Versatile thiol-based reactions for micrometer- and nanometer-scale photopatterning of polymers and biomolecules
Thiol-based chemistry provides a mild and versatile tool for surface functionalization. In the present work, mercaptosilane films were patterned by utilizing UV-induced photo-oxidation of the thiol to yield sulfonate groups via contact and interferometric lithography (IL). These photo-generated sulfonic acid groups were used for selective immobilization of amino-functionalized molecules after activation with triphenylphosphine ditriflate (TPPDF). Moreover, protein-resistant poly(oligoethyleneglycolmethacrylate) (POEGMA) brushes were grown from the intact thiol groups by a surface-induced polymerization reaction. Exploiting both reactions it is possible to couple amino-labelled nitrilotriacetic acid (NH2-NTA) to sulfonate-functionalized regions, enabling the site-specific binding of green fluorescent protein (GFP) to regions defined lithographically, while exploiting the protein-resistant character of POEGMA brushes to prevent non-specific protein adsorption to previously masked areas. The outstanding reactivity of thiol groups paves the way towards novel strategies for the fabrication of complex protein nanopatterns beyond thiol–ene chemistry
Uterine prolapse prevention in Eastern Nepal: the perspectives of women and health care professionals
Abstract:Uterine prolapse is a major reproductive health issue in Nepal. There is a wide range of literature available on the causes and risk factors of uterine prolapse and on the ways to prevent and treat it. There is still a lack of published evidence on what prevention and treatment services are working well or the attitudes toward them. This paper presents the findings of a qualitative study on primary and secondary prevention of uterine prolapse in Eastern Nepal.Method:The study involved eight focus group discussions with 71 women in six villages of the eastern districts of Siraha and Saptari and 14 qualitative interviews with health professionals from the local to central level. The group discussions and interviews covered the awareness levels of uterine prolapse and its prevention and treatment, as well as participants’ opinions on and experiences with the services offered.Results:It was found that patriarchy, gender discrimination, and cultural traditions such as early marriage and pregnancy make it difficult for people to discontinue uterine prolapse risk behaviors. Women are aware of risk factors, prevention, and treatment, but are powerless to change their situations. Health professionals and women are fond of surgery as treatment, but opinions on the use of ring pessaries and pelvic floor muscle training are split.Conclusion:The main recommendation that can be drawn from this study is that research on the effectiveness of early treatments, such as ring pessaries and exercise, should be conducted. Furthermore, the involvement of other target groups (husbands, adolescents, and mothers-in-law) needs to be increased in order to make it easier for women to adapt low-risk behaviors. Finally, uterine prolapse prevention should be better integrated in national reproductive health services. Enforcing transparency, monitoring systems, and collaborations are important factors that should be considered as well.Keywords: uterine prolapse, primary prevention, secondary prevention, awareness, Nepa

Gender Discrimination in Hiring: Evidence from a Cross-National Harmonized Field Experiment

which permits unrestricted reuse, distribution, and reproduction in any medium, provided the original work is properly citedGender discrimination is often regarded as an important driver of women’s disadvantage in the labour market, yet earlier studies show mixed results. However, because different studies employ different research designs, the estimates of discrimination cannot be compared across countries. By utilizing data from the first harmonized comparative field experiment on gender discrimination in hiring in six countries, we can directly compare employers’ callbacks to fictitious male and female applicants. The countries included vary in a number of key institutional, economic, and cultural dimensions, yet we found no sign of discrimination against women. This cross-national finding constitutes an important and robust piece of evidence. Second, we found discrimination against men in Germany, the Netherlands, Spain, and the UK, and no discrimination against men in Norway and the United States. However, in the pooled data the gender gradient hardly differs across countries. Optimal particle parameters for CLC and CLR processes - predictions by intra-particle transport models and experimental validation
Validated models for predicting oxidation and reduction kinetics of multi-component porous particles in chemical looping combustion (CLC) and chemical looping reforming (CLR) processes are of key importance to identify the rate limiting step in these processes. Since particle properties (i.e., their composition, porosity, pore size, grain size, etc.) can be adjusted by modern synthesis techniques, there is an open question on the optimal set of these properties that would lead to the most economic process. We introduce a general open-source simulation environment, called ParScale that can be used to simulate models relevant for CLC and CLR processes, and hence can be used for their optimization. Most important, ParScale features a generalized one-dimensional spherical discretization which enables the user to predict an arbitrary number of reactions within non-isothermal porous particles consisting of multiple solid (reactive or inert) species. We perform an optimization study (constrained by typical process requirements like the maximum reaction time) for an isothermal first-order reaction, as well as for an n-th order reaction typical for hematite reduction. Very Late Antigen-4 (α<inf>4</inf>β<inf>1</inf> Integrin) Targeted PET Imaging of Multiple Myeloma
Biomedical imaging techniques such as skeletal survey and 18F-fluorodeoxyglucose (FDG)/Positron Emission Tomography (PET) are frequently used to diagnose and stage multiple myeloma (MM) patients. However, skeletal survey has limited sensitivity as it can detect osteolytic lesions only after 30-50% cortical bone destruction, and FDG is a marker of cell metabolism that has limited sensitivity for intramedullary lesions in MM. Targeted, and non-invasive novel probes are needed to sensitively and selectively image the unique molecular signatures and cellular processes associated with MM. Very late antigen-4 (VLA-4; also called α4β1 integrin) is over-expressed on MM cells, and is one of the key mediators of myeloma cell adhesion to the bone marrow (BM) that promotes MM cell trafficking and drug resistance. Here we describe a proof-of-principle, novel molecular imaging strategy for MM tumors using a VLA-4 targeted PET radiopharmaceutical, 64Cu-CB-TE1A1P-LLP2A. Cell uptake studies in a VLA-4-positive murine MM cell line, 5TGM1, demonstrated receptor specific uptake (P<0.0001, block vs. non-block). Tissue biodistribution at 2 h of 64Cu-CB-TE1A1P-LLP2A in 5TGM1 tumor bearing syngeneic KaLwRij mice demonstrated high radiotracer uptake in the tumor (12±4.5%ID/g), and in the VLA-4 rich organs, spleen (8.8±1.0%ID/g) and marrow (11.6±2.0%ID/g). Small animal PET/CT imaging with 64Cu-CB-TE1A1P-LLP2A demonstrated high uptake in the 5TGM1 tumors (SUV 6.6±1.1). There was a 3-fold reduction in the in vivo tumor uptake in the presence of blocking agent (2.3±0.4). Additionally, 64Cu-CB-TE1A1P-LLP2A demonstrated high binding to the human MM cell line RPMI-8226 that was significantly reduced in the presence of the cold targeting agent. These results provide pre-clinical evidence that VLA-4-targeted imaging using 64Cu-CB-TE1A1P-LLP2A is a novel approach to imaging MM tumors. © 2013 Soodgupta et al
