    Merkel Cells as Putative Regulatory Cells in Skin Disorders: An In Vitro Study

    Merkel cells (MCs) are involved in mechanoreception, but several lines of evidence suggest that they may also participate in skin disorders through the release of neuropeptides and hormones. In addition, MC hyperplasias have been reported in inflammatory skin diseases. However, neither proliferation nor reactions to the epidermal environment have been demonstrated. We established a culture model enriched in swine MCs to analyze their proliferative capability and to discover MC survival factors and modulators of MC neuroendocrine properties. In culture, MCs reacted to bFGF by extending outgrowths. Conversely, neurotrophins failed to induce cell spreading, suggesting that they do not act as a growth factor for MCs. For the first time, we provide evidence of proliferation in culture through Ki-67 immunoreactivity. We also found that MCs reacted to histamine or activation of the proton gated/osmoreceptor TRPV4 by releasing vasoactive intestinal peptide (VIP). Since VIP is involved in many pathophysiological processes, its release suggests a putative regulatory role for MCs in skin disorders. Moreover, in contrast to mechanotransduction, neuropeptide exocytosis was Ca2+-independent, as inhibition of Ca2+ channels or culture in the absence of Ca2+ failed to decrease the amount of VIP released. We conclude that neuropeptide release and neurotransmitter exocytosis may be two distinct pathways that are differentially regulated

    Collagen-producing lung cell atlas identifies multiple subsets with distinct localization and relevance to fibrosis

    Collagen-producing cells maintain the complex architecture of the lung and drive pathologic scarring in pulmonary fibrosis. Here we perform single-cell RNA-sequencing to identify all collagen-producing cells in normal and fibrotic lungs. We characterize multiple collagen-producing subpopulations with distinct anatomical localizations in different compartments of murine lungs. One subpopulation, characterized by expression of Cthrc1 (collagen triple helix repeat containing 1), emerges in fibrotic lungs and expresses the highest levels of collagens. Single-cell RNA-sequencing of human lungs, including those from idiopathic pulmonary fibrosis and scleroderma patients, demonstrate similar heterogeneity and CTHRC1-expressing fibroblasts present uniquely in fibrotic lungs. Immunostaining and in situ hybridization show that these cells are concentrated within fibroblastic foci. We purify collagen-producing subpopulations and find disease-relevant phenotypes of Cthrc1-expressing fibroblasts in in vitro and adoptive transfer experiments. Our atlas of collagen-producing cells provides a roadmap for studying the roles of these unique populations in homeostasis and pathologic fibrosis

    Prospect for HEW Applications of Multivariate Statistical Analysis in Microscopy and Spectroscopy

    Multivariate Statistical Analysis has been developed in order to process large data sets and to extract from them the significant information.It has been introduced in electron microscopy in order to process image series of macromolecules and to classify individual images into several subsets of the series. This kind of series can be considered as a spatial series.In fact, electron microscopy and spectroscopy are using more and more image sequences in order to be able to access more precisely the content of the object studied. Among the different types of sequences which are already used and which will be used more extensively in the near future, one can mention : —spatial series (several slightly different objects, whose images are to be classified and combined ; or several microanalytical spectra recorded at different places of the object).—time series : in order to study changes induced in the object, one has to record successive images or spectra.—series in energy : one possibility to study the behavior of a specimen is to vary the accelerating voltage, to record several images or spectra and process the whole data set. Another exemple of series in energy is given by energy loss elemental mapping (or Auger elemental mapping) where several energy filtered images must be recorded (below and above the edge of interest) in order to get a “true” elemental map.—more “exotic” series can also be obtained, for instance series obtained by varying the spot size in MEB, series obtained with different configured detectors in STEM...</jats:p

    Effect of rhizobial inoculation on growth of Calliandra tree species under nursery conditions

    A nursery experiment was conducted in un-sterilized soil in Senegal using six Calliandra species or provenances inoculated with a mixture of seven rhizobial strains. Plant growth was assessed periodically at 1, 2, 3, 12 and 18 months whereas nodulation, shoot and root dry weights were assessed at 12 and 18 months after planting (MAP). Un-inoculated seedlings of all the six species died at 12 months after planting. Results of growth assessments were variable with significant differences (P < 0.05) between C. calothyrsus Meissn provenances (Flores and San Ramón) and C. juzepczukii Standley in height at 1, 2 and 3 MAP. Shoot dry weights of San Ramón provenance of C. calothyrsus Meissn were also significantly different from those of C. acapulcensis (Britton and Rose) Standley and C. longepedicellata (Mc Vaugh) H. Hern and Macqueen at 18 MAP. In contrast, C. glandiflora (L’Her.) Benth grew poorly and did not nodulate. Although rhizobial inoculation improved shoot and root dry weights some differences were observed among the Calliandra species in response to the inoculation, which suggested the occurrence of interaction between the rhizobial strains and the host plant species tested
