Taurine Treatment Modulates Circadian Rhythms in Mice Fed A High Fat Diet
Close ties have been made among certain nutrients, obesity, type 2 diabetes and circadian clocks. Among nutrients, taurine has been documented as being effective against obesity and type 2 diabetes. However, the impact of taurine on circadian clocks has not been elucidated. We investigated whether taurine can modulate or correct disturbances in daily rhythms caused by a high-fat diet in mice. Male C57BL/6 mice were divided in four groups: control (C), control + taurine (C+T), high-fat diet (HFD) and HFD + taurine (HFD+T). They were administered 2% taurine in their drinking water for 10 weeks. Mice were euthanized at 6:00, 12:00, 18:00, and 24:00. HFD mice increased body weight, visceral fat and food intake, as well as higher levels of glucose, insulin and leptin, throughout the 24 h. Taurine prevented increments in food intake, body weight and visceral fat, improved glucose tolerance and insulin sensitivity and reduced disturbances in the 24 h patterns of plasma insulin and leptin. HFD downregulated the expression of clock genes Rev-erbα, Bmal1, and Per1 in pancreatic islets. Taurine normalized the gene and protein expression of PER1 in beta-cells, which suggests that it could be beneficial for the correction of daily rhythms and the amelioration of obesity and diabetes
Comparison of adeno-associated virus pseudotype 1, 2, and 8 vectors administered by intramuscular injection in the treatment of murine phenylketonuria
Phenylketonuria (PKU) is caused by hepatic phenylalanine hydroxylase (PAH) deficiency and is associated with systemic accumulation of phenylalanine (Phe). Previously we demonstrated correction of murine PKU after intravenous injection of a recombinant type 2 adeno-associated viral vector pseudotyped with type 8 capsid (rAAV2/8), which successfully directed hepatic transduction and Pah gene expression. Here, we report that liver PAH activity and phenylalanine clearance were also restored in PAH-deficient mice after simple intramuscular injection of either AAV2 pseudotype 1 (rAAV2/1) or rAAV2/8 vectors. Serotype 2 AAV vector (rAAV2/2) was also investigated, but long-term phenylalanine clearance has been observed only for pseudotypes 1 and 8. Therapeutic correction was shown in both male and female mice, albeit more effectively in males, in which correction lasted for the entire period of the experiment (>1 year). Although phenylalanine levels began to rise in female mice at about 8-10 months after rAAV2/8 injection they remained only mildly hyperphenylalaninemic thereafter and subsequent supplementation with synthetic tetrahydrobiopterin resulted in a transient decrease in blood phenylalanine. Alternatively, subsequent administration of a second vector with a different AAV pseudotype to avoid immunity against the previously administrated vector was also successful for long-term treatment of female PKU mice. Overall, this relatively less invasive gene transfer approach completes our previous studies and allows comparison of complementary strategies in the development of efficient PKU gene therapy protocols
SMILE: A joint ESA/CAS mission to investigate the interaction between the solar wind and Earth's magnetosphere
The Solar wind Magnetosphere Ionosphere Link Explorer (SMILE) is a collaborative science mission between ESA and the Chinese Academy of Sciences (CAS). SMILE is a novel self-standing mission to observe the coupling of the solar wind and Earth's magnetosphere via X-Ray imaging of the solar wind -- magnetosphere interaction zones, UV imaging of global auroral distributions and simultaneous in-situ solar wind, magnetosheath plasma and magnetic field measurements. The SMILE mission proposal was submitted by a consortium of European, Chinese and Canadian scientists following a joint call for mission by ESA and CAS. It was formally selected by ESA's Science Programme Committee (SPC) as an element of the ESA Science Program in November 2015, with the goal of a launch at the end of 2021.
In order to achieve its scientific objectives, the SMILE payload will comprise four instruments: the Soft X-ray Imager (SXI), which will spectrally map the Earth's magnetopause, magnetosheath and magnetospheric cusps; the UltraViolet Imager (UVI), dedicated to imaging the auroral regions; the Light Ion Analyser (LIA) and the MAGnetometer (MAG), which will establish the solar wind properties simultaneously with the imaging instruments. We report on the status of the mission and payload developments and the findings of a design study carried out in parallel at the concurrent design facilities (CDF) of ESA and CAS in October/November 2015. © (2016) COPYRIGHT Society of Photo-Optical Instrumentation Engineers (SPIE). Topoisomer Differentiation of Molecular Knots by FTICR MS: Lessons from Class II Lasso Peptides
Lasso peptides constitute a class of bioactive peptides sharing a knotted
structure where the C-terminal tail of the peptide is threaded through and
trapped within an N-terminalmacrolactamring. The structural characterization of
lasso structures and differentiation from their unthreaded topoisomers is not
trivial and generally requires the use of complementary biochemical and
spectroscopic methods. Here we investigated two antimicrobial peptides
belonging to the class II lasso peptide family and their corresponding
unthreaded topoisomers: microcin J25 (MccJ25), which is known to yield
two-peptide product ions specific of the lasso structure under collisioninduced
dissociation (CID), and capistruin, for which CID does not permit to
unambiguously assign the lasso structure. The two pairs of topoisomers were
analyzed by electrospray ionization Fourier transform ion cyclotron resonance
mass spectrometry (ESI-FTICR MS) upon CID, infrared multiple photon
dissociation (IRMPD), and electron capture dissociation (ECD). CID and
ECDspectra clearly permitted to differentiate MccJ25 from its non-lasso
topoisomer MccJ25-Icm, while for capistruin, only ECD was informative and
showed different extent of hydrogen migration (formation of c\bullet/z from
c/z\bullet) for the threaded and unthreaded topoisomers. The ECD spectra of the
triply-charged MccJ25 and MccJ25-lcm showed a series of radical b-type product
ions {\eth}b0In{\TH}. We proposed that these ions are specific of
cyclic-branched peptides and result from a dual c/z\bullet and y/b
dissociation, in the ring and in the tail, respectively. This work shows the
potentiality of ECD for structural characterization of peptide topoisomers, as
well as the effect of conformation on hydrogen migration subsequent to electron
capture
Cell-Oriented Modeling of Angiogenesis
Due to its significant involvement in various physiological and pathological conditions, angiogenesis (the development of new blood vessels from an existing vasculature) represents an important area of the actual biological research and a field in which mathematical modeling proved particularly useful in supporting the experimental work. In this paper, we focus on a specific modeling strategy, known as “cell-centered” approach. This type of mathematical models work at a “mesoscopic scale,” assuming the cell as the natural level of abstraction for computational modeling of development. They treat cells phenomenologically, considering their essential behaviors to study how tissue structure and organization emerge from the collective dynamics of multiple cells. The main contributions of the cell-oriented approach to the study of the angiogenic process will be described. From one side, they have generated “basic science understanding” about the process of capillary assembly during development, growth, and pathology. On the other side, models were also developed supporting “applied biomedical research” for the purpose of identifying new therapeutic targets and clinically relevant approaches for either inhibiting or stimulating angiogenesis
