Dispersal Dynamics of the Bivalve Gemma Gemma in a Patchy Environment
The purpose of this study was to analyze the dispersal dynamics of the ovoviviparous bivalve Gemma gemma (hereafter referred to as Gemma) in an environment disturbed by the pit-digging activities of horseshoe crabs, Limulus polyphemus. Gemma broods its young and has no planktonic larval stage, so all dispersal is the result of juvenile and adult movement. Animal movement was measured using natural crab pits, hand-dug simulated crab pits, and cylindrical bottom traps in the intertidal zone at Tom\u27s Cove, Virginia, USA.
This study demonstrated that horseshoe crabs create localized patches with reduced densities of Gemma, that all sizes and ages of Gemma quickly disperse into these low density patches, and that the mechanism of dispersal is passive bedload and suspended load transport. Freshly excavated natural pits had significantly lower Gemma densities than did undisturbed background sediment, but there were no significant differences in total density of other species, number of species, and species diversity (H\u27). Equitability (J\u27) was greater in pits than in controls because of the reduced abundance of Gemma, the numerically dominant species. Newly dug simulated crab pits also had significantly lower Gemma densities than controls and returned to control levels by the next day. Density recovery trajectories for individually marked pits showed consistent responses in summer and fall, but not in winter when low Gemma abundance resulted in greater variability among pits.
Significant positive correlations between the volume of sediment and the number of Gemma collected per bottom trap support the hypothesis that Gemma dispersal is a passive transport phenomenon. Assuming no active, density-dependent movement, the product of the Gemma density frequency distribution in undisturbed background sediment and the frequency distribution of sediment volume collected per trap created a predicted Gemma frequency distribution in traps that matched the actual distribution. Absolute dispersal rates and relative dispersal rates (absolute dispersal rate divided by background density in undisturbed sediment) into pits and traps were greater in summer than winter. Dispersal rate results suggest that increased horseshoe crab disturbance in summer may cause an increase in Gemma transport. Because Gemma individuals are dispersed by hydrodynamic action, it was expected that small, young individuals would be most easily transported in the bedload. There was, however, little evidence that movement into pits and traps was size- or age-selective.
Most recent benthic dispersal research has focused on the large-scale movement and settlement patterns of invertebrate larvae. The results from this study illustrate that dispersal of bottom-dwelling juveniles and adults plays an important role in regulating the local distribution and abundance of Gemma. Previous workers have shown that young Gemma live in dense aggregations and that growth and fecundity are reduced at such high densities, leading to population crashes. This study demonstrated a mechanism by which Gemma disperses into low-density patches where intraspecific competition may be mitigated, possibly resulting in enhanced individual reproductive success and population fitness
The Impact of Sexual or Physical Abuse History on Pain‐Related Outcomes Among Blacks and Whites with Chronic Pain: Gender Influence
Objectives. Physical and sexual abuses commonly co‐occur with chronic pain. We hypothesized that: 1) abuse history questions would form distinct factors that relate differently to pain perceptions and pain outcomes; 2) abuse history consequences on physical and mental health differ by gender; and 3) different abuse types and age of occurrence (childhood vs adolescent/adulthood) predict different negative outcomes. Methods. Chronic pain patients at a tertiary care pain center provided data (64% women, 50% black) through a confidential survey. Factors were formed for abuse type and age. Linear regression, controlling for socio‐demographic information, was used to examine the relationship between abuse and abuse by sex interactions with pain‐related outcomes. Results. Six 3‐item abuse factors (α = 0.77–0.91)—sexual molestation, sexual penetration, and physical abuse—were identified in both childhood and adulthood. Lifetime prevalence of abuse was 70% for men and 65% for women. Women experienced lower physical abuse ( P = 0.01) in childhood, and higher penetration ( P = 0.02) in adulthood. Decreased general health was associated with all abuse types ( P < 0.05) in childhood. Affective pain was associated with all childhood abuse scales and adulthood molestation, though childhood molestation only for men ( P = 0.04). Disability was associated with childhood ( P = 0.02) and adulthood rape ( P = 0.04). Men with childhood or adulthood molestation ( P = 0.02; P = 0.02) reported higher post‐traumatic stress disorder. Conclusions. Our study confirms physical and mental health, and pain‐related outcomes are affected by abuse history for men and women. A Cure for HIV Infection: "Not in My Lifetime" or "Just Around the Corner"?
With the advent and stunning success of combination antiretroviral therapy (ART) to prolong and improve quality of life for persons with HIV infection, HIV research has been afforded the opportunity to pivot towards studies aimed at finding "a cure." The mere idea that cure of HIV might be possible has energized researchers and the community towards achieving this goal. Funding agencies, both governmental and private, have targeted HIV cure as a high priority; many in the field have responded to these initiatives and the cure research agenda is robust. In this "salon" two editors of Pathogens and Immunity, Michael Lederman and Daniel Douek ask whether curing HIV is a realistic, scalable objective. We start with an overview perspective and have asked a number of prominent HIV researchers to add to the discussion
The effects of Hordeum spontaneum C. Koch. germplasm on barley production
Several studies have shown that Hordeum spontaneum contains potentially useful genes for improving agronomic traits of cultivated barley. In this study, the value of seven accessions of H. spontaneum C. Koch as donors of both cytoplasmic and nuclear genes affecting agronomic traits of barley were investigated;Favorable effects of H. spontaneum cytoplasm were found for straw yield, unit straw weight, and vegetative growth index. These favorable effects were actually attributed to nuclear-cytoplasmic interactions as indicated by the presence of cytoplasm-mating interactions and absence of cytoplasmic-generation interactions. Thus, H. spontaneum cytoplasmic genome must have interacted with H. vulgare nuclear genome. H. vulgare cytoplasmic effects were found to be superior to H. spontaneum for grain yield and harvest index. These cytoplasmic effects were attributed to nuclear-cytoplasmic interactions as indicated by the presence of cytoplasm-generation interactions. Therefore, H. vulgare cytoplasm must have interacted with either (a) H. spontaneum nuclear genes alone, or (b) specific combinations of H. spontaneum and H. vulgare nuclear genes;The seven accessions used in this study were assigned to four ecotypes and tested for variability among and within ecotypes by evaluating ecotype segregates. Superior performance for specific traits was, at least in part, a result of specific ecotypes. The most obvious effect was the superior straw yield performance from coastal ecotype segregates; however, these ecotype segregates also had the poorest harvest index. Segregates from matings involving northern and southern ecotypes had the highest harvest index. Based on the small sample size, variability among and within ecotype segregates suggests the possibility of obtaining superior progenies by sampling a larger number of accessions among and within ecotype categories. Topological Defects and Interactions in Nematic Emulsions
Inverse nematic emulsions in which surfactant-coated water droplets are
dispersed in a nematic host fluid have distinctive properties that set them
apart from dispersions of two isotropic fluids or of nematic droplets in an
isotropic fluid. We present a comprehensive theoretical study of the
distortions produced in the nematic host by the dispersed droplets and of
solvent mediated dipolar interactions between droplets that lead to their
experimentally observed chaining. A single droplet in a nematic host acts like
a macroscopic hedgehog defect. Global boundary conditions force the nucleation
of compensating topological defects in the nematic host. Using variational
techniques, we show that in the lowest energy configuration, a single water
droplet draws a single hedgehog out of the nematic host to form a tightly bound
dipole. Configurations in which the water droplet is encircled by a
disclination ring have higher energy. The droplet-dipole induces distortions in
the nematic host that lead to an effective dipole-dipole interaction between

HIV-1 protease inhibitors and clinical malaria: A secondary analysis of the AIDS Clinical Trials Group A5208 study
HIV-1 protease inhibitors (PIs) have antimalarial activity in vitro and in murine models. The potential beneficial effect of HIV-1 PIs on malaria has not been studied in clinical settings. We used data from Adult AIDS Clinical Trials Group A5208 sites where malaria is endemic to compare the incidence of clinically diagnosed malaria among HIV-infected adult women randomized to either lopinavir/ritonavir (LPV/r)-based antiretroviral therapy (ART) or to nevirapine (NVP)-based ART. We calculated hazard ratios and 95% confidence intervals. We conducted a recurrent events analysis that included both first and second clinical malarial episodes and also conducted analyses to assess the sensitivity of results to outcome misclassification. Among the 445 women in this analysis, 137 (31%) received a clinical diagnosis of malaria at least once during follow-up. Of these 137, 72 (53%) were randomized to LPV/r-based ART. Assignment to the LPV/r treatment group (n = 226) was not consistent with a large decrease in the hazard of first clinical malarial episode (hazard ratio = 1.11 [0.79 to 1.56]). The results were similar in the recurrent events analysis. Sensitivity analyses indicated the results were robust to reasonable levels of outcome misclassification. In this study, the treatment with LPV/r compared to NVP had no apparent beneficial effect on the incidence of clinical malaria among HIV-infected adult women. Additional research concerning the effects of PI-based therapy on the incidence of malaria diagnosed by more specific criteria and among groups at a higher risk for severe disease is warranted. Comparative gender analysis of the efficacy and safety of atazanavir/ritonavir and lopinavir/ritonavir at 96 weeks in the CASTLE study.
OBJECTIVES: To examine whether the overall results of the CASTLE study pertain to both genders, we analysed the efficacy and safety of atazanavir/ritonavir and lopinavir/ritonavir in 277 female and 606 male patients in the open-label, multinational trial over 96 weeks. The trial is registered with ClinicalTrials.gov, number NCT00272779.
METHODS: Treatment-naive patients aged ≥ 18 years with HIV-1 RNA ≥ 5000 copies/mL were randomized to receive either atazanavir/ritonavir 300/100 mg once daily or lopinavir/ritonavir 400/100 mg twice daily, with fixed-dose tenofovir/emtricitabine 300/200 mg once daily.
RESULTS: At week 96, confirmed virological response rates (HIV RNA \u3c50 copies\u3e/mL; intent-to-treat analysis) were higher in women and men receiving atazanavir/ritonavir than those receiving lopinavir/ritonavir and lower in women than men in both treatment arms (67% of women and 77% of men on atazanavir/ritonavir and 63% of women and 71% of men on lopinavir/ritonavir). These differences were not observed in the on-treatment analysis. Mean change in CD4 cell count from baseline to week 96 was 265 cells/mm(3) for women and 269 cells/mm(3) for men on atazanavir/ritonavir and 298 cells/mm(3) for women and 286 cells/mm(3) for men on lopinavir/ritonavir. Discontinuation rates were higher in women than men in each treatment arm (22% of women and 15% of men on atazanavir/ritonavir and 29% of women and 18% of men on lopinavir/ritonavir). In women and men, grade 2-4 nausea and diarrhoea were more frequent in the lopinavir/ritonavir group; jaundice and hyperbilirubinaemia occurred more frequently in the atazanavir/ritonavir group.
CONCLUSIONS: Once-daily atazanavir/ritonavir is an effective and well-tolerated therapeutic option for women and men with HIV-1 infection. The sex-based differences in response may be due to higher discontinuation rates in women
An essential thioredoxin-type protein of Trypanosoma brucei acts as redox-regulated mitochondrial chaperone
Most known thioredoxin-type proteins (Trx) participate in redox pathways, using two highly conserved cysteine residues to catalyze thiol-disulfide exchange reactions. Here we demonstrate that the so far unexplored Trx2 from African trypanosomes (Trypanosoma brucei) lacks protein disulfide reductase activity but functions as an effective temperature-activated and redox-regulated chaperone. Immunofluorescence microscopy and fractionated cell lysis revealed that Trx2 is located in the mitochondrion of the parasite. RNA-interference and gene knock-out approaches showed that depletion of Trx2 impairs growth of both mammalian bloodstream and insect stage procyclic parasites. Procyclic cells lacking Trx2 stop proliferation under standard culture conditions at 27°C and are unable to survive prolonged exposure to 37°C, indicating that Trx2 plays a vital role that becomes augmented under heat stress. Moreover, we found that Trx2 contributes to the in vivo infectivity of T. brucei. Remarkably, a Trx2 version, in which all five cysteines were replaced by serine residues, complements for the wildtype protein in conditional knock-out cells and confers parasite infectivity in the mouse model. Characterization of the recombinant protein revealed that Trx2 can coordinate an iron sulfur cluster and is highly sensitive towards spontaneous oxidation. Moreover, we discovered that both wildtype and mutant Trx2 protect other proteins against thermal aggregation and preserve their ability to refold upon return to non-stress conditions. Activation of the chaperone function of Trx2 appears to be triggered by temperature-mediated structural changes and inhibited by oxidative disulfide bond formation. Our studies indicate that Trx2 acts as a novel chaperone in the unique single mitochondrion of T. brucei and reveal a new perspective regarding the physiological function of thioredoxin-type proteins in trypanosomes
