The DAG1 transcription factor negatively regulates the seed-to-seedling transition in Arabidopsis acting on ABA and GA levels
BACKGROUND:
In seeds, the transition from dormancy to germination is regulated by abscisic acid (ABA) and gibberellins (GAs), and involves chromatin remodelling. Particularly, the repressive mark H3K27 trimethylation (H3K27me3) has been shown to target many master regulators of this transition. DAG1 (DOF AFFECTING GERMINATION1), is a negative regulator of seed germination in Arabidopsis, and directly represses the GA biosynthetic gene GA3ox1 (gibberellin 3-β-dioxygenase 1). We set to investigate the role of DAG1 in seed dormancy and maturation with respect to epigenetic and hormonal control.
RESULTS:
We show that DAG1 expression is controlled at the epigenetic level through the H3K27me3 mark during the seed-to-seedling transition, and that DAG1 directly represses also the ABA catabolic gene CYP707A2; consistently, the ABA level is lower while the GA level is higher in dag1 mutant seeds. Furthermore, both DAG1 expression and protein stability are controlled by GAs.
CONCLUSIONS:
Our results point to DAG1 as a key player in the control of the developmental switch between seed dormancy and germination

In 2000 zijn twee nieuwe rassenproeven witte asperge aangelegd in Meterik, Nederland (PPO) en in Geel, België (PDLT). Er zijn 10 dezelfde rassen aangeplant op beide plaatsen. De rassen zijn gezaaid bij plantenkwekerij de Kemp in Meterik in 1999. Het sortiment bestaat uit de Hollandse rassen Backlim, Gijnlim, Thielim, Grolim afkomstig van Asparagus BV te Horst en de Duitse rassen Eposs, Ramada, Ramos, Ravel , Rally (96013) en Rapsody (96071) afkomstig van SWD Saatsucht. De Nederlandse rassen worden verkocht door Nunhem en de Duitse door Royal Sluis
Infrared activity of hydrogen molecules trapped in Si
The rovibrational-translational states of a hydrogen molecule moving in a cage site in Si, when subjected to an electrical field arising from its surroundings, are investigated. The wave functions are expressed in terms of basis functions consisting of the eigenfunctions of the molecule confined to move in the cavity and rovibrational states of the free molecule. The energy levels, intensities of infrared and Raman transitions, effects of uniaxial stress, and a neighboring oxygen defect are found and compared with existing experimental data
Interleukin-6 Gene-Deficient Mice Show Impaired Defense against Pneumococcal Pneumonia
Induction of pneumonia in C57Bl/6 mice by intranasal inoculation with 106 cfu of Streptococcus pneumoniae resulted in sustained expression of interleukin (IL)-6 mRNA in lungs and increases in lung and plasma IL-6 concentrations. In IL-6-deficient (IL-6−/−) mice, pneumonia was associated with higher lung levels of the proinflammatory cytokines tumor necrosis factor-α, IL-1β, and interferon-γ and of the antiinflammatory cytokine IL-10 than in wild type (IL-6+/+) mice (all P < .05). Also, the plasma concentrations of soluble tumor necrosis factor receptors were higher in IL-6−/− mice (P < .05), while the acute-phase protein response was strongly attenuated (P < .01). Lungs harvested from IL-6−/− mice 40 h after inoculation contained more S. pneumoniae colonies (P < .05). IL-6−/− mice died significantly earlier from pneumococcal pneumonia than did IL-6+/+ mice (P < .05). Metastable liquid-liquid phase transition in a single-component system with only one crystal phase and no density anomaly
We investigate the phase behavior of a single-component system in 3
dimensions with spherically-symmetric, pairwise-additive, soft-core
interactions with an attractive well at a long distance, a repulsive soft-core
shoulder at an intermediate distance, and a hard-core repulsion at a short
distance, similar to potentials used to describe liquid systems such as
colloids, protein solutions, or liquid metals. We showed [Nature {\bf 409}, 692
(2001)] that, even with no evidences of the density anomaly, the phase diagram
has two first-order fluid-fluid phase transitions, one ending in a
gas--low-density liquid (LDL) critical point, and the other in a
gas--high-density liquid (HDL) critical point, with a LDL-HDL phase transition
at low temperatures. Here we use integral equation calculations to explore the
3-parameter space of the soft-core potential and we perform molecular dynamics
simulations in the interesting region of parameters. For the equilibrium phase
diagram we analyze the structure of the crystal phase and find that, within the
considered range of densities, the structure is independent of the density.
Then, we analyze in detail the fluid metastable phases and, by explicit
thermodynamic calculation in the supercooled phase, we show the absence of the
density anomaly. We suggest that this absence is related to the presence of

Function and central projections of gustatory receptor neurons on the antenna of the noctuid moth Spodoptera littoralis
Chemosensory information is crucial for most insects to feed and reproduce. Olfactory signals are mainly used at a distance, whereas gustatory stimuli play an important role when insects directly contact chemical substrates. In noctuid moths, although the antennae are the main olfactory organ, they also bear taste sensilla. These taste sensilla detect sugars and hence are involved in appetitive learning but could also play an important role in food evaluation by detecting salts and bitter substances. To investigate this, we measured the responses of individual taste sensilla on the antennae of Spodoptera littoralis to sugars and salts using tip recordings. We also traced the projections of their neuronal axons into the brain. In each sensillum, we found one or two neurons responding to sugars: one NaCl-responsive and one water-sensitive neuron. Responses of these neurons were dose-dependent and similar across different locations on the antenna. Responses were dependent on the sex for sucrose and on both sex and location for glucose and fructose. We did not observe a spatial map for the projections from specific regions of the antennae to the deutocerebrum or the tritocerebrum/suboesophageal ganglion complex. In accordance with physiological recordings, back-fills from individual sensilla revealed up to four axons, in most cases targeting different projection zones
Personalised service? Changing the role of the government librarian
Investigates the feasibility of personalised information service in a government department. A qualitative methodology explored stakeholder opinions on the remit, marketing, resourcing and measurement of the service. A questionnaire and interviews gathered experiences of personalised provision across the government sector. Potential users were similarly surveyed to discuss how the service could meet their needs. Data were analysed using coding techniques to identify emerging theory. Lessons learned from government librarians centred on clarifying requirements, balancing workloads and selective marketing. The user survey showed low usage and awareness of existing specialist services, but high levels of need and interest in services repackaged as a tailored offering. Fieldwork confirmed findings from the literature on the scope for adding value through information management advice, information skills training and substantive research assistance and the need to understand business processes and develop effective partnerships.
