Meiosis progression and donor age affect expression profile of DNA repair genes in bovine oocytes
Several genetic and physiological factors increase the risk of DNA damage in mammalian oocytes. Two critical events are meiosis progression, from maturation to fertilization, due to the extensive chromatin remodelling during genome decondensation and aging which is associated to a progressive oxidative stress. In this work, we studied the transcriptional patterns of three genes, RAD51, APEX-1 and MLH1, involved in DNA repair mechanisms. The analyses were performed by Real-Time quantitative PCR (RT-qPCR) in immature and in vitro matured oocytes collected from 17 ± 3 mo old heifers and 94 ± 20 mo old cows. Batches of 30-50 oocytes for each group (three replicates) were collected from ovarian follicles of slaughtered animals. The oocytes were freed from cumulus cells at the time of follicle removal, or after IVM carried out in M199 supplemented with 10% foetal calf serum, 10 IU LH /ml, 0.1 IU FSH /ml and 1 µg 17β-oestradiol/ml. Total RNA was extracted by Trizol method. The expression of bovine GAPDH gene was used as internal standard, while primers for bovine RAD51, APEX-1 and MLH1 genes were designed from DNA sequences retrieved from GeneBank. Results obtained indicate a clear up-regulation of RAD51, APEX-1 and MLH1 genes after IVM ranging between 2- and 4-fold compared to GV oocytes. However, only RAD51 showed a significant transcript increase between the immature oocytes collected from young and old individuals. This finding candidates RAD51 as gene marker for discriminating bovine immature oocytes in relation to the donor age
Regulation of mammary gland branching morphogenesis by the extracellular matrix and its remodeling enzymes.
A considerable body of research indicates that mammary gland branching morphogenesis is dependent, in part, on the extracellular matrix (ECM), ECM-receptors, such as integrins and other ECM receptors, and ECM-degrading enzymes, including matrix metalloproteinases (MMPs) and their inhibitors, tissue inhibitors of metalloproteinases (TIMPs). There is some evidence that these ECM cues affect one or more of the following processes: cell survival, polarity, proliferation, differentiation, adhesion, and migration. Both three-dimensional culture models and genetic manipulations of the mouse mammary gland have been used to study the signaling pathways that affect these processes. However, the precise mechanisms of ECM-directed mammary morphogenesis are not well understood. Mammary morphogenesis involves epithelial 'invasion' of adipose tissue, a process akin to invasion by breast cancer cells, although the former is a highly regulated developmental process. How these morphogenic pathways are integrated in the normal gland and how they become dysregulated and subverted in the progression of breast cancer also remain largely unanswered questions
What influences national and foreign physicians’ geographic distribution? An analysis of medical doctors’ residence location in Portugal
Background
The debate over physicians’ geographical distribution has attracted the attention of the
economic and public health literature over the last forty years. Nonetheless, it is still to date
unclear what influences physicians’ location, and whether foreign physicians contribute to fill
the geographical gaps left by national doctors in any given country. The present research sets
out to investigate the current distribution of national and international physicians in Portugal, with the objective to understand its determinants and provide an evidence base for policymakers to identify policies to influence it.
Methods
A cross-sectional study of physicians currently registered in Portugal was conducted to describe the population and explore the association of physician residence patterns with relevant personal and municipality characteristics. Data from the Portuguese Medical Council
on physicians’ residence and characteristics were analysed, as well as data from the National Institute of Statistics on municipalities’ population, living standards and health care network.
Descriptive statistics, chi-square tests, negative binomial and logistic regression modelling were applied to determine: (a) municipality characteristics predicting Portuguese and International physicians’ geographical distribution, and; (b) doctors’ characteristics that could
increase the odds of residing outside the country’s metropolitan areas.
Results
There were 39,473 physicians in Portugal in 2008, 51.1% of whom male, and 40.2% between
41 and 55 years of age. They were predominantly Portuguese (90.5%), with Spanish,
Brazilian and African nationalities also represented. Population, Population’s Purchasing
Power, Nurses per capita and Municipality Development Index (MDI) were the municipality
characteristics displaying the strongest association with national physicians’ location. For
foreign physicians, the MDI was not statistically significant, while municipalities’ foreign
population applying for residence appeared to be an additional positive factor in their location
decisions. In general, being foreigner and male resulted to be the physician characteristics
increasing the odds of residing outside the metropolitan areas. However, among the
internationals, older doctors were more likely to reside outside metropolitan areas. Being
Spanish or Brazilian (but not of African origin) was found to increase the odds of being based
outside the Lisbon and Oporto metropolitan areas.
Conclusions
The present study showed the relevance of studying one country’s physician population to
understand the factors driving national and international doctors’ location decisions. A more
nuanced understanding of national and foreign doctors’ location appears to be needed to
design more effective policies to reduce the imbalance of medical services across

Fine Tuning of Ca(V)1.3 Ca2+ Channel Properties in Adult Inner Hair Cells Positioned in the Most Sensitive Region of the Gerbil Cochlea
Hearing relies on faithful signal transmission by cochlear inner hair cells (IHCs) onto auditory fibres over a wide frequency
and intensity range. Exocytosis at IHC ribbon synapses is triggered by Ca2+
inflow through CaV1.3 (L-type) Ca2+
channels. We
investigated the macroscopic (whole-cell) and elementary (cell-attached) properties of Ca2+
currents in IHCs positioned at
the middle turn (frequency ,2 kHz) of the adult gerbil cochlea, which is their most sensitive hearing region. Using near
physiological recordings conditions (body temperature and a Na+
based extracellular solution), we found that the
macroscopic Ca2+
current activates and deactivates very rapidly (time constant below 1 ms) and inactivates slowly and only
partially. Single-channel recordings showed an elementary conductance of 15 pS, a sub-ms latency to first opening, and a
very low steady-state open probability (Po: 0.024 in response to 500-ms depolarizing steps at ,218 mV). The value of Po
was significantly larger (0.06) in the first 40 ms of membrane depolarization, which corresponds to the time when most Ca2+
channel openings occurred clustered in bursts (mean burst duration: 19 ms). Both the Po and the mean burst duration were
smaller than those previously reported in high-frequency basal IHCs. Finally, we found that middle turn IHCs are likely to
express about 4 times more Ca2+
channels per ribbon than basal cells. We propose that middle-turn IHCs finely-tune CaV1.3
Ca2+
channel gating in order to provide reliable information upon timing and intensity of lower-frequency sounds
Effects of Dietary Restriction on Cancer Development and Progression
The effects of caloric restriction on tumor growth and progression are known for
over a century. Indeed, fasting has been practiced for millennia, but just recently
has emerged the protective role that it may exert toward cells. Fasting cycles are
able to reprogram the cellular metabolism, by inducing protection against oxidative
stress and prolonging cellular longevity. The reduction of calorie intake as
well as short- or long-term fasting has been shown to protect against chronic and
degenerative diseases, such as diabetes, cardiovascular pathologies, and cancer.
In vitro and in vivo preclinical models showed that different restriction dietary
regimens may be effective against cancer onset and progression, by enhancing
therapy response and reducing its toxic side effects. Fasting-mediated beneficial
effects seem to be due to the reduction of inflammatory response and downregulation
of nutrient-related signaling pathways able to modulate cell proliferation
and apoptosis. In this chapter, we will discuss the most significant studies
present in literature regarding the molecular mechanisms by which dietary
restriction may contribute to prevent cancer onset, reduce its progression, and
positively affect the response to the treatments
Ergothioneine Biosynthesis and Functionality in the Opportunistic Fungal Pathogen, Aspergillus fumigatus.
Ergothioneine (EGT; 2-mercaptohistidine trimethylbetaine) is a trimethylated and sulphurised histidine derivative which exhibits antioxidant properties. Here we report that deletion of Aspergillus fumigatus egtA (AFUA_2G15650), which encodes a trimodular enzyme, abrogated EGT biosynthesis in this opportunistic pathogen. EGT biosynthetic deficiency in A. fumigatus significantly reduced resistance to elevated H2O2 and menadione, respectively, impaired gliotoxin production and resulted in attenuated conidiation. Quantitative proteomic analysis revealed substantial proteomic remodelling in ΔegtA compared to wild-type under both basal and ROS conditions, whereby the abundance of 290 proteins was altered. Specifically, the reciprocal differential abundance of cystathionine γ-synthase and β-lyase, respectively, influenced cystathionine availability to effect EGT biosynthesis. A combined deficiency in EGT biosynthesis and the oxidative stress response regulator Yap1, which led to extreme oxidative stress susceptibility, decreased resistance to heavy metals and production of the extracellular siderophore triacetylfusarinine C and increased accumulation of the intracellular siderophore ferricrocin. EGT dissipated H2O2 in vitro, and elevated intracellular GSH levels accompanied abrogation of EGT biosynthesis. EGT deficiency only decreased resistance to high H2O2 levels which suggests functionality as an auxiliary antioxidant, required for growth at elevated oxidative stress conditions. Combined, these data reveal new interactions between cellular redox homeostasis, secondary metabolism and metal ion homeostasis

Impact of Safety-Related Dose Reductions or Discontinuations on Sustained Virologic Response in HCV-Infected Patients: Results from the GUARD-C Cohort.
BACKGROUND: Despite the introduction of direct-acting antiviral agents for chronic hepatitis C virus (HCV) infection, peginterferon alfa/ribavirin remains relevant in many resource-constrained settings. The non-randomized GUARD-C cohort investigated baseline predictors of safety-related dose reductions or discontinuations (sr-RD) and their impact on sustained virologic response (SVR) in patients receiving peginterferon alfa/ribavirin in routine practice. METHODS: A total of 3181 HCV-mono-infected treatment-naive patients were assigned to 24 or 48 weeks of peginterferon alfa/ribavirin by their physician. Patients were categorized by time-to-first sr-RD (Week 4/12). Detailed analyses of the impact of sr-RD on SVR24 (HCV RNA <50 IU/mL) were conducted in 951 Caucasian, noncirrhotic genotype (G)1 patients assigned to peginterferon alfa-2a/ribavirin for 48 weeks. The probability of SVR24 was identified by a baseline scoring system (range: 0-9 points) on which scores of 5 to 9 and <5 represent high and low probability of SVR24, respectively. RESULTS: SVR24 rates were 46.1% (754/1634), 77.1% (279/362), 68.0% (514/756), and 51.3% (203/396), respectively, in G1, 2, 3, and 4 patients. Overall, 16.9% and 21.8% patients experienced ≥1 sr-RD for peginterferon alfa and ribavirin, respectively. Among Caucasian noncirrhotic G1 patients: female sex, lower body mass index, pre-existing cardiovascular/pulmonary disease, and low hematological indices were prognostic factors of sr-RD; SVR24 was lower in patients with ≥1 vs. no sr-RD by Week 4 (37.9% vs. 54.4%; P = 0.0046) and Week 12 (41.7% vs. 55.3%; P = 0.0016); sr-RD by Week 4/12 significantly reduced SVR24 in patients with scores <5 but not ≥5. CONCLUSIONS: In conclusion, sr-RD to peginterferon alfa-2a/ribavirin significantly impacts on SVR24 rates in treatment-naive G1 noncirrhotic Caucasian patients. Baseline characteristics can help select patients with a high probability of SVR24 and a low probability of sr-RD with peginterferon alfa-2a/ribavirin.This study was sponsored by F. Alignment of the ALICE Inner Tracking System with cosmic-ray tracks
37 pages, 15 figures, revised version, accepted by JINSTALICE (A Large Ion Collider Experiment) is the LHC (Large Hadron Collider) experiment devoted to investigating the strongly interacting matter created in nucleus-nucleus collisions at the LHC energies. The ALICE ITS, Inner Tracking System, consists of six cylindrical layers of silicon detectors with three different technologies; in the outward direction: two layers of pixel detectors, two layers each of drift, and strip detectors. The number of parameters to be determined in the spatial alignment of the 2198 sensor modules of the ITS is about 13,000. The target alignment precision is well below 10 micron in some cases (pixels). The sources of alignment information include survey measurements, and the reconstructed tracks from cosmic rays and from proton-proton collisions. The main track-based alignment method uses the Millepede global approach. An iterative local method was developed and used as well. We present the results obtained for the ITS alignment using about 10^5 charged tracks from cosmic rays that have been collected during summer 2008, with the ALICE solenoidal magnet switched off.Peer reviewe
