te2O5X2 (X=Cl,
Br). A model of the magnetic structure associated with the propagation vectors
k'Cl ~ -0.150,0.422,1/2 and k'Br ~ -0.172,0.356,1/2 and stable below TN=18 K
for X=Cl and TN=11 K for X=Br is proposed. A feature of the model, common to
both the bromide and chloride, is a canted coplanar motif for the 4 Cu2+ spins
on each tetrahedron which rotates on a helix from cell to cell following the
propagation vector. The Cu2+magnetic moment determined for X=Br, 0.395(5)muB,
is significantly less than for X=Cl, 0.88(1)muB at 2K. The magnetic structure
of the chloride associated with the wave-vector k' differs from that determined
previously for the wave vector k~0.150,0.422,1/2 [O. Zaharko et.al. Phys. Rev.
Lett. 93, 217206 (2004)]
Fv antibodies to aflatoxin B1 derived from a pre-immunized antibody phage display library system
The production and characterization of recombinant antibodies to aflatoxin B[SUB1] (AFB[SUB1]), a potent mycotoxin and carcinogen is described. The antibody fragments produced were then applied for use in a surface plasmon resonance-based biosensor (BIAcore), which measures biomolecular interactions in 'real-time'. Single chain Fv (scFv) antibodies were generated to aflatoxin B1 from an established phage display system, which incorporated a range of different plasmids for efficient scFv expression. The scFv's were used in the development of a competitive ELISA, and also for the development of surface plasmon resonance (SPR)-based inhibition immunoassays. They were found to be suitable for the detection of AFB[SUB1], in this format, with the assays being sensitive and reproducible

Laser Microdissection of the Alveolar Duct Enables Single-Cell Genomic Analysis
Complex tissues such as the lung are composed of structural hierarchies such as alveoli, alveolar ducts, and lobules. Some structural units, such as the alveolar duct, appear to participate in tissue repair as well as the development of bronchioalveolar carcinoma. Here, we demonstrate an approach to conduct laser microdissection of the lung alveolar duct for single-cell PCR analysis. Our approach involved three steps. (1) The initial preparation used mechanical sectioning of the lung tissue with sufficient thickness to encompass the structure of interest. In the case of the alveolar duct, the precision-cut lung slices were 200 μm thick; the slices were processed using near-physiologic conditions to preserve the state of viable cells. (2) The lung slices were examined by transmission light microscopy to target the alveolar duct. The air-filled lung was sufficiently accessible by light microscopy that counterstains or fluorescent labels were unnecessary to identify the alveolar duct. (3) The enzymatic and microfluidic isolation of single cells allowed for the harvest of as few as several thousand cells for PCR analysis. Microfluidics based arrays were used to measure the expression of selected marker genes in individual cells to characterize different cell populations. Preliminary work suggests the unique value of this approach to understand the intra- and intercellular interactions within the regenerating alveolar duct
Measurement of the Eta Production in Proton Proton Collisions with the COSY Time of Flight Spectrometer
The reaction pp -> pp eta was measured at excess energies of 15 and 41 MeV at
an external target of the Juelich Cooler Synchrotron COSY with the Time of
Flight Spectrometer. About 25000 events were measured for the excess energy of
15 MeV and about 8000 for 41 MeV. Both protons of the process pp eta were
detected with an acceptance of nearly 100% and the eta was reconstructed by the
missing mass technique. For both excess energies the angular distributions are
found to be nearly isotropic. In the invariant mass distributions strong
deviations from the pure phase space distributions are seen.Comment: 15 pages, 14 figures, 4 table
ALMA reveals the aftermath of a white dwarf–brown dwarf merger in CK Vulpeculae
We present Atacama Large Millimeter–Submillimeter Array (ALMA) observations of CK Vulpeculae which is identified with “Nova Vulpeculae 1670”. They trace obscuring dust in the inner regions of the associated nebulosity. The dust forms two cocoons, each extending ∼5″ north and south of the presumed location of the central star. Brighter emission is in a more compact east–west structure (2″ × 1″) where the cocoons intersect. We detect line emission in NH2CHO, CN, four organic molecules and C17O. CN lines trace bubbles within the dusty cocoons; CH3OH a north–south S–shaped jet; and other molecules a central cloud with a structure aligned with the innermost dust structure. The major axis of the overall dust and gas bubble structure has a projected inclination of ∼24° with respect to a 71″ extended “hourglass” nebulosity, previously seen in Hα. Three cocoon limbs align with dark lanes in the inner regions of the same Hα images. The central 2″ × 1″ dust is resolved into a structure consistent with a warped dusty disc. The velocity structure of the jets indicates an origin at the centre of this disc and precession with an unknown period. Deceleration regions at both the northern and southern tips of the jets are roughly coincident with additional diffuse dust emission over regions approximately 2″ across. These structures are consistent with a bipolar outflow expanding into surrounding high density material. We suggest that a white dwarf and brown dwarf merged between 1670 and 1672, with the observed structures and extraordinary isotopic abundances generated as a result
Role of the mesoamygdaloid dopamine projection in emotional learning
Amygdala dopamine is crucially involved in the acquisition of Pavlovian associations, as measured via conditioned approach to the location of the unconditioned stimulus (US). However, learning begins before skeletomotor output, so this study assessed whether amygdala dopamine is also involved in earlier 'emotional' learning. A variant of the conditioned reinforcement (CR) procedure was validated where training was restricted to curtail the development of selective conditioned approach to the US location, and effects of amygdala dopamine manipulations before training or later CR testing assessed. Experiment 1a presented a light paired (CS+ group) or unpaired (CS- group) with a US. There were 1, 2 or 10 sessions, 4 trials per session. Then, the US was removed, and two novel levers presented. One lever (CR+) presented the light, and lever pressing was recorded. Experiment 1b also included a tone stimulus. Experiment 2 applied intra-amygdala R(+) 7-OH-DPAT (10 nmol/1.0 A mu l/side) before two training sessions (Experiment 2a) or a CR session (Experiment 2b). For Experiments 1a and 1b, the CS+ group preferred the CR+ lever across all sessions. Conditioned alcove approach during 1 or 2 training sessions or associated CR tests was low and nonspecific. In Experiment 2a, R(+) 7-OH-DPAT before training greatly diminished lever pressing during a subsequent CR test, preferentially on the CR+ lever. For Experiment 2b, R(+) 7-OH-DPAT infusions before the CR test also reduced lever pressing. Manipulations of amygdala dopamine impact the earliest stage of learning in which emotional reactions may be most prevalent
