    On the shopfloor: exploring the impact of teacher trade unions on school-based industrial relations

    Teachers are highly unionised workers and their trade unions exert an important influence on the shaping and implementation of educational policy. Despite this importance there is relatively little analysis of the impact of teacher trade unions in educational management literature. Very little empirical research has sought to establish the impact of teacher unions at school level. In an era of devolved management and quasi-markets this omission is significant. New personnel issues continue to emerge at school level and this may well generate increased trade union activity at the workplace. This article explores the extent to which devolved management is drawing school-based union representation into a more prominent role. It argues that whilst there can be significant differences between individual schools, increased school autonomy is raising the profile of trade union activity in the workplace, and this needs to be better reflected in educational management research

    Delineation of a unique protein-protein interaction site on the surface of the estrogen receptor

    Recent studies have identified a series of estrogen receptor (ER)interacting peptides that recognize sites that are distinct from the classic coregulator recruitment (AF2) region. Here, we report the structural and functional characterization of an ER alpha-specific peptide that binds to the liganded receptor in an AF2-independent manner. The 2-angstrom crystal structure of the ER/peptide complex reveals a binding site that is centered on a shallow depression on the beta-hairpin face of the ligand-binding domain. The peptide binds in an unusual extended conformation and makes multiple contacts with the ligand-binding domain. The location and architecture of the binding site provides an insight into the peptide's ER subtype specificity and ligand interaction preferences. In vivo, an engineered coactivator containing the peptide motif is able to strongly enhance the transcriptional activity of liganded ER alpha, particularly in the presence of 4-hydroxytamoxifen. Furthermore, disruption of this binding surface alters ER's response to the coregulator TIF2. Together, these results indicate that this previously unknown interaction site represents a bona fide control surface involved in regulating receptor activity

    A signature motif mediating selective interactions of BCL11A with the NR2E/F subfamily of orphan nuclear receptors

    Despite their physiological importance, selective interactions between nuclear receptors (NRs) and their cofactors are poorly understood. Here, we describe a novel signature motif (F/YSXXLXXL/Y) in the developmental regulator BCL11A that facilitates its selective interaction with members of the NR2E/F subfamily. Two copies of this motif (named here as RID1 and RID2) permit BCL11A to bind COUP-TFs (NR2F1;NR2F2;NR2F6) and Tailless/TLX (NR2E1), whereas RID1, but not RID2, binds PNR (NR2E3). We confirmed the existence of endogenous BCL11A/TLX complexes in mouse cortex tissue. No interactions of RID1 and RID2 with 20 other ligand-binding domains from different NR subtypes were observed. We show that RID1 and RID2 are required for BCL11A-mediated repression of endogenous γ-globin gene and the regulatory non-coding transcript Bgl3, and we identify COUP-TFII binding sites within the Bgl3 locus. In addition to their importance for BCL11A function, we show that F/YSXXLXXL/Y motifs are conserved in other NR cofactors. A single FSXXLXXL motif in the NR-binding SET domain protein NSD1 facilitates its interactions with the NR2E/F subfamily. However, the NSD1 motif incorporates features of both LXXLL and FSXXLXXL motifs, giving it a distinct NR-binding pattern in contrast to other cofactors. In summary, our results provide new insights into the selectivity of NR/cofactor complex formation

    Actitudes y percepciones de los docentes universitarios en el proceso de adopción de una innovación. La iniciativa OpenCourseWare en la Universidad de Valencia (España)

    El proyecto OpenCourseWare, repositorio de recursos educativos en abierto, se inició en la Universidad de Valencia en 2008 y en 2009, sólo incluía 10 asignaturas, ninguna de ellas del área de ciencias de la salud. El objetivo de este trabajo es investigar las actitudes de los docentes de esta área así como sus percepciones con relación a este repositorio para determinar cuáles son las estrategias necesarias para extender su uso. Se parte de la teoría de la difusión de las innovaciones de Rogers (2003) y de un enfoque cualitativo a través del muestreo teórico, entrevistas semiestructuradas y codificación abierta. Los resultados indican la existencia de dos grandes grupos de profesores. Los adoptantes tempranos con una predisposición alta a participar en la iniciativa y con una percepción positiva de sus atributos y los mainstream o corriente principal más reacios a participar y con una percepción negativa respecto a alguno de sus atributos. La identificación de las características de estos dos grandes grupos marca las líneas de acción y de investigación futura en el ámbito del marketing y difusión, formación, sensibilización y motivación para extender el uso del OCW. The OpenCourseWare project, an open learning resource repository, was set up at the University of Valencia between 2008 and 2009. It initially consisted of only 10 subjects, none of which was in the field of health sciences. The objective of this research is to investigate the attitudes and perceptions of teaching staff in the field of health sciences with regard to the project in order to identify strategies to increase its use and expand its coverage. The diffusion of innovations theory as propounded by Rogers is used, along with a qualitative approach using theoretical sampling, semi-structured interviews and open coding. The results indicate the existence of two categories of teaching staff. Firstly, those who adopt an innovation early, have a marked inclination to participate in the initiative and a positive perception of the characteristics of the innovation. Secondly, a larger category of those more reluctant to participate in it and with a negative perception of one or more of its characteristics. Identifying the nature of these two groups provides guidelines for future action and research in the marketing and distribution of such projects, and the training, awareness and motivation required in order to extend the use of OCW

    Fibronectin rescues estrogen receptor α from lysosomal degradation in breast cancer cells

    Estrogen receptor α (ERα) is expressed in tissues as diverse as brains and mammary glands. In breast cancer, ERα is a key regulator of tumor progression. Therefore, understanding what activates ERα is critical for cancer treatment in particular and cell biology in general. Using biochemical approaches and superresolution microscopy, we show that estrogen drives membrane ERα into endosomes in breast cancer cells and that its fate is determined by the presence of fibronectin (FN) in the extracellular matrix; it is trafficked to lysosomes in the absence of FN and avoids the lysosomal compartment in its presence. In this context, FN prolongs ERα half-life and strengthens its transcriptional activity. We show that ERα is associated with β1-integrin at the membrane, and this integrin follows the same endocytosis and subcellular trafficking pathway triggered by estrogen. Moreover, ERα+ vesicles are present within human breast tissues, and colocalization with β1-integrin is detected primarily in tumors.     Teacher unionism in changing times: is this the real “new unionism”?

    This article provides a case study of union change in an environment in which radical school restructuring is taking place, and active strategies to weaken and marginalize organized teachers are being pursued by the state. The case study union is the National Union of Teachers in England. The article explores a number of different strategies open to teacher unions, utilizing a framework provided by Turner (2004). Drawing on data collected at a national level, and in three local authority areas, I argue that the National Union of Teachers’ response to the erosion of collective bargaining is best presented as an amalgam of strategies focused on workplace organizing, political campaigning, and coalition building. The data demonstrate considerable congruence between national and local strategies, although local data reveal considerable challenges in implementation and consequently considerable unevenness in local experiences

    The leadership component of Kelly’s Mobilisation Theory : contribution, tensions, limitations and further development

    This reassessment of Kelly’s analysis of the relationship of activist leadership to collective action within the overall jigsaw of mobilisation theory draws on social movement literature, studies by industrial relations scholars utilising aspects of Kelly’s approach – including this author’s own work – and related research on union leadership within collective mobilisation. In the process, it identifies and celebrates how Kelly’s work, whilst contributing a distinct and substantive actor-related approach, recognised that leadership is one ingredient amongst other factors, including important structural opportunities and constraints. It considers three potential ambiguities/tensions within Kelly’s conceptualisation of leadership related to the social construction of workers’ interests, spontaneity of workers’ action and the ‘leader/follower’ interplay. The review also identifies two important limitations, related to the union member/bureaucracy dynamic and the role of left-wing political leadership, and concludes by signalling different forms of leadership relationships on which further refinement and development would be fruitful

    Coregulator Control of Androgen Receptor Action by a Novel Nuclear Receptor-Binding Motif

    The androgen receptor (AR) is a ligand-activated transcription factor that is essential for prostate cancer development. It is activated by androgens through its ligand-binding domain (LBD), which consists predominantly of 11 α-helices. Upon ligand binding, the last helix is reorganized to an agonist conformation termed activator function-2 (AF-2) for coactivator binding. Several coactivators bind to the AF-2 pocket through conserved LXXLL or FXXLF sequences to enhance the activity of the receptor. Recently, a small compound-binding surface adjacent to AF-2 has been identified as an allosteric modulator of the AF-2 activity and is termed binding function-3 (BF-3). However, the role of BF-3 in vivo is currently unknown, and little is understood about what proteins can bind to it. Here we demonstrate that a duplicated GARRPR motif at the N terminus of the cochaperone Bag-1L functions through the BF-3 pocket. These findings are supported by the fact that a selective BF-3 inhibitor or mutations within the BF-3 pocket abolish the interaction between the GARRPR motif(s) and the BF-3. Conversely, amino acid exchanges in the two GARRPR motifs of Bag-1L can impair the interaction between Bag-1L and AR without altering the ability of Bag-1L to bind to chromatin. Furthermore, the mutant Bag-1L increases androgen-dependent activation of a subset of AR targets in a genome-wide transcriptome analysis, demonstrating a repressive function of the GARRPR/BF-3 interaction. We have therefore identified GARRPR as a novel BF-3 regulatory sequence important for fine-tuning the activity of the AR

    Heterodimers of photoreceptor-specific nuclear receptor (PNR/NR2E3) and peroxisome proliferator-activated receptor (PPARγ) are disrupted by retinal disease-associated mutations

    Photoreceptor-specific nuclear receptor (PNR/NR2E3) and Tailless homolog (TLX/NR2E1) are human orthologs of the NR2E group, a subgroup of phylogenetically related members of the Nuclear Receptor (NR) superfamily of transcription factors. We assessed the ability of these NRs to form heterodimers with other members of the human NRs representing all major subgroups. The TLX ligand binding domain (LBD) did not appear to form homodimers or interact directly with any other NR tested. The PNR LBD was able to form homodimers, but also exhibited robust interactions with the LBDs of PPARγ/NR1C3 and TRβ/NR1A2. The binding of PNR to PPARγ was specific for this paralog, as no interaction was observed with the LBDs of PPARαNR1C1 or PPARδNR1C2. In support of these findings, PPARγ and PNR were found to be co-expressed in human retinal tissue extracts and could be co-immunoprecipitated as a native complex. Selected sequence variants in the PNR LBD associated with human retinopathies, or a mutation in the dimerization region of PPARγ LBD associated with familial partial lipodystrophy type 3, were found to disrupt PNR/PPARγ complex formation. Wild type PNR, but not a PNR309G mutant, was able to repress PPARγ-mediated transcription in reporter assays. In summary our results reveal novel heterodimer interactions in the NR superfamily, suggesting previously unknown functional interactions of PNR with PPARγ and TRβ that have potential importance in retinal development and disease
