    Nano Ag@Bioactive Microspheres From Marine Sponge Clathria Frondifera: Fabrication, Fortification, Characterization and Antibacterial Potential Evaluation

    Abstract Bioresources are attaining much importance in the discovery of drugs and delivering agents. In particular, marine sponges are of great interest due to their metabolites production for the survival in risky environment. The incorporation of silver nanoparticles with marine sponge derived metabolites was reported for the first time. In this work, a facile material has been generated of great efficacy in solving environmental and health issues, as a recipe of silver and marine sponge Clathria frondifera, named as Ag Fortified Sponge spheres (AFS). AFS spheres were successfully synthesized after method optimization, using the various extracts of marine sponge Clathria frondifera as effective reducing agent in Ag (I) to Ag (0) reduction. Bioactive material from marine sponge and AgNP from the reduction of AgNO3 solution stablishing one another and thus AFS spheres were attaining long lifetime along with enhanced antimicrobial activity. The characterization of synthesized AFS and other AgNPs (1-4) has done using FT-IR, PXRD, FESEM, TEM, UV-vis and PL data. The cytotoxic response of AFS was assessed by 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) and morphological changes. AFS are exact spherical, micro sized and effective in inhibiting the growth of both gram positive and gram negative bacteria. Anticancer studies were also carried out and ensued with excellent activity in the HELA cells.</jats:p

    A pH-sensitive guar gum-: Grafted -lysine-?-cyclodextrin drug carrier for the controlled release of 5-flourouracil into cancer cells

    The physiological environment is a crucial factor in biomedical systems, which can be regulated with relative ease both in vitro and in vivo. Control of pH has emerged as a powerful strategy in cancer therapy because pH has a profound effect on the interaction of a polymeric-based drug delivery system with tumor cells. In this regard, the enhancement of pH response capability was demonstrated with a 5-flourouracil (5-FU)-loaded guar gum (GG)-grafted-lysine-?-cyclodextrin (L-?-CD) drug carrier. The size, charge, morphology, and encapsulation efficiency of the 5-FU-loaded polymeric carrier were characterized for the control and sustained release of the drug. In a specific cancer pH environment, 5-FU-loaded GG-g-L-?-CD could rapidly swallow, disassemble, and release 5-FU by accepting or losing electrons. In vitro observations indicate that GG-g-L-?-CD dramatically releases 5-FU in an acidic environment rather than in a basic environment. In vitro antitumor activity tests showed that 5-FU-loaded GG-g-L-?-CD had a higher cytotoxicity against KB cells, with an IC50 value of 1.38 ?g ml-1. The reactive oxygen species (ROS) generated by 5-FU in the KB cells showed efficient suppression of tumour cell growth. The Hoechst assay revealed the active nature of 5-FU in the cell nucleus of the KB cells. The potential mitochondrial damage by apoptosis in KB cells greatly increased cell death. Therefore, due to its active nature, the pH-sensitive 5-FU-loaded GG-g-L-?-CD carrier is a potential drug delivery system for safe and effective cancer therapy, and it can be useful for inhibiting tumour cell growth.MR acknowledges the major financial support received from the University Grants Commission (UGC), Government of India, for financial support under the plan of ‘‘UGC-MRP’’ (F.     Fabrication of Progesterone-Loaded Nanofibers for the Drug Delivery Applications in Bovine

    Progesterone is a potent drug for synchronization of the estrus and ovulation cycles in bovine. At present, the estrus cycle of bovine is controlled by the insertion of progesterone-embedded silicone bands. The disadvantage of nondegradable polymer inserts is to require for disposal of these bands after their use. The study currently focuses on preparation of biodegradable progesterone-incorporated nanofiber for estrus synchronization. Three different concentrations (1.2, 1.9, and 2.5 g) of progesterone-impregnated nanofibers were fabricated using electrospinning. The spun membrane were characterized by scanning electron microscopy, X-ray diffraction, differential scanning calorimetry, thermogravimetric analysis, and Fourier transform infrared spectroscopy. Uniform surface morphology, narrow size distribution, and interaction between progesterone and zein were confirmed by SEM. FTIR spectroscopy indicated miscibility and interaction between zein and progesterone. X-ray analysis indicated that the size of zein crystallites increased with progesterone content in nanofibers. Significant differences in thermal behavior of progesterone-impregnated nanofiber were observed by DSC. Cell viability studies of progesterone-loaded nanofiber were examined using MTT assay. In vitro release experiment is to identify the suitable progesterone concentration for estrus synchronization. This study confirms that progesterone-impregnated nanofibers are an ideal vehicle for progesterone delivery for estrus synchronization of bovines
