However, there is little information on the pattern of CD4 (Th1) and CD8 (Tc1) T-cell response to AFP in patients with HCC and their association with the clinical characteristics of patients.
Methods: We therefore analysed CD4 and CD8 T-cell responses to a panel of AFP-derived peptides in a total of 31 HCC patients and 14 controls, using an intracellular cytokine assay for IFN-γ.
Results: Anti-AFP Tc1 responses were detected in 28.5% of controls, as well as in 25% of HCC patients with Okuda I (early tumour stage) and in 31.6% of HCC patients with stage II or III (late tumour stages). An anti-AFP Th1 response was detected only in HCC patients (58.3% with Okuda stage I tumours and 15.8% with Okuda stage II or III tumours). Anti-AFP Th1 response was mainly detected in HCC patients who had normal or mildly elevated serum AFP concentrations (P=0.00188), whereas there was no significant difference between serum AFP concentrations in these patients and the presence of an anti-AFP Tc1 response. A Th1 response was detected in 44% of HCC patients with a Child–Pugh A score (early stage of cirrhosis), whereas this was detected in only 15% with a B or C score (late-stage cirrhosis). In contrast, a Tc1 response was detected in 17% of HCC patients with a Child–Pugh A score and in 46% with a B or C score.
Conclusion: These results suggest that anti-AFP Th1 responses are more likely to be present in patients who are in an early stage of disease (for both tumour stage and liver cirrhosis), whereas anti-AFP Tc1 responses are more likely to be present in patients with late-stage liver cirrhosis. Qtls Controlling Seed Weight and Days to Flowering in Mungbean [Vigna Radiata (L.) Wilczek], Their Conservation in Azuki Bean [V. Angularis (Ohwi) Ohwi & Ohashi] and Rice Bean [V. Umbellata (Thunb.) Ohwi & Ohashi]
Mungbean (Vigna radiata (L.) Wilczek) is a socio-economically important legume crop of Asia. Varieties with large seed size and early maturity are preferred in commercial production. In this study, we identified quantitative trait loci (QTL) controlling seed weight and days to flowering in mungbean. The mapping population comprises 155 F2-derived lines from a cross between Kamphaeng Saen 1 (large-seeded and early flowering) and V4718 (small-seeded and late flowering). The F2 population was analyzed with 67 simple sequence repeat markers. The F2:3 families were evaluated for 100-seed weigh and days to flowering in two years, 2008 (one season) and 2011 (two seasons). Composite interval mapping identified six QTLs for 100-seed weight and 5 QTLs for days to flowering. Three genomic regions harbored QTLs for both seed weight and days to flowering, revealing association between the two traits. Comparison of QTLs for both traits found in this study with those reported in azuki bean (Vigna angularis (Willd.) Ohwi & Ohashi) and rice bean (Vigna umbellata (Thunb.) Ohwi & Ohashi) revealed that several QTLs are conserved among the three Vigna species
Genome-Wide Association with Diabetes-Related Traits in the Framingham Heart Study
BACKGROUND: Susceptibility to type 2 diabetes may be conferred by genetic variants having modest effects on risk. Genome-wide fixed marker arrays offer a novel approach to detect these variants. METHODS: We used the Affymetrix 100K SNP array in 1,087 Framingham Offspring Study family members to examine genetic associations with three diabetes-related quantitative glucose traits (fasting plasma glucose (FPG), hemoglobin A1c, 28-yr time-averaged FPG (tFPG)), three insulin traits (fasting insulin, HOMA-insulin resistance, and 0–120 min insulin sensitivity index); and with risk for diabetes. We used additive generalized estimating equations (GEE) and family-based association test (FBAT) models to test associations of SNP genotypes with sex-age-age2-adjusted residual trait values, and Cox survival models to test incident diabetes. RESULTS: We found 415 SNPs associated (at p 1%) 100K SNPs in LD (r2 > 0.05) with ABCC8 A1369S (rs757110), KCNJ11 E23K (rs5219), or SNPs in CAPN10 or HNFa. PPARG P12A (rs1801282) was not significantly associated with diabetes or related traits. CONCLUSION: Framingham 100K SNP data is a resource for association tests of known and novel genes with diabetes and related traits posted at. PENGARUH DISIPLIN KERJA DAN MOTIVASI TERHADAP KINERJA PEGAWAI DI DIVISI PENCATAT METER PERUSAHAAN DAERAH AIR MINUM (PDAM) TIRTAWENING KOTA BANDUNG
ABSTRAK
Penelitian ini diawali dengan adanya temuan indikasi masalah yang terdapat
pada kinerja pegawai di Divisi Pencatat Meter Perusahaan Daerah Air Minum
(PDAM) Tirtawwening Kota Bandung yang masih di bawah rata-rata. Masalah
kinerja pegawai ini dapat dilihat dari disiplin kerja pegawai yang masih rendah
dan masih rendahnya motivasi yang dimiliki pegawai.
Penelitian ini bertujuan untuk mengetahui seberapa besar pengaruh disiplin
kerja dan motivasi terhadap kinerja pegawai di Divisi Pencatat Meter Perusahaan
Daerah Air Minum (PDAM) Tirtawening Kota Bandung. Metode yang digunakan
dalam penelitian ini adalah metode deskriptif dan verifikatif. Adapun populasi
dalam penelitian ini yaitu sebanyak 95 orang pegawai, dengan menggunakan
penelitian sensus dimana semua anggota di jadikan responden. Teknik
pengumpulan data yang dilakukan oleh peneliti adalah observasi, wawancara dan
penyebaran kuesioner. Alat analisis data adalah analisis regresi linier berganda,
analisis korelasi berganda dan analisis koefisien determinasi.
Penelitian ini menghasilkan pengaruh disiplin kerja dan motivasi terhadap
kinerja pegawai kuat dan positif yaitu sebesar 0,765 atau 76,5%. Kontribusi
variabel disiplin kerja terhadap variabel kinerja pegawai sebesar 30,4% dan
kontribusi variabel motivasi terhadap variabel kinerja pegawai sebesar 46,1%.

Immunization expands B cells specific to HIV-1 V3 glycan in mice and macaques.
Broadly neutralizing monoclonal antibodies protect against infection with HIV-1 in animal models, suggesting that a vaccine that elicits these antibodies would be protective in humans. However, it has not yet been possible to induce adequate serological responses by vaccination. Here, to activate B cells that express precursors of broadly neutralizing antibodies within polyclonal repertoires, we developed an immunogen, RC1, that facilitates the recognition of the variable loop 3 (V3)-glycan patch on the envelope protein of HIV-1. RC1 conceals non-conserved immunodominant regions by the addition of glycans and/or multimerization on virus-like particles. Immunization of mice, rabbits and rhesus macaques with RC1 elicited serological responses that targeted the V3-glycan patch. Antibody cloning and cryo-electron microscopy structures of antibody-envelope complexes confirmed that immunization with RC1 expands clones of B cells that carry the anti-V3-glycan patch antibodies, which resemble precursors of human broadly neutralizing antibodies. Thus, RC1 may be a suitable priming immunogen for sequential vaccination strategies in the context of polyclonal repertoires
Inhibitor of DNA binding 1 as a secreted angiogenic transcription factor in rheumatoid arthritis
Abstract
Introduction
Rheumatoid arthritis (RA) is characterized by enhanced blood vessel development in joint synovium. This involves the recruitment of endothelial progenitor cells (EPCs), allowing for de novo vessel formation and pro-inflammatory cell infiltration. Inhibitor of DNA Binding 1 (Id1) is a transcription factor characteristic of EPCs that influences cell maturation.
Method
Enzyme-linked immunosorbant assay (ELISA) and polymerase chain reaction (PCR) were used to examine Id1 levels in synovial fluid (SF) and endothelial cells (ECs), respectively. Immunohistology was used to determine the expression of Id1 in synovial tissue (ST). Human dermal microvascular EC (HMVEC) migration and tube forming assays were used to determine if recombinant human Id1 (rhuId1) and/or RA SF immunodepleted Id1 showed angiogenic activity. We also utilized the RA ST severe combined immunodeficient (SCID) mouse chimera to examine if Id1 recruits EPCs to RA synovium.
Results
ST samples immunostained for Id1 showed heightened expression in RA compared to osteoarthritis (OA) and normal (NL) ST. By immunofluorescence staining, we found significantly more Id1 in RA compared to OA and NL vasculature, showing that Id1 expressing cells, and therefore EPCs, are most active in vascular remodeling in the RA synovium. We also detected significantly more Id1 in RA compared to OA and other arthritis SFs by ELISA, which correlates highly with Chemokine (C-X-C motif) ligand 16 (CXCL16) levels. In vitro chemotaxis assays showed that Id1 is highly chemotactic for HMVECs and can be attenuated by inhibition of Nuclear Factor κB and phosphoinositide 3-kinase. Using in vitro Matrigel assays, we found that HMVECs form tubes in response to rhuId1 and that Id1 immunodepleted from RA SF profoundly decreases tube formation in Matrigel in vitro. PCR showed that Id1 mRNA could be up-regulated in EPCs compared to HMVECs in response to CXCL16. Finally, using the K/BxN serum induced arthritis model, we found that EC CXCR6 correlated with Id1 expression by immunohistochemistry.
Conclusions
We conclude that Id1 correlates highly with CXCL16 expression, EPC recruitment, and blood vessel formation in the RA joint, and that Id1 is potently angiogenic and can be up-regulated in EPCs by CXCL16.http://deepblue.lib.umich.edu/bitstream/2027.42/110805/1/13075_2013_Article_4258.pd
