NEET young people and the labour market: working on the margins
This paper is based on findings from a longitudinal study of twenty young people who have spent significant periods of time categorised as NEET (not in education, employment or training). Drawing on three years of ethnographic research conducted across two local authorities in the north of England, it focuses on the lived experience of a set of young people as they move between various sites of exclusion and participation in the labour market. Central to the paper are the experiences of three individuals and their attempts to begin work in the retail, care and catering industries.
The paper illustrates a range of tensions between the aspirations of young people and the opportunities open to them. It provides a critical insight into some of the conditions which characterise work on the fringes of the labour market and the inter-play between these and the attitudes, values and dispositions of the young people taking part in the research. The paper’s findings challenge popular discourses about young people on the margins of participation and pose questions about the articulation between education, work and training for those seeking to enter the labour market
Structural and Functional Analysis of a β2-Adrenergic Receptor Complex with GRK5.
The phosphorylation of agonist-occupied G-protein-coupled receptors (GPCRs) by GPCR kinases (GRKs) functions to turn off G-protein signaling and turn on arrestin-mediated signaling. While a structural understanding of GPCR/G-protein and GPCR/arrestin complexes has emerged in recent years, the molecular architecture of a GPCR/GRK complex remains poorly defined. We used a comprehensive integrated approach of cross-linking, hydrogen-deuterium exchange mass spectrometry (MS), electron microscopy, mutagenesis, molecular dynamics simulations, and computational docking to analyze GRK5 interaction with the β2-adrenergic receptor (β2AR). These studies revealed a dynamic mechanism of complex formation that involves large conformational changes in the GRK5 RH/catalytic domain interface upon receptor binding. These changes facilitate contacts between intracellular loops 2 and 3 and the C terminus of the β2AR with the GRK5 RH bundle subdomain, membrane-binding surface, and kinase catalytic cleft, respectively. These studies significantly contribute to our understanding of the mechanism by which GRKs regulate the function of activated GPCRs. PAPERCLIP
The transcriptional repressor complex FRS7-FRS12 regulates flowering time and growth in Arabidopsis
Most living organisms developed systems to efficiently time environmental changes. The plant-clock acts in coordination with external signals to generate output responses determining seasonal growth and flowering time. Here, we show that two Arabidopsis thaliana transcription factors, FAR1 RELATED SEQUENCE 7 (FRS7) and FRS12, act as negative regulators of these processes. These proteins accumulate particularly in short-day conditions and interact to form a complex. Loss-of-function of FRS7 and FRS12 results in early flowering plants with overly elongated hypocotyls mainly in short days. We demonstrate by molecular analysis that FRS7 and FRS12 affect these developmental processes in part by binding to the promoters and repressing the expression of GIGANTEA and PHYTOCHROME INTERACTING FACTOR 4 as well as several of their downstream signalling targets. Our data reveal a molecular machinery that controls the photoperiodic regulation of flowering and growth and offer insight into how plants adapt to seasonal changes
Cooperative Metaheuristics for Exploring Proteomic Data
Most combinatorial optimization problems cannotbe solved exactly. A class of methods, calledmetaheuristics, has proved its efficiency togive good approximated solutions in areasonable time. Cooperative metaheuristics area sub-set of metaheuristics, which implies aparallel exploration of the search space byseveral entities with information exchangebetween them. The importance of informationexchange in the optimization process is relatedto the building block hypothesis ofevolutionary algorithms, which is based onthese two questions: what is the pertinentinformation of a given potential solution andhow this information can be shared? Aclassification of cooperative metaheuristicsmethods depending on the nature of cooperationinvolved is presented and the specificproperties of each class, as well as a way tocombine them, is discussed. Severalimprovements in the field of metaheuristics arealso given. In particular, a method to regulatethe use of classical genetic operators and todefine new more pertinent ones is proposed,taking advantage of a building block structuredrepresentation of the explored space. Ahierarchical approach resting on multiplelevels of cooperative metaheuristics is finallypresented, leading to the definition of acomplete concerted cooperation strategy. Someapplications of these concepts to difficultproteomics problems, including automaticprotein identification, biological motifinference and multiple sequence alignment arepresented. For each application, an innovativemethod based on the cooperation concept isgiven and compared with classical approaches.In the protein identification problem, a firstlevel of cooperation using swarm intelligenceis applied to the comparison of massspectrometric data with biological sequencedatabase, followed by a genetic programmingmethod to discover an optimal scoring function.The multiple sequence alignment problem isdecomposed in three steps involving severalevolutionary processes to infer different kindof biological motifs and a concertedcooperation strategy to build the sequencealignment according to their motif conten
Arc-Heater Facility for Hot Hydrogen Exposure of Nuclear Thermal Rocket Materials
A hyper-thermal environment simulator is described for hot hydrogen exposure of nuclear thermal rocket material specimens and component development. This newly established testing capability uses a high-power, multi-gas, segmented arc-heater to produce high-temperature pressurized hydrogen flows representative of practical reactor core environments and is intended to serve. as a low cost test facility for the purpose of investigating and characterizing candidate fueUstructura1 materials and improving associated processing/fabrication techniques. Design and development efforts are thoroughly summarized, including thermal hydraulics analysis and simulation results, and facility operating characteristics are reported, as determined from a series of baseline performance mapping tests

The Milky Way Tomography with SDSS: III. Stellar Kinematics
We study Milky Way kinematics using a sample of 18.8 million main-sequence
stars with r<20 and proper-motion measurements derived from SDSS and POSS
astrometry, including ~170,000 stars with radial-velocity measurements from the
SDSS spectroscopic survey. Distances to stars are determined using a
photometric parallax relation, covering a distance range from ~100 pc to 10 kpc
over a quarter of the sky at high Galactic latitudes (|b|>20 degrees). We find
that in the region defined by 1 kpc <Z< 5 kpc and 3 kpc <R< 13 kpc, the
rotational velocity for disk stars smoothly decreases, and all three components
of the velocity dispersion increase, with distance from the Galactic plane. In
contrast, the velocity ellipsoid for halo stars is aligned with a spherical
coordinate system and appears to be spatially invariant within the probed
volume. The velocity distribution of nearby ( kpc) K/M stars is complex,
and cannot be described by a standard Schwarzschild ellipsoid. For stars in a
distance-limited subsample of stars (<100 pc), we detect a multimodal velocity
distribution consistent with that seen by HIPPARCOS. This strong
non-Gaussianity significantly affects the measurements of the velocity
ellipsoid tilt and vertex deviation when using the Schwarzschild approximation.
We develop and test a simple descriptive model for the overall kinematic
behavior that captures these features over most of the probed volume, and can
be used to search for substructure in kinematic and metallicity space. We use
this model to predict further improvements in kinematic mapping of the Galaxy
