NASA Team Collaboration Pilot: Enabling NASA's Virtual Teams
Most NASA projects and work activities are accomplished by teams of people. These teams are often geographically distributed - across NASA centers and NASA external partners, both domestic and international. NASA "virtual" teams are stressed by the challenge of getting team work done - across geographic boundaries and time zones. To get distributed work done, teams rely on established methods - travel, telephones, Video Teleconferencing (NASA VITS), and email. Time is our most critical resource - and team members are hindered by the overhead of travel and the difficulties of coordinating work across their virtual teams. Modern, Internet based team collaboration tools offer the potential to dramatically improve the ability of virtual teams to get distributed work done
Results of an Advanced Fan Stage Operating Over a Wide Range of Speed and Bypass Ratio
NASA s Fundamental Aeronautics Program is investigating turbine-based combined cycle (TBCC) propulsion systems for access to space because it provides the potential for aircraft-like, space-launch operations that may significantly reduce launch costs and improve safety. To this end, National Aeronautics and Space Administration (NASA) and General Electric (GE) teamed to design a Mach 4 variable cycle turbofan/ramjet engine for access to space. To enable the wide operating range of a Mach 4+ variable cycle turbofan ramjet required the development of a unique fan stage design capable of multi-point operation to accommodate variations in bypass ratio (10 ), fan speed (7 ), inlet mass flow (3.5 ), inlet pressure (8 ), and inlet temperature (3 ). In this paper, NASA has set out to characterize a TBCC engine fan stage aerodynamic performance and stability limits over a wide operating range including power-on and hypersonic-unique "windmill" operation. Herein, we will present the fan stage design, and the experimental test results of the fan stage operating from 15 to 100 percent corrected design speed. Whereas, in the companion paper, we will provide an assessment of NASA s APNASA code s ability to predict the fan stage performance and operability over a wide range of speed and bypass ratio
Neuropilin-1 mediates vascular permeability independently of vascular endothelial growth factor receptor-2 activation
Neuropilin-1 (NRP1) regulates developmental and pathological angiogenesis, arteriogenesis, and vascular permeability, acting as a coreceptor for semaphorin 3A (Sema3A) and the 165–amino acid isoform of vascular endothelial growth factor A (VEGF-A165). NRP1 is also the receptor for the CendR peptides, a class of cell- and tissue-penetrating peptides with a specific R-x-x-R carboxyl-terminal motif. Because the cytoplasmic domain of NRP1 lacks catalytic activity, NRP1 is mainly thought to act through the recruitment and binding to other receptors. We report here that the NRP1 intracellular domain mediates vascular permeability. Stimulation with VEGF-A165, a ligand-blocking antibody, and a CendR peptide led to NRP1 accumulation at cell-cell contacts in endothelial cell monolayers, increased cellular permeability in vitro and vascular leakage in vivo. Biochemical analyses, VEGF receptor-2 (VEGFR-2) silencing, and the use of a specific VEGFR blocker established that the effects induced by the CendR peptide and the antibody were independent of VEGFR-2. Moreover, leakage assays in mice expressing a mutant NRP1 lacking the cytoplasmic domain revealed that this domain was required for NRP1-induced vascular permeability in vivo. Hence, these data define a vascular permeability pathway mediated by NRP1 but independent of VEGFR-2 activation
Inlet Flow Test Calibration for a Small Axial Compressor Facility. Part 1: Design and Experimental Results
An axial compressor test rig has been designed for the operation of small turbomachines. The inlet region consisted of a long flowpath region with two series of support struts and a flapped inlet guide vane. A flow test was run to calibrate and determine the source and magnitudes of the loss mechanisms in the inlet for a highly loaded two-stage axial compressor test. Several flow conditions and IGV angle settings were established in which detailed surveys were completed. Boundary layer bleed was also provided along the casing of the inlet behind the support struts and ahead of the IGV. A detailed discussion of the flowpath design along with a summary of the experimental results are provided in Part 1

Several computational fluid dynamics (CFD) calculations were made for selected flow conditions established during the test. Good agreement between the CFD and test data were obtained for these test conditions
Aerodynamic Synthesis of a Centrifugal Impeller Using CFD and Measurements
The performance and flow structure in an unshrouded impeller of approximately 4:1 pressure ratio is synthesized on the basis of a detailed analysis of 3D viscous CFD results and aerodynamic measurements. A good data match was obtained between CFD and measurements using laser anemometry and pneumatic probes. This solidified the role of the CFD model as a reliable representation of the impeller internal flow structure and integrated performance. Results are presented showing the loss production and secondary flow structure in the impeller. The results indicate that while the overall impeller efficiency is high, the impeller shroud static pressure recovery potential is underdeveloped leading to a performance degradation in the downstream diffusing element. Thus, a case is made for a follow-on impeller parametric design study to improve the flow quality. A strategy for aerodynamic performance enhancement is outlined and an estimate of the gain in overall impeller efficiency that might be realized through improvements to the relative diffusion process is provided
VEGF binding to NRP1 is essential for VEGF stimulation of endothelial cell migration, complex formation between NRP1 and VEGFR2, and signaling via FAK Tyr407 phosphorylation
In endothelial cells, neuropilin-1 (NRP1) binds vascular endothelial growth factor (VEGF)-A and is thought to act as a coreceptor for kinase insert domain-containing receptor (KDR) by associating with KDR and enhancing VEGF signaling. Here we report mutations in the NRP1 b1 domain (Y297A and D320A), which result in complete loss of VEGF binding. Overexpression of Y297A and D320A NRP1 in human umbilical vein endothelial cells reduced high-affinity VEGF binding and migration toward a VEGF gradient, and markedly inhibited VEGF-induced angiogenesis in a coculture cell model. The Y297A NRP1 mutant also disrupted complexation between NRP1 and KDR and decreased VEGF-dependent phosphorylation of focal adhesion kinase at Tyr407, but had little effect on other signaling pathways. Y297A NRP1, however, heterodimerized with wild-type NRP1 and NRP2 indicating that nonbinding NRP1 mutants can act in a dominant-negative manner through formation of NRP1 dimers with reduced binding affinity for VEGF. These findings indicate that VEGF binding to NRP1 has specific effects on endothelial cell signaling and is important for endothelial cell migration and angiogenesis mediated via complex formation between NRP1 and KDR and increased signaling to focal adhesions. Identification of key residues essential for VEGF binding and biological functions provides the basis for a rational design of antagonists of VEGF binding to NRP1
Laser Anemometer Measurements of the Flow Field in a 4:1 Pressure Ratio Centrifugal Impeller
A laser-doppler anemometer was used to obtain flow-field velocity measurements in a 4:1 pressure ratio, 4.54 kg/s (10 lbm/s), centrifugal impeller, with splitter blades and backsweep, which was configured with a vaneless diffuser. Measured through-flow velocities are reported for ten quasi-orthogonal survey planes at locations ranging from 1% to 99% of main blade chord. Measured through-flow velocities are compared to those predicted by a 3-D viscous steady flow analysis (Dawes) code. The measurements show the development and progression through the impeller and vaneless diffuser of a through-flow velocity deficit which results from the tip clearance flow and accumulation of low momentum fluid centrifuged from the blade and hub surfaces. Flow traces from the CFD analysis show the origin of this deficit which begins to grow in the inlet region of the impeller where it is first detected near the suction surface side of the passage. It then moves toward the pressure side of the channel, due to the movement of tip clearance flow across the impeller passage, where it is cut by the splitter blade leading edge. As blade loading increases toward the rear of the channel the deficit region is driven back toward the suction surface by the cross-passage pressure gradient. There is no evidence of a large wake region that might result from flow separation and the impeller efficiency is relatively high. The flow field in this impeller is quite similar to that documented previously by NASA Lewis in a large low-speed backswept impeller
Experimental Results of the First Two Stages of an Advanced Transonic Core Compressor Under Isolated and Multi-Stage Conditions
NASA's Environmentally Responsible Aviation (ERA) Program calls for investigation of the technology barriers associated with improved fuel efficiency of large gas turbine engines. Under ERA the task for a High Pressure Ratio Core Technology program calls for a higher overall pressure ratio of 60 to 70. This mean that the HPC would have to almost double in pressure ratio and keep its high level of efficiency. The challenge is how to match the corrected mass flow rate of the front two supersonic high reaction and high corrected tip speed stages with a total pressure ratio of 3.5. NASA and GE teamed to address this challenge by using the initial geometry of an advanced GE compressor design to meet the requirements of the first 2 stages of the very high pressure ratio core compressor. The rig was configured to run as a 2 stage machine, with Strut and IGV, Rotor 1 and Stator 1 run as independent tests which were then followed by adding the second stage. The goal is to fully understand the stage performances under isolated and multi-stage conditions and fully understand any differences and provide a detailed aerodynamic data set for CFD validation. Full use was made of steady and unsteady measurement methods to isolate fluid dynamics loss source mechanisms due to interaction and endwalls. The paper will present the description of the compressor test article, its predicted performance and operability, and the experimental results for both the single stage and two stage configurations. We focus the detailed measurements on 97 and 100 of design speed at 3 vane setting angles
Blocking endothelial apoptosis revascularises the retina in a model of ischemic retinopathy
Aberrant, neovascular retinal blood vessel growth is a vision-threatening complication in ischemic retinal diseases. It is driven by retinal hypoxia frequently caused by capillary nonperfusion and endothelial cell (EC) loss. We investigated the role of EC apoptosis in this process using a mouse model of ischemic retinopathy, in which vessel closure and EC apoptosis cause capillary regression and retinal ischemia followed by neovascularization. Protecting ECs from apoptosis in this model did not prevent capillary closure or retinal ischemia. Nonetheless, it prevented the clearance of ECs from closed capillaries, delaying vessel regression and allowing ECs to persist in clusters throughout the ischemic zone. In response to hypoxia, these preserved ECs underwent a vessel sprouting response and rapidly reassembled into a functional vascular network. This alleviated retinal hypoxia, preventing subsequent pathogenic neovascularization. Vessel reassembly was not limited by VEGFA neutralization, suggesting it was not dependent on the excess VEGFA produced by the ischemic retina. Neutralization of ANG2 did not prevent vessel reassembly, but did impair subsequent angiogenic expansion of the reassembled vessels. Blockade of EC apoptosis may promote ischemic tissue revascularization by preserving ECs within ischemic tissue that retain the capacity to reassemble a functional network and rapidly restore blood supply
