    Selection of alfalfa genotypes for resistance to the foliar pathogen Curvularia geniculata

    ABSTRACT Foliar diseases impose severe restrictions on the persistence and productivity of Medicago sativa, both of which may be increased by developing disease resistant and more competitive genotypes that can improve pasture quality. We found Curvularia geniculata as the principal alfalfa foliar pathogen in the Brazilian state of Rio Grande do Sul (RS). Growth chamber experiments evaluated the resistance of alfalfa genotypes ‘E1C4’, ‘CPPSul’, ‘ABT 805’ and ‘CUF-101’ to C. geniculata as compared the control ‘Crioula’ genotype. These genotypes were also evaluated in field trials at a sea level site in Eldorado do Sul in central RS and at two sites £200 m above sea level, one in Bagé municipality in south west RS and the other at a farm near the town of Alto Feliz in north east RS. Plants were spray-inoculated with 1.6 x 106 ml-1 of C. geniculata spores and visually evaluated for leaf damage 14 days later. The C. geniculata infection rates varied from zero to 100%. Alfalfa persistence and forage mean dry mass (DM) production at the Eldorado site were measured during different seasons from November 2013 to January 2015 by calculating the incidence of invasive plants and morphologically separating leaves from stems and calculating both leaf and stem DM. Data were analysed using mixed statistical models. The best results for persistence and forage DM were shown by the ‘CPPSul’ genotypes (DM = 16,600 kg ha-1) and ‘Crioula’ (DM = 15,750 kg ha-1). These two genotypes will be used for subsequent investigations and selection cycles

    A number of genes that confer resistance to coffee leaf rust (S H 1–S H 9) have been identified within the genus Coffea, but despite many years of research on this pathosystem, the complementary avirulence genes of Hemileia vastatrix have not been reported. After identification of H. vastatrix effector candidate genes (HvECs) expressed at different stages of its lifecycle, we established an assay to characterize HvEC proteins by delivering them into coffee cells via the type-three secretion system (T3SS) of Pseudomonas syringae pv. garcae (Psgc). Employing a calmodulin-dependent adenylate cyclase assay, we demonstrate that Psgc recognizes a heterologous P. syringae T3SS secretion signal which enables us to translocate HvECs into the cytoplasm of coffee cells. Using this Psgc-adapted effector detector vector (EDV) system, we found that HvEC-016 suppresses the growth of Psgc on coffee genotypes with the S H 1 resistance gene. Suppression of bacterial blight symptoms in S H 1 plants was associated with reduced bacterial multiplication. By contrast, HvEC-016 enhanced bacterial multiplication in S H 1-lacking plants. Our findings suggest that HvEC-016 may be recognized by the plant immune system in a S H 1-dependent manner. Thus, our experimental approach is an effective tool for the characterization of effector/avirulence proteins of this important pathogen

    Adubação nitrogenada e potássica na severidade da antracnose em dois cultivares de milho

    O equilíbrio nutricional pode contribuir para a resistência das plantas às doenças. Com o objetivo de avaliar o efeito da interação entre as adubações nitrogenada e potássica na severidade das lesões da antracnose foliar e na nutrição mineral da cultura do milho, foram instalados dois experimentos, em casa de vegetação com dois cultivares, o DAS 2B710 (moderadamente resistente à doença) e o BRS 1010 (susceptível), cinco doses de N (75, 150, 300, 600 e 1200 mg dm-3) e cinco doses de K 63, 125, 250, 500 e 1000 mg dm-3). O delineamento experimental para cada cultivar foi em blocos inteiramente ao acaso, em esquema fatorial 5 x 5, com 25 tratamentos e quatro repetições. Cada vaso com quatro plantas constituiu uma parcela experimental. As doses de N e K foram divididas em quatro parcelas, com intervalos de dez dias a partir da semeadura, para os dois cultivares. Aos 21 dias após a semeadura, as plantas foram inoculadas e levadas para câmara úmida, com fotoperíodo de 16 horas, no escuro durante três dias consecutivos. Avaliou-se a severidade das lesões diariamente, até 11 dias após a inoculação. Aos 43 dias após a semeadura, a parte aérea das plantas foi colhida, secada e moída para determinar os teores de N e de K. A quantidade de área foliar lesionada dependeu da interação entre os nutrientes. Em ambos os cultivares, os menores valores de severidade foram observados na menor dose de N, combinada com a maior dose de K. A severidade das lesões observada no cultivar moderadamente resistente foi 41% menor que a observada no cultivar susceptível.     Circulating CD36 Is Reduced in HNF1A-MODY Carriers

    INTRODUCTION: Premature atherosclerosis is a significant cause of morbidity and mortality in type 2 diabetes mellitus. Maturity onset diabetes of the young (MODY) accounts for approximately 2% of all diabetes, with mutations in the transcription factor; hepatocyte nuclear factor 1 alpha (HNF1A) accounting for the majority of MODY cases. There is somewhat limited data available on the prevalence of macrovascular disease in HNF1A-MODY carriers with diabetes. Marked insulin resistance and the associated dyslipidaemia are not clinical features of HNF1A-MODY carriers. The scavenger protein CD36 has been shown to play a substantial role in the pathogenesis of atherosclerosis, largely through its interaction with oxidised LDL. Higher levels of monocyte CD36 and plasma CD36(sCD36) are seen to cluster with insulin resistance and diabetes. The aim of this study was to determine levels of sCD36 in participants with HNF1A-MODY diabetes and to compare them with unaffected normoglycaemic family members and participants with type 2 diabetes mellitus. METHODS: We recruited 37 participants with HNF1A-MODY diabetes and compared levels of sCD36 with BMI-matched participants with type 2 diabetes mellitus and normoglycaemic HNF1A-MODY negative family controls. Levels of sCD36 were correlated with phenotypic and biochemical parameters. RESULTS: HNF1A-MODY participants were lean, normotensive, with higher HDL and lower triglyceride levels when compared to controls and participants with type 2 diabetes mellitus. sCD36 was also significantly lower in HNF1A-MODY participants when compared to both the normoglycaemic family controls and to lean participants with type 2 diabetes mellitus. CONCLUSION: In conclusion, sCD36 is significantly lower in lean participants with HNF1A-MODY diabetes when compared to weight-matched normoglycaemic familial HNF1A-MODY negative controls and to lean participants with type 2 diabetes mellitus. Lower levels of this pro-atherogenic marker may result from the higher HDL component in the lipid profile of HNF1A-MODY participants
