Rejection Sensitivity, Perceived Power, and HIV Risk in the Relationships of Low-Income Urban Women
The psychological processes associated with HIV infection in long-term relationships differ from those operative in casual sexual encounters, and relatively little research has considered the aspects of personality applicable in the ongoing heterosexual relationships in which women are at greatest risk. Sensitivity to rejection has been linked with efforts to prevent rejection at a cost to the self and, therefore, may be relevant to the health risks that many women incur in relationships. We examined the association of rejection sensitivity with women\u27s sexual risk behavior in a sample of women at heightened risk for HIV exposure. Women in long-term heterosexual relationships (N = 159) were recruited for study participation in the hospital emergency room serving a low-income neighborhood in New York City, in 2001-2003. Rejection sensitivity and known HIV risk factors were assessed using verbally administered questionnaires. Rejection sensitivity was associated with lower perceived relationship power and, in turn, more frequent unprotected sex with a partner perceived to be at risk for HIV. These results held when controlling for other HIV risk factors including partner violence, economic dependence, and substance use. Understanding the association of rejection concerns with lower perceived personal power in relationships may be important for HIV prevention
Pengaruh Kecerdasan Emosional terhadap Perilaku Konsumsi Siswa di SMA
This thesis aims to find out how the influence of emotional intelligence on the consumer behavior of students in SMA Hidayatul Muhsinin Kubu Raya Regency Academic Year 2013/2014. The methos used is associative method with the form of causal relationship research. With a population of 111 students and a total sample of 84 students by using proportional random sampling technique. The data collection technique used are direct communication technique, indirect communication technique and document technique with the data collecting tools such as interview guideline, questionnaire and document. Based on the data analysis, there is the influence of emotional intelligence on the consumer behavior, the rate of the influence is 31,5 %. The emotional intelligence level of students in SMA Hidayatul Muhsinin Kubu Raya Regency in good category with a percentage of 65,5 % whereas the rate of consumer behavior of students in SMA Hidayatul Muhsinin Kubu Raya Regency in good category with a percentage of 58,3 %
Adverse prognostic and predictive significance of low DNA-dependent protein kinase catalytic subunit (DNA-PKcs) expression in early-stage breast cancers
Background: DNA-dependent protein kinase catalytic subunit (DNA-PKcs), a serine threonine kinase belonging to the PIKK family (phosphoinositide 3-kinase-like-family of protein kinase), is a critical component of the non-homologous end joining (NHEJ) pathway required for the repair of DNA double strand breaks. DNA-PKcs may be involved in breast cancer pathogenesis. Methods: We evaluated clinicopathological significance of DNA-PKcs protein expression in 1161 tumours and DNA-PKcs mRNA expression in 1950 tumours. We correlated DNA-PKcs to other markers of aggressive phenotypes, DNA repair, apoptosis and cell cycle regulation. Results: Low DNA-PKcs protein expression was associated with higher tumour grade, higher mitotic index, tumour de-differentiation and tumour type (ps<0.05). Absence of BRCA1, low XRCC1/SMUG1/APE1/Polβ were also more likely in low DNA-PKcs expressing tumours (ps<0.05). Low DNA-PKcs protein expression was significantly associated with worse breast cancer specific survival (BCCS) in univariate and multivariate analysis (ps<0.01). At the mRNA level, low DNA-PKcs was associated with PAM50.Her2 and PAM50.LumA molecular phenotypes (ps<0.01) and poor BCSS. In patients with ER positive tumours who received endocrine therapy, low DNA-PKcs (protein and mRNA) was associated with poor survival. In ER negative patients, low DNA-PKcs mRNA remains significantly associated with adverse outcome. Conclusions: Our study suggests that low DNA-PKcs expression may have prognostic and predictive significance in breast cancers

QUANTITATIVE ANALYSIS OF BIOACTIVE COMPOUNDS FROM TAGETES ERECTA (LINN.)
Objectives: This study investigates the quantitative analysis of the major phytoconstituents of Tagetes erecta (Linn.) and proves their therapeuticvalue as medicines in both leaf and flower extracts.Methods: The various bioactive compounds were quantitatively analyzed as per standard methods.Results: The present study registered carbohydrates, coumarine, alkaloids, and quinone which are of medicinal importance. During the study, quinonerecorded the highest amount of phytocomponents in both the leaf and flower samples.Conclusion: The significant result of the present study revealed that the flower extract exhibited more amounts of phytochemicals than the others,and this could be exploded for potential drug development in future.Keywords: Phytoconstituents, Coumarine, Alkaloids, Quinone, Extraction, Solvents
Clinicopathological and functional significance of RECQL1 helicase in sporadic breast cancers
RECQL1, a key member of the RecQ family of DNA helicases, is required for DNA replication and DNA repair. Two recent studies have shown that germline RECQL1 mutations are associated with increased breast cancer susceptibility. Whether altered RECQL1 expression has clinicopathologic significance in sporadic breast cancers is unknown. We evaluated RECQL1 at the transcriptomic level (METABRIC cohort, n = 1,977) and at the protein level [cohort 1, n = 897; cohort 2, n = 252; cohort 3 (BRCA germline deficient), n = 74]. In RECQL1-depleted breast cancer cells, we investigated anthracycline sensitivity. High RECQL1 mRNA was associated with intClust.3 (P = 0.026), which is characterized by low genomic instability. On the other hand, low RECQL1 mRNA was linked to intClust.8 [luminal A estrogen receptor–positive (ER+) subgroup; P = 0.0455] and intClust.9 (luminal B ER+ subgroup; P = 0.0346) molecular phenotypes. Low RECQL1 expression was associated with shorter breast cancer–specific survival (P = 0.001). At the protein level, low nuclear RECQL1 level was associated with larger tumor size, lymph node positivity, high tumor grade, high mitotic index, pleomorphism, dedifferentiation, ER negativity, and HER-2 overexpression (P < 0.05). In ER+ tumors that received endocrine therapy, low RECQL1 was associated with poor survival (P = 0.008). However, in ER− tumors that received anthracycline-based chemotherapy, high RECQL1 was associated with poor survival (P = 0.048). In RECQL1-depleted breast cancer cell lines, we confirmed doxorubicin sensitivity, which was associated with DNA double-strand breaks accumulation, S-phase cell-cycle arrest, and apoptosis. We conclude that RECQL1 has prognostic and predictive significance in breast cancers

Molecular characterization and phylogenetic analysis of betasatellite molecules associated with okra yellow vein mosaic disease in Sri Lanka
Okra production in Sri Lanka has been severely affected by okra yellow vein mosaic disease (OYVMD), which is caused by
begomoviruses and associated betasatellites. These betasatellite molecules commonly determine the development and severity of the disease. Therefore, knowledge about the genetic variability of betasatellites associated with OYVMD could assist okra breeding programs in the selection of resistant varieties. The present study aimed to characterize the betasatellite DNA sequences associated with OYVMD in Sri Lanka and to determine their phylogenetic relationships. Betasatellite DNA of six virus isolates from widely separated geographical locations were sequenced and compared with already reported begomovirus betasatellites. The betasatellite molecules have features common to other betasatellite DNAs: a conserved nonanucleotide TAATATTAC, a coding sequence for the protein βC1, an adenine rich region and a satellite conserved region. Nucleotide diversity among the isolates was relatively low (π = 0.034). A recombination event was detected at a specific region in the genome of all isolates. The isolates shared >96% sequence identity with bhendi yellow vein betasatellites reported from India and phylogenetic analysis confirmed their genetic relationship
RECQL4 helicase has oncogenic potential in sporadic breast cancers
RECQL4 helicase is a molecular motor that unwinds DNA, a process essential during DNA replication and DNA repair. Germ-line mutations in RECQL4 cause type II Rothmund–Thomson syndrome (RTS), characterized by a premature ageing phenotype and cancer predisposition. RECQL4 is widely considered to be a tumour suppressor, although its role in human breast cancer is largely unknown. As the RECQL4 gene is localized to chromosome 8q24, a site frequently amplified in sporadic breast cancers, we hypothesized that it may play an oncogenic role in breast tumourigenesis. To address this, we analysed large cohorts for gene copy number changes (n = 1977), mRNA expression (n = 1977) and protein level (n = 1902). Breast cancer incidence was also explored in 58 patients with type II RTS. DNA replication dynamics and chemosensitivity was evaluated in RECQL4-depleted breast cancer cells in vitro. Amplification or gain in gene copy number (30.6%), high-level mRNA expression (51%) and high levels of protein (23%) significantly associated with aggressive tumour behaviour, including lymph node positivity, larger tumour size, HER2 overexpression, ER-negativity, triple-negative phenotypes and poor survival. RECQL4 depletion impaired the DNA replication rate and increased chemosensitivity in cultured breast cancer cells. Thus, although recognized as a ’safe guardian of the genome’, our data provide compelling evidence that RECQL4 is tumour promoting in established breast cancers
