    Avaliação de risco em mioloma múltiplo: resultados preliminares do grupo brasileiro de estudos de mieloma

    The Durie/Salmon staging system continues to be used worldwide in patients with multiple myeloma. However, in recent years, new systems have been proposed. The International Myeloma Working Group performed a retrospective study with 11,179 patients and proposed an "International Staging System" utilizing serum levels of â2 microglobulin and albumin. In addition, current research has focused on the usefulness of cytogenetic and molecular data as prognostic factors. These data suggest that these parameters are powerful discriminators of a poor prognostic group of myeloma patients. Indeed, these prognostic indexes have been utilized in clinical trials, with interesting and encouraging results.O esquema de Durie / Salmon continua a ser utilizado para estadiar os pacientes com mieloma múltiplo. Recentemente, um novo sistema mais simples e eficaz foi proposto. O "International Myeloma Working Group" realizou um estudo retrospectivo com 11.179 pacientes e a partir destes dados propôs a criação de um "International Staging System (ISS)" utilizando os níveis séricos de ß2 microglobulina e de albumina ao diagnóstico. Além do ISS a pesquisa está voltada para identificar alterações citogenéticas e moleculares que se correlacionem com o prognóstico no mieloma múltiplo.     A concretização do princípio ne bis in idem e o sistema de responsabilização da improbidade administrativa

    O presente estudo versa sobre a incidência de mais de um regime de responsabilização sobre o mesmo fato, com foco no regime da improbidade administrativa, a partir das alterações legislativas trazidas pela Lei 14.230/21. A partir da tensão entre o dever de o Estado responsabilizar aqueles que infringem a função administrativa com base nos sistemas de responsabilização criados no bojo do Estado Democrático de Direito sobre um mesmo fato e o dever de o Estado garantir e concretizar direitos fundamentais dos cidadãos na aplicação de sanções, pretende-se analisar, nas alterações trazidas pela 14.230/21, se há normas que se referem à aplicação concomitante dos sistemas de responsabilização sobre o mesmo fato - normas de referência. Ainda, de forma específica, como estas novas normas de referência tratam da incidência do sistema de responsabilização da improbidade administrativa e anticorrupção, regulamentada pela Lei nº 12.846/13, para, ao final, verificar-se se as novas regras jurídicas concretizam o princípio do ne bis in idem e reconfiguram a noção de independência entre as instâncias no sentido de vetor interpretativ

    CarPatch: A Synthetic Benchmark for Radiance Field Evaluation on Vehicle Components

    Neural Radiance Fields (NeRFs) have gained widespread recognition as a highly effective technique for representing 3D reconstructions of objects and scenes derived from sets of images. Despite their efficiency, NeRF models can pose challenges in certain scenarios such as vehicle inspection, where the lack of sufficient data or the presence of challenging elements (e.g. reflections) strongly impact the accuracy of the reconstruction. To this aim, we introduce CarPatch, a novel synthetic benchmark of vehicles. In addition to a set of images annotated with their intrinsic and extrinsic camera parameters, the corresponding depth maps and semantic segmentation masks have been generated for each view. Global and part-based metrics have been defined and used to evaluate, compare, and better characterize some state-of-the-art techniques. The dataset is publicly released at https://aimagelab.ing.unimore.it/go/carpatch and can be used as an evaluation guide and as a baseline for future work on this challenging topic.Comment: Accepted at ICIAP202

    Redox stress proteins are involved in adaptation response of the hyperthermoacidophilic archaeon Sulfolobus solfataricus to nickel challenge

    <p>Abstract</p> <p>Background</p> <p>Exposure to nickel (Ni) and its chemical derivatives has been associated with severe health effects in human. On the contrary, poor knowledge has been acquired on target physiological processes or molecular mechanisms of this metal in model organisms, including Bacteria and Archaea. In this study, we describe an analysis focused at identifying proteins involved in the recovery of the archaeon <it>Sulfolobus solfataricus </it>strain MT4 from Ni-induced stress.</p> <p>Results</p> <p>To this purpose, <it>Sulfolobus solfataricus </it>was grown in the presence of the highest nickel sulphate concentration still allowing cells to survive; crude extracts from treated and untreated cells were compared at the proteome level by using a bi-dimensional chromatography approach. We identified several proteins specifically repressed or induced as result of Ni treatment. Observed up-regulated proteins were largely endowed with the ability to trigger recovery from oxidative and osmotic stress in other biological systems. It is noteworthy that most of the proteins induced following Ni treatment perform similar functions and a few have eukaryal homologue counterparts.</p> <p>Conclusion</p> <p>These findings suggest a series of preferential gene expression pathways activated in adaptation response to metal challenge.</p

    [Purpose]  This article addresses the configuration of social control as a constitutional principle and its importance within the scope of Law no. 11,445/2007. [Methodology/approach/design] It analyzes the rules of social control in Law no. 11,445/2007 and the impact of changes promoted by Law no. 14,026/2020. [Findings] It identifies the influence of social control on the regionalization of the basic sanitation service and the legality of the Paulista Law no. 17,231/2021, from the viewpoint of social control, which instituted regional units in the State of Sao Paulo.[Purpose]  This article addresses the configuration of social control as a constitutional principle and its importance within the scope of Law no. 11,.445/2007. [Methodology/approach/design] It analyzes the rules of social control in Law no. 11,445/2007 and the impact of changes promoted by Law no. 14,026/2020. [Resultados] Ao final, este artigo identifica a influência do controle social na regionalização do serviço de saneamento básico e a legalidade da Lei Paulista 17.231/21 no aspecto do controle social, que instituiu unidades regionais no Estado de São Paulo.

    Case report: Prader-Willi syndrome and inflammatory arthritis-An important consideration

    BackgroundPrader-Willi syndrome (PWS) is a multisystemic genetically determined disorder. Musculoskeletal manifestations are common in most patients. We report the cases of two children with PWS who developed inflammatory arthritis, complicated with chronic anterior bilateral uveitis in one case. To our knowledge, no previous reports of such an association exist. Case presentationCase 1 was of a 3-year-old girl diagnosed with PWS who developed arthritis of the right knee with morning stiffness, joint swelling, and limited range of motion. Other causes of arthritis were ruled out. Increased inflammatory markers, antinuclear antibody (ANA) positivity, and hypertrophic synovitis on ultrasound confirmed the diagnosis of inflammatory arthritis compatible with juvenile idiopathic arthritis (JIA). Despite the treatment with methotrexate, arthritis progressed, and etanercept was added. The patient reached and maintained articular remission while on combined MTX and etanercept treatment during 9 years of follow-up. Case 2 was of a 6-year-old boy diagnosed with PWS who developed arthritis of the right knee. Laboratory investigations showed mildly increased acute phase reactants, microcytic anemia, and ANA positivity at high titer (titer 1:1,280). Infectious and other causes of arthritis were excluded. Ultrasound confirmed the presence of joint effusion and synovial thickening, and synovial fluid analysis was consistent with inflammatory arthrosynovitis (white blood cell count of 14,200/mu l) compatible with JIA. Shortly after the diagnosis, the ophthalmologic evaluation revealed the presence of bilateral anterior uveitis. Despite MTX and topical corticosteroid, ocular inflammation persisted and adalimumab was added. At the last follow-up, 9 months later, the child experienced inactivity of arthritis and uveitis with normal growth. ConclusionsWe aim to raise awareness of this possible association among pediatricians since arthritis might be underestimated due to high pain tolerance, behavioral disturbances, and other musculoskeletal abnormalities in PWS patients

    Enhancement of Human Prolactin Synthesis by Sodium Butyrate Addition to Serum-Free CHO Cell Culture

    Sodium butyrate (NaBu) has been used as a productivity enhancer for the synthesis of recombinant proteins in Chinese hamster ovary (CHO) cells. Thus, the influence of NaBu on the production of recombinant human prolactin (hPRL) from CHO cells was investigated for the first time. CHO cell cultures were submitted to a treatment with different concentrations of NaBu (0.25 to 4 mM). Quantitative and qualitative analyses by reverse-phase high-performance liquid chromatography (RP-HPLC) and Western blot or SDS-PAGE, carried out directly on CHO-conditioned medium, showed that the highest hPRL expression was obtained with 1 mM NaBu. In vitro biological assays based on noble rat lymphoma (Nb2) and mouse pro-B lymphoma (Ba/F3-LLP) cells were carried out on purified hPRL. Its bioactivity in the presence of NaBu was not apparently different from that of the First International Reference Reagent of recombinant hPRL (WHO 97/714). Our results show that NaBu increased the synthesis of recombinant hPRL in CHO cells, apparently without compromising either its structure or function
