    Wt1 is required for cardiovascular progenitor cell formation through transcriptional control of Snail and E-cadherin

    Epicardial epithelial-mesenchymal transition (EMT) is hypothesized to generate cardiovascular progenitor cells that differentiate into various cell types, including coronary smooth muscle and endothelial cells, perivascular and cardiac interstitial fibroblasts and cardiomyocytes. Here we show that an epicardial-specific knockout of Wt1 leads to a reduction of mesenchymal progenitor cells and their derivatives. We demonstrate that Wt1 is essential for repression of the epithelial phenotype in epicardial cells and during Embryonic Stem (ES) cell differentiation, through direct transcriptional regulation of Snail (Snai1) and E-cadherin (Cdh1), two of the major mediators of EMT. Some mesodermal lineages fail to form in Wt1 null embryoid bodies but this effect is rescued by the expression of Snai1, underlining the importance of EMT in generating these differentiated cells. These new insights into the molecular mechanisms regulating cardiovascular progenitor cells and EMT will shed light on the pathogenesis of heart diseases and may help the development of cell based therapies

    Incidence, prevalence and mortality rates of malaria in Ethiopia from 1990 to 2015: analysis of the global burden of diseases 2015

    Background: In Ethiopia there is no complete registration system to measure disease burden and risk factors accurately. In this study, the 2015 Global Burden of Diseases, Injuries and Risk factors (GBD) data were used to analyse the incidence, prevalence and mortality rates of malaria in Ethiopia over the last 25 years. Methods: GBD 2015 used verbal autopsy (VA) surveys, reports, and published scientific articles to estimate the burden of malaria in Ethiopia. Age and gender-specific causes of death for malaria were estimated using Cause of Death Ensemble Modelling (CODEm). Results: The number of new cases of malaria declined from 2.8 million (95% uncertainty interval (UI): 1.4-4.5million) in 1990 to 621,345 (95% UI: 462,230-797,442) in 2015. Malaria caused an estimated 30,323.9 deaths (95% UI: 11,533.3-61,215.3) in 1990 and 1,561.7 deaths (95% UI: 752.8-2,660.5) in 2015, a 94.8% reduction over the 25 years. Age-standardized mortality rate of malaria has declined by 96.5% between 1990 and 2015 with an annual rate of change (ARC) of 13.4%. Age-standardized malaria incidence rate among all ages and gender declined by 88.7% between 1990 and 2015. The number of disability-adjusted life years lost (DALY) due to malaria decreased from 2.2 million (95% UI: 0.76-4.7 million) in 1990 to 0.18 million (95% UI: 0.12-0.26 million) in 2015, with a total reduction 91.7%. Similarly, age-standardized DALY rate declined by 94.8% during the same period. Conclusions: Ethiopia has achieved a 50% reduction target of malaria of the Millennium Development Goals (MDGs). The country should strengthen its malaria control and treatment strategies to achieve the Sustainable Development Goals (SDG)

    Food Insecurity, Food Assistance, and Psychological Distress among University Students: Cross-Sectional Survey Western Australia, 2020

    University students have been identified as a population sub-group vulnerable to food insecurity. This vulnerability increased in 2020 due to the COVID-19 pandemic. This study aimed to assess factors associated with food insecurity among university students and the differences between students with and without children. A cross-sectional survey of (n = 213) students attending one university in Western Australia measured food insecurity, psychological distress, and socio-demographic characteristics. Logistic regression analyses were conducted to identify factors associated with food insecurity. Forty-eight percent of students who responded to the survey had experienced food insecurity in 2020. International students who were studying in Australia were nine times more likely to experience food insecurity than domestic students (AOR = 9.13; 95% CI = 2.32–35.97). International students with children were more likely to experience food insecurity than international students without children (p < 0.001) and domestic students with (p < 0.001) or without children (p < 0.001). For each unit increase in depression level, the likelihood of experiencing food insecurity increased (AOR = 1.62; 95% CI = 1.12–2.33). Findings show a higher prevalence of food insecurity among international university students and students with children during the COVID-19 pandemic and that food insecurity was associated with higher levels of psychological distress. These findings highlight the need for targeted interventions to mitigate the risk of food insecurity among Australian university students, particularly among international students, students with children, and those experiencing psychological distress

    De-identification of primary care electronic medical records free-text data in Ontario, Canada

    <p>Abstract</p> <p>Background</p> <p>Electronic medical records (EMRs) represent a potentially rich source of health information for research but the free-text in EMRs often contains identifying information. While de-identification tools have been developed for free-text, none have been developed or tested for the full range of primary care EMR data</p> <p>Methods</p> <p>We used <it>deid </it>open source de-identification software and modified it for an Ontario context for use on primary care EMR data. We developed the modified program on a training set of 1000 free-text records from one group practice and then tested it on two validation sets from a random sample of 700 free-text EMR records from 17 different physicians from 7 different practices in 5 different cities and 500 free-text records from a group practice that was in a different city than the group practice that was used for the training set. We measured the sensitivity/recall, precision, specificity, accuracy and F-measure of the modified tool against manually tagged free-text records to remove patient and physician names, locations, addresses, medical record, health card and telephone numbers.</p> <p>Results</p> <p>We found that the modified training program performed with a sensitivity of 88.3%, specificity of 91.4%, precision of 91.3%, accuracy of 89.9% and F-measure of 0.90. The validations sets had sensitivities of 86.7% and 80.2%, specificities of 91.4% and 87.7%, precisions of 91.1% and 87.4%, accuracies of 89.0% and 83.8% and F-measures of 0.89 and 0.84 for the first and second validation sets respectively.</p> <p>Conclusion</p> <p>The <it>deid </it>program can be modified to reasonably accurately de-identify free-text primary care EMR records while preserving clinical content.</p

    Adoption Studies on Improved Chickpea Varieties in Ethiopia

    Chickpea (Cicer arietinum L.) is one of the most important food legumes in Ethiopia contributing to about 17% of the countries’ total pulse production. Ethiopia is the largest chickpea growing country in Africa, with a share of about 37% in area and 48% in production. During 2003/2004, Ethiopia produced 135,930 m t of chickpea from an area of 168,089 ha. There has been an increase of 12% in area and 34% in production since 1981/1982. Most of the chickpea production goes for domestic consumption. However, there has been substantial export of chickpea during the past five years, with maximum of 48,549 t (valued at US$14.7 million) during 2002 (FAOSTAT 2005). Chickpea is an important source of dietary protein and minerals for many Ethiopians who cannot afford animal products. It is used in various forms, e.g., green seeds, dried seeds, dehulled-splits and flour. Chickpea straw is highly valued as animal feed. The farmers recognize the importance of legumes in improving soil fertility and thus grow chickpea and other legumes in rotation with cereals. The Debre Zeit Agricultural Research Center (DZARC) has been the premier institute for chickpea research in Ethiopia. It has collaborated with the International Crops Research Institute for the Semi-Arid Tropics (ICRISAT), Patancheru, India, and the International Center for Agricultural Research in the Dry Areas (ICARDA), Aleppo, Syria, in chickpea improvement and released 10 chickpea varieties in Ethiopia. Of these, three (DZ-10-4, DZ-10-11 and Dubie) were developed from its own breeding materials, five (Mariye, Worku, Akaki, Shasho and Chefe) from the breeding materials supplied by ICRISAT, and two (Arerti and Habru) from the breeding materials supplied by ICARDA.........

    Integrated human-animal sero-surveillance of Brucellosis in the pastoral Afar and Somali regions of Ethiopia

    BACKGROUND: Brucellosis is widespread in Ethiopia with variable reported prevalence depending on the geographical area, husbandry practices and animal species. However, there is limited information on the disease prevalence amongst pastoral communities, whose life is intricately linked with their livestock. METHODOLOGY: We conducted an integrated human-animal brucellosis sero-surveillance study in two adjacent pastoral regions, Afar and Somali region (SRS). This cross-sectional study included 13 woredas (districts) and 650 households. Blood samples were collected from people and livestock species (cattle, camel, goats and sheep). Sera were analyzed with C-ELISA for camels and shoats (sheep and goats), with I-ELISA for cattle and IgG ELISA for humans. Descriptive and inferential statistics analyses were performed. RESULTS: A total of 5469 sera were tested by ELISA. Prevalence of livestock was 9.0% in Afar and 8.6% in SRS (ranging from 0.6 to 20.2% at woreda level). In humans, prevalence was 48.3% in Afar and 34.9% in SRS (ranging from 0.0 to 74.5% at woreda level). 68.4% of all households in Afar and 57.5% of households in SRS had at least one animal reactor. Overall, 4.1% of animals had a history of abortion. The proportion of animals with abortion history was higher in seropositive animals than in seronegative animals. Risk factor analysis showed that female animals were significantly at higher risk of being reactors (p = 0.013). Among the species, cattle had the least risk of being reactors (p = 0.014). In humans, there was a clear regional association of disease prevalence (p = 0.002). The older the people, the highest the odds of being seropositive. CONCLUSION: Brucellosis is widespread in humans and animals in pastoral communities of Afar and SRS with the existence of geographical hotspots. No clear association was seen between human and particular livestock species prevalence, hence there was no indication as whether B. abortus or B. melitensis are circulating in these areas, which warrants further molecular research prior to embarking on a national control programs. Such programs will need to be tailored to the pastoral context

    Expression of the transcription factor, TFII-I, during post-implantation mouse embryonic development

    <p>Abstract</p> <p>Background</p> <p>General transcription factor (TFII-I) is a multi-functional transcription factor encoded by the Gtf2i gene, that has been demonstrated to regulate transcription of genes critical for development. Because of the broad range of genes regulated by TFII-I as well as its potential role in a significant neuro-developmental disorder, developing a comprehensive expression profile is critical to the study of this transcription factor. We sought to define the timing and pattern of expression of TFII-I in post-implantation embryos at a time during which many putative TFII-I target genes are expressed.</p> <p>Findings</p> <p>Antibodies to the N-terminus of TFII-I were used to probe embryonic mouse sections. TFII-I protein is widely expressed in the developing embryo. TFII-I is expressed throughout the period from E8-E16. However, within this period there are striking shifts in localization from cytoplasmic predominant to nuclear. TFII-I expression varies in both a spatial and temporal fashion. There is extensive expression in neural precursors at E8. This expression persists at later stages. TFII-I is expressed in developing lung, heart and gut structures. There is no evidence of isoform specific expression. Available data regarding expression patterns at both an RNA and protein level throughout development are also comprehensively reviewed.</p> <p>Conclusions</p> <p>Our immunohistochemical studies of the temporal and spatial expression patterns of TFII-I in mouse embryonic sections are consistent with the hypothesis that hemizygous deletion of <it>GTF2I </it>in individuals with Williams-Beuren Syndrome contributes to the distinct cognitive and physiological symptoms associated with the disorder.</p

    Assessing spatio-temporal rainfall variability in a tropical mountain area (Ethiopia) using NOAA’s rainfall estimates

    Seasonal and interannual variation in rainfall can cause massive economic loss for farmers and pastoralists, not only because of deficient total rainfall amounts but also because of long dry spells within the rainy season. The semi-arid to sub-humid mountain climate of the North Ethiopian Highlands is especially vulnerable to rainfall anomalies. In this article, spatio-temporal rainfall patterns are analysed on a regional scale in the North Ethiopian Highlands using satellite-derived rainfall estimates (RFEs). To counter the weak correlation in the dry season, only the rainy season rainfall from March till September is used, responsible for approximately 91% of the annual rainfall. Validation analysis demonstrates that the RFEs are well correlated with the meteorological station (MS) rainfall data, i.e. 85% for RFE 1.0 (1996-2000) and 80% for RFE 2.0 (2001-2006). However, discrepancies indicate that RFEs generally underestimate MS rainfall and the scatter around the trendlines indicates that the estimation by RFEs can be in gross error. A local calibration of RFE with rain gauge information is validated as a technique to improve RFEs for a regional mountainous study area. Slope gradient, slope aspect, and elevation have no added value in the calibration of the RFEs. The estimation of monthly rainfall using this calibration model improved on average by 8%. Based upon the calibration model, annual rainfall maps and an average isohyet map for the period 1996-2006 were constructed. The maps show a general northeast-southwest gradient of increasing rainfall in the study area and a sharp east-west gradient in its northern part. Slope gradient, slope aspect, elevation, easting, and northing were evaluated as explanatory factors for the spatial variability of annual rainfall in a stepwise multiple regression with the calibrated average of RFE 1.0 as dependent variable. Easting and northing are the only significant contributing variables (R-2=0.86), of which easting has proved to be the most important factor (R-2=0.72). The scatter around the individual trendlines of easting and northing corresponds to an increase in rainfall variability in the drier regions. Despite the remaining underestimation of rainfall in the southern part of the study area, the improved estimation of spatio-temporal rainfall variability in a mountainous region by RFEs is valuable as input to a wide range of scientific models
