    Mutations of penicillin acylase residue B71 extend substrate specificity by decreasing steric constraints for substrate binding

    Two mutant forms of penicillin acylase from Escherichia coli strains, selected using directed evolution for the ability to use glutaryl-L-leucine for growth [Forney, Wong and Ferber (1989) Appl. Environ. Microbiol. 55, 2550-2555], are changed within one codon, replacing the B-chain residue Phe(B71) with either Cys or Leu. Increases of up to a factor of ten in k(cat)/K-m values for substrates possessing a phenylacetyl leaving group are consistent with a decrease in K-s. Values of k(cat/)K(m) for glutaryl-L-leucine are increased at least 100-fold. A decrease in k(cat)/K-m for the CySB71 mutant with increased pH is consistent with binding of the uncharged glutaryl group. The mutant proteins are more resistant to urea denaturation monitored by protein fluorescence, to inactivation in the presence of substrate either in the presence of urea or at high pH, and to heat inactivation. The crystal structure of the Leu(B71) mutant protein, solved to 2 X resolution, shows a flip of the side chain of Phe(B256) into the periphery of the catalytic centre, associated with loss of the pi-stacking interactions between Phe(B256) and Phe(B71). Molecular modelling demonstrates that glutaryl-L-leucine may bind with the uncharged glutaryl group in the S-1 subsite of either the wild-type or the Leu(B71) mutant but with greater potential freedom of rotation of the substrate leucine moiety in the complex with the mutant protein. This implies a smaller decrease in the conformational entropy of the substrate on binding to the mutant proteins and consequently greater catalytic activity

    Predicting the distribution of canine leishmaniasis in western Europe based on environmental variables.

    The domestic dog is the reservoir host of Leishmania infantum, the causative agent of zoonotic visceral leishmaniasis endemic in Mediterranean Europe. Targeted control requires predictive risk maps of canine leishmaniasis (CanL), which are now explored. We databased 2187 published and unpublished surveys of CanL in southern Europe. A total of 947 western surveys met inclusion criteria for analysis, including serological identification of infection (504, 369 dogs tested 1971-2006). Seroprevalence was 23 2% overall (median 10%). Logistic regression models within a GIS framework identified the main environmental predictors of CanL seroprevalence in Portugal, Spain, France and Italy, or in France alone. A 10-fold cross-validation approach determined model capacity to predict point-values of seroprevalence and the correct seroprevalence class (20%). Both the four-country and France-only models performed reasonably well for predicting correctly the 20% seroprevalence classes (AUC >0 70). However, the France-only model performed much better for France than the four-country model. The four-country model adequately predicted regions of CanL emergence in northern Italy (<5% seroprevalence). Both models poorly predicted intermediate point seroprevalences (5-20%) within regional foci, because surveys were biased towards known rural foci and Mediterranean bioclimates.     On-site forest fire smoke detection by low-power autonomous vision sensor

    Trabajo presentado a la VI International Conference on Forest Fire Research celebrada en Coimbra (Portugal) del 15 al 18 de noviembre de 2010.Early detection plays a crucial role to prevent forest fires from spreading. Wireless vision sensor networks deployed throughout high-risk areas can perform fine-grained surveillance and thereby very early detection and precise location of forest fires. One of the fundamental requirements that need to be met at the network nodes is reliable low-power on-site image processing. It greatly simplifies the communication infrastructure of the network as only alarm signals instead of complete images are transmitted, anticipating thus a very competitive cost. As a first approximation to fulfill such a requirement, this paper reports the results achieved from field tests carried out in collaboration with the Andalusian Fire-Fighting Service (INFOCA). Two controlled burns of forest debris were realized (www.youtube.com/user/vmoteProject). Smoke was successfully detected on-site by the EyeRISTM v1.2, a general-purpose autonomous vision system, built by AnaFocus Ltd., in which a vision algorithm was programmed. No false alarm was triggered despite the significant motion other than smoke present in the scene. Finally, as a further step, we describe the preliminary laboratory results obtained from a prototype vision chip which implements, at very low energy cost, some image processing primitives oriented to environmental monitoring.This work is funded by CICE/JA and MICINN (Spain) through projects 2006-TIC-2352 and TEC2009-11812 respectively.Peer Reviewe

    Liberal governmentality in Spain: bodies, minds, and the medical construction of the “outsider,” 1870–1910

    This paper traces the fragility of the subject in the period extending from the aftermath of the Sexenio through to the early twentieth century. In particular, two case studies are focused upon: the question of gender “deviance” and the figure of the genius, in order to understand how medicine participated in the construction of “outsider” identities within the context of the emerging liberal order. How did liberal rationales exclude or curtail certain wayward expressions of identity and subjectivity? What consequences did the marking of “excessive” figures or outsiders have for notions of inclusiveness and citizenship within the late-nineteenth-century liberal order? By concentrating primarily on medical texts and journals published during the period, this study builds on existing research to tease out answers to these questions

    Enhancing magnetorheological effect using bimodal suspensions in the singlemultidomain limit

    We demonstrate a new route to enhance the magnetorheological effect using bimodal suspensions in the single-multidomain limit. Experimental results are satisfactorily compared to 3D finite element method simulations.     Evaluation of urinary hydrogen peroxide as an oxidative stress biomarker in a healthy Japanese population

    The usefulness of urinary hydrogen peroxide (H2O2) as an oxidative stress biomarker was evaluated in 766 healthy Japanese. The mean level of urinary concentrations of H2O2 was 5.66 +/- 8.27 mu mol/g creatinine, and was significantly higher in females than in males. Significant correlations of H2O2 were observed with age, aspartate aminotransferase (AST), alanine aminotransferase (ALT), total cholesterol (TC), low-density lipoprotein cholesterol (LDL-c), insulin, 8-hydroxy-2'-deoxyguanosine (8-OHdG), and exercise habit in females. In both sexes, H2O2 showed a significant correlation with 8-OHdG. By a multiple logistic regression analysis, urinary H2O2 was positively associated with urinary 8-OHdG and TC and was inversely associated with insulin. By stratification of sex and age, the association of urinary H2O2 with TC was positive in both sexes under 50 years old and was inverse in males over 50 years old, and that with insulin was inverse in males over 50 years old and in females under 50 years old. Moreover, by stratification of sex and age, a positive association of H2O2 with exercise and an inverse association of H2O2 with alcohol consumption became clear in males under 50 years old, although there were no significant odds for H2O2 after adjustment for covariates. In conclusion, the present results suggest that urinary H2O2 is a useful biomarker for oxidative stress, showing an association with 8-OHdG, TC, and insulin independently

    Geographic Range Expansion and Taxonomic Notes of the Shortfin Neoscopelid Neoscopelus cf. microchir (Myctophiformes: Neoscopelidae) in the North-Eastern Atlantic

    One specimen of the shortfin neoscopelid Neoscopelus microchir Matsubara, 1943, has been recorded for the first time on the Porcupine Bank, southwestern Ireland, providing a new northern limit of distribution for the eastern Atlantic. Morphometric and meristic parameters confirm the taxonomic identification. However, DNA barcoding shows deficiencies in current taxonomy and the potential occurrence of cryptic species. On this basis, the specimen is cautiously reported as Neosopelus cf. microchir pending a taxonomic revision of the genus.Versión del edito

    Seropositivity rates for agents of canine vector-borne diseases in Spain : a multicentre study

    Background: Controlling canine vector-borne diseases (CVBD) is a major concern, since some of these diseases are serious zoonoses. This study was designed to determine seropositivity rates in Spain for agents causing the following five CVBD: leishmaniosis (Leishmania infantum: Li), heartworm (Dirofilaria immitis: Di), ehrlichiosis (Ehrlichia canis: Ec), anaplasmosis (Anaplasma phagocytophilum/Anaplasma platys: An) and Lyme disease (Borrelia burgdorferi: Bb). Methods: Anti-An, -Bb, and -Ec antibodies and the Di antigen were determined using the 4DX SNAP® Test (IDEXX Laboratories) and anti-L. infantum (Li) antibodies using the Leishmania SNAP® Test (IDEXX Laboratories) in blood and/or serum samples. Results: Among 1100 dogs examined, overall seropositivity rates were: Li (15.7%), Ec (5%), An (3.1%), Di (1.25%) and Bb (0.4%). While seropositivity towards Bb and Di was similar in all geographic regions, rates were significantly higher in the east of Spain (8.3%) for An, significantly higher in the north (20%) for Ec, and significantly higher in the Southeast (46.6%) and South (27.4%), and significantly lower in the north (0%) for Li. No statistical associations were observed between sex and the CVBD analyzed (p ≥ 0.05) while the following associations with other variables were detected: a higher seropositivity to Ec (40%) and Bb (6.7%) in dogs under one year of age compared with adults (p < 0.05); and a higher seropositivity to An and Li in dogs that lived outdoors versus indoors (p = 0.01; p < 0.001, respectively). Seropositivity rates of 2.1%, 0%, 1.7%, 0.5% and 4.2% were recorded respectively for An, Bb, Ec, Di and Li in dogs with no clinical signs (n = 556) versus 3.8%, 0.6%, 7.5%, 1.8% and 25.9% for those with signs (n = 507) suggestive of a CVBD. Conclusion: The data obtained indicate a risk for dogs in Spain of acquiring any of the five CVBD examined. Veterinarians in the different regions should include these diseases in their differential diagnoses and recommend the use of repellents and other prophylactic measures to prevent disease transmission by arthropod vectors. Public health authorities also need to become more involved in the problem, since some of the CVBD examined here also affect humans

    Functional traits related to competition for light influence tree diameter increments in a biodiversity manipulation experiment

    Understanding how functional traits and diversity modulate plant interactions within forests is becoming a widespread research goal in ecology. We applied neighbourhood analysis to a Mediterranean biodiversity manipulation experiment (IDENT-Macomer) to assess the importance of functional traits in predicting tree diameter increments (DI) in a mixed forest. We used tree functional traits to weigh the neighbourhood competition index (NCI) and functional dispersion (FDis), which is a functional diversity metric. We found that functional traits affect competitive performance across species within a mixed forest and that resource acquisition is based primarily on trait hierarchy. We also found that traits related to competition for light, such as maximum plant height (Hmax), are the best predictors of DI. Our results reveal that NCI is a more reliable predictor than FDis, but the combination of both effects helps to better explain differences in DI. Finally, our findings show that gathering functional trait data is a practise that should be prioritised in mixed forest management due to the predictive importance of NCI and FDis in experiments with high density and species diversity

    Ternary solid-ferrofluid-liquid magnetorheological fluids

    A new kind of magnetorheological fluid is proposed that exhibits both enhanced magnetorheological effect and kinetic stability against sedimentation.
