    Integrated use of residues from olive mill and winery for lipase production by solid state fermentation with Aspergillus sp

    Two phase olive mill waste (TPOMW) is presently the major waste produced by the olive mill industry. This waste has potential to be used as substrate for solid state fermentation (SSF) despite of its high concentration of phenolic compounds and low nitrogen content. In this work, it is demonstrated that mixtures of TPOMW with winery wastes support the production of lipase by Aspergillus spp. By agar plate screening, Aspergillus niger MUM 03.58, Aspergillus ibericus MUM 03.49 and Aspergillus uvarum MUM 08.01 were chosen for lipase production by SSF. Plackett-Burman experimental design was employed to evaluate the effect of substrate composition and time on lipase production. The highest amounts of lipase were produced by A. ibericus on a mixture of TPOMW, urea and exhausted grape mark (EGM). Urea was found to be the most influent factor for the lipase production. Further optimization of lipase production by A. ibericus using a full factorial design (32) conducted to optimal conditions of substrate composition (0.073 g urea/g and 25% of EGM) achieving 18.67 U/g of lipolytic activity.Jose Manuel Salgado is grateful for Postdoctoral fellowship (EX-2010-0402) of Education Ministry of Spanish Government.     Synthesis, crystal structure and spectroscopic characterization of a new organic bismuthate (III) [C9H28N4][Bi2Cl10].H2O

    The chemical preparation, crystal structure and spectroscopic characterization of [C9H28N4][Bi2Cl10].H2O have been reported. This compound crystallizes in the monoclinic system in space group P21/c and cell parameters a = 12.2385 (6), b = 17.3062 (7), c = 13.0772 (6) Å, β = 104.475 (5)°, Z = 4 and V = 2681.9 (2) Å3. Its crystal structure has been determined and refined to R = 0.049, using 5848 independent reflections. The atomic arrangement can be described by an alternation of organic and inorganic layers. The inorganic layer built up of [Bi2Cl10]4– bioctahedra arranged in sandwich between the organic layer. The organic groups are interconnected by the water molecules through N-H…O(W) hydrogen bonds to form infinite zig-zag chains spreading along the b-axis. These Chains are themselves interconnected by means of the N–H…Cl hydrogen bonds originating from [Bi2Cl10]4– anions, to form a three-dimensional network. Intermolecular Cl…Cl interactions between adjacent dimeric [Bi2Cl10]4– anions have been observed. The compound was also characterized by FT-IR and Raman spectrscopies

    Sodium lauryl ether sulfate (SLES) degradation by nitrate-reducing bacteria

    The online version of this article (doi:10.1007/s00253-017-8212-x) contains supplementary material, which is available to authorized users.The surfactant sodium lauryl ether sulfate (SLES) is widely used in the composition of detergents and frequently ends up in wastewater treatment plants (WWTPs). While aerobic SLES degradation is well studied, little is known about the fate of this compound in anoxic environments, such as denitrification tanks of WWTPs, nor about the bacteria involved in the anoxic biodegradation. Here, we used SLES as sole carbon and energy source, at concentrations ranging from 50 to 1000 mg L1, to enrich and isolate nitrate-reducing bacteria from activated sludge of a WWTP with the anaerobic-anoxic-oxic (A2/O) concept. In the 50 mg L1 enrichment, Comamonas (50%), Pseudomonas (24%), and Alicycliphilus (12%) were present at higher relative abundance, while Pseudomonas (53%) became dominant in the 1000 mg L1 enrichment. Aeromonas hydrophila strain S7, Pseudomonas stutzeri strain S8, and Pseudomonas nitroreducens strain S11 were isolated from the enriched cultures. Under denitrifying conditions, strains S8 and S11 degraded 500 mg L1 SLES in less than 1 day, while strain S7 required more than 6 days. Strains S8 and S11 also showed a remarkable resistance to SLES, being able to grow and reduce nitrate with SLES concentrations up to 40 g L1. Strain S11 turned out to be the best anoxic SLES degrader, degrading up to 41% of 500 mg L1. The comparison between SLES anoxic and oxic degradation by strain S11 revealed differences in SLES cleavage, degradation, and sulfate accumulation; both ester and ether cleavage were probably employed in SLES anoxic degradation by strain S11.This research was supported by the Spanish Ministry of Education and Science (contract project CTQ2007-64324 and 447 CONSOLIDER-CSD 2007-00055).     Experimental Analysis on the Counter-Current Dumitrescu- Taylor Bubble Flow in a Smooth Vertical Conduct of Small Diameter

    In this paper, an experimental investigation on the Dumitrescu-Taylor bubble in counter-current laminar downward flow in vertical pipe of a small internal diameter pipe is presented. The experimental design is realized to work for low and stable liquid flow rates. The Dumitrescu-Taylor bubble may be stationary or can be in motion with an ascending or descending velocity, and this displacement depends on the downward liquid flow rates. Consequently, the advantage of this device is to carry out the measurements of the velocities inside the gas Dumitrescu-Taylor bubble by Laser Doppler Velocimetry (LDV). Starting from the visual observations and image acquisitions with a fast camera, a qualitative description was brought on the hydrodynamic behavior of the liquid film and the ripples created at the bottom of the Dumitrescu-Taylor bubble. The experimental results show a presence of a long toroïdal vortex inside the gas bubble. It should be noted that previous work using a hot wire does not show the existence of this vortex. Additionally, other hydrodynamic magnitudes were measured as the liquid film thickness, the Dumitrescu-Taylor bubble rising velocity as well as the erosion bubble.     Reorganization of Coherent Structures Downstream a Circular Cylinder Located between Two Parallel Walls

    Experiments were performed at low Reynolds numbers in the range 75 Re 275 in the wake of a circular cylinder of dc diameter placed symmetrically between two parallel walls of H height. 2D2C particle image velocimetry (PIV) was used to investigate the flow downstream the cylinder. In the unsteady flow regime downstream the cylinder, the detached primary vortices (Pi) interact with walls generating secondary ones (Pi’) and modify the cylinder wake dynamic. The kinematical properties (advection velocity, circulation, rotation kinetic energy, etc.) of the generated secondary vortices are studied and compared with the primary ones in order to show how the walls influence the von Kármán vortex street. The authors propose here a relation between the circulations and kinetic energies of primary and secondary vortices

    In this paper we studied a beta type Stirling machine. At first, we present the adopted theoretical quasi-stationary model. Then, we pass to the physical and geometrical presentation of this machine. The Latter was experimented according to two configurations: motor configuration and receiver configuration. For the first configuration, in order to improve the performances of the machine, we proceeded to the insulation of the machine hot room to reduce losses by radiation. For the second configuration, the machine is experimented as a heat pump and refrigerator. Comparisons between the theoretical and experimental results are also presented. We finally validated the results obtained by the model with experiments

    Screening of winery and olive mill wastes for lignocellulolytic enzyme production from Aspergillus species by solid-state fermentation

    Wastes from olive oil and wine industries (as exhausted grape marc, vineshoot trimmings, two-phase olive mill waste, vinasses, and olive mill wastewater) were evaluated for lignocellulolytic enzyme production (as endocellulases, endoxylanases, and feruloyl esterases) by solid-state fermentation (SSF) with Aspergillus niger, Aspergillus ibericus, and Aspergillus uvarum. To study the effect of different solid medium composition and time in enzyme production, a PlackettBurman experimental design was used. Variables that had a higher positive effect in lignocellulolytic enzyme production were urea, time, and exhausted grape marc. The maximum values of enzymatic activity per unit of substrate dry mass were found with A. niger for feruloyl esterase. Enzymatic extracts from SSF with A. niger achieved maximum feruloyl esterase activity (89.53 U/g) and endoxylanase activity (3.06 U/g) and with A. uvarum for endocellulase activity (6.77 U/g).
