    Design of new highly functional polymer grafted polyhipes for proteins immobilization

    PolyHIPE have proven to be useful in a large variety of applications included column filtration/separation, supported organic chemistry, as media for tissue engineering and 3D cell culture.1 The ability to conveniently modify pHIPE surfaces with functional groups is essential to opening new applications areas. The most promising method to conveniently modify pHIPE surface with a high density of functional groups is the “grafting from” approach. Stable polymer brushes covalently attached to the surface posses excellent mechanical and chemical robustness and offer the flexibility to introduce a large variety of functional monomers.2 We developed a new and unique pHIPE platform by incorporation of a polymerizable monomer with amino group into the HIPE available for different post in situ polymerization. The pHIPE with amino groups on the surface (pHIPE-NH2) can be directly used for the ring opening polymerization of amino acids N-carboxyanhydrates (NCAs) monomers to make pHIPE-g-polypeptide (such as pHIPE-g-poly(L-Benzyl Glutamate)) or easily converted to an atom transfer radical polymerization (ATRP) initiator for activators generated electron transfer (AGET) ATRP of tert-Butyl acrylate monomers. The polymers grafted can be deprotected to form pHIPE-g-poly(glutamic acid) or pHIPE-g-poly(acrylic acid) with reactive groups, on the surface of the pHIPE, available for further bioconjugation

    Detection and diversity of a putative novel heterogeneous polymorphic proline-glycine repeat (Pgr) protein in the footrot pathogen Dichelobacter nodosus

    Dichelobacter nodosus, a Gram-negative anaerobic bacterium, is the essential causative agent of footrot in sheep. Currently, depending on the clinical presentation in the field, footrot is described as benign or virulent; D. nodosus strains have also been classified as benign or virulent, but this designation is not always consistent with clinical disease. The aim of this study was to determine the diversity of the pgr gene, which encodes a putative proline-glycine repeat protein (Pgr). The pgr gene was present in all 100 isolates of D. nodosus that were examined and, based on sequence analysis had two variants, pgrA and pgrB. In pgrA, there were two coding tandem repeat regions, R1 and R2: different strains had variable numbers of repeats within these regions. The R1 and R2 were absent from pgrB. Both variants were present in strains from Australia, Sweden and the UK, however, only pgrB was detected in isolates from Western Australia. The pgrA gene was detected in D. nodosus from tissue samples from two flocks in the UK with virulent footrot and only pgrB from a flock with no virulent or benign footrot for >10 years. Bioinformatic analysis of the putative PgrA protein indicated that it contained a collagen-like cell surface anchor motif. These results suggest that the pgr gene may be a useful molecular marker for epidemiological studies

    Using Reflection to Promote Success in Community College Mathematics

    Community college mathematics faculty have instituted changes to address inadequate preparation of incoming students to learn college-level mathematics. However, they could do more to develop student cognitive capabilities to enable underprepared students to successfully learn college mathematics. This basic qualitative study examined the perceptions of community college mathematics instructors who made their classrooms more reflective, including using reflection as an instruction activity for students entering college underprepared to learn college-level mathematics. The conceptual framework consisted of Baxter Magolda’s epistemological reflection model of college student development and Schoenfeld’s theoretical model for teaching mathematics. Open coding of interviews with 10 community college mathematics instructors in the United States suggested instructors agreed that improvement in teaching mathematics is needed. They reported that reflection fostered development of epistemological and cognitive abilities of underprepared students who were newly successful in learning mathematics. Instructors perceived that reflection activities improved student learning outcomes, with reflection first starting individually and then shared by explaining solutions to the instructor and other students, leading to a growing confidence. In addition, instructors noticed that reflection improved their own teaching practice over time. This study may contribute to positive social change by helping community college faculty and leaders better understand how to improve learning outcomes for underprepared students to become newly successful in learning mathematics and by doing so, remain enrolled in community college and complete their degrees

    Polypeptide-grafted macroporous polyHIPE by surface-initiated N-Carboxyanhydride (NCA) polymerization as a platform for bioconjugation

    A new class of functional macroporous monoliths from polymerized high internal phase emulsion (polyHIPE) with tunable surface functional groups was developed by direct polypeptide surface grafting. In the first step, amino-functional polyHIPEs were obtained by the addition of 4-vinylbenzyl or 4-vinylbenzylphthalimide to the styrenic emulsion and thermal radical polymerization. The obtained monoliths present the expected open-cell morphology and a high surface area. The incorporated amino group was successfully utilized to initiate the ring-opening polymer- ization of benzyl-L-glutamate N-carboxyanhydride (BLG NCA) and benzyloxycarbonyl-L-lysine (Lys(Z)) NCA, which resulted in a dense homogeneous coating of polypeptides throughout the internal polyHIPE surfaces as confirmed by SEM and FTIR analysis. The amount of polypeptide grafted to the polyHIPE surfaces could be modulated by varying the initial ratio of amino acid NCA to amino-functional polyHIPE. Subsequent removal of the polypeptide protecting groups yielded highly functional polyHIPE-g-poly(glutamic acid) and polyHIPE-g- poly(lysine). Both types of polypeptide-grafted monoliths responded to pH by changes in their hydrohilicity. The possibility to use the high density of function (−COOH or −NH2) for secondary reaction was demonstrated by the successful bioconjugation of enhanced green fluorescent protein (eGFP) and fluorescein isocyanate (FITC) on the polymer 3D-scaffold surface. The amount of eGFP and FITC conjugated to the polypeptide-grafted polyHIPE was significantly higher than to the amino- functional polyHIPE, signifying the advantage of polypeptide grafting to achieve highly functional polyHIPEs

Для качественного развития массового въездного и внутреннего туризма в регионах России, в частности в городе Новосибирске, необходимо уделить отдельное внимание экскурсионному обслуживанию. Именно обзорные экскурсии создают первое впечатление о городе у его гостей. От экскурсионной деятельности во многом зависит как воспримут туристы посещаемые места, захотят ли побывать в них еще раз. Поэтому целью моей работы является разработка пакета экскурсионных маршрутов по принципу Hop-on Hop-off для туристов по городу Новосибирску.The last decade in the Russian Federation has increased the interest of the government in the integrated development of tourism: both in the whole state and in certain regions. For the qualitative development of mass inbound and domestic tourism in the regions of Russia, in particular in the city of Novosibirsk, it is necessary to pay special attention to excursion services. It is the sightseeing tours that create the first impression of the city at its guests. From the excursion activity in many respects depends on how the tourists will perceive the places visited, whether they will want to visit them again. Therefore the purpose of my work is elaboration of a package of sightseeing routes on the principle of Hop-on Hop-off for tourists around the city of Novosibirsk
