    The ULK3 Kinase Is Critical for Convergent Control of Cancer-Associated Fibroblast Activation by CSL and GLI.

    The connection between signaling pathways activating cancer-associated fibroblasts (CAFs) remains to be determined. Metabolic alterations linked to autophagy have also been implicated in CAF activation. CSL/RBPJ, a transcriptional repressor that mediates Notch signaling, suppresses the gene expression program(s), leading to stromal senescence and CAF activation. Deregulated GLI signaling can also contribute to CAF conversion. Here, we report that compromised CSL function depends on GLI activation for conversion of human dermal fibroblasts into CAFs, separately from cellular senescence. Decreased CSL upregulates the expression of the ULK3 kinase, which binds and activates GLI2. Increased ULK3 also induces autophagy, which is unlinked from GLI and CAF activation. ULK3 upregulation occurs in the CAFs of several tumor types, and ULK3 silencing suppresses the tumor-enhancing properties of these cells. Thus, ULK3 links two key signaling pathways involved in CAF conversion and is an attractive target for stroma-focused anti-cancer intervention

    Combined use of x-ray fluorescence microscopy, phase contrast imaging for high resolution quantitative iron mapping in inflamed cells

    X-ray fluorescence microscopy (XRFM) is a powerful technique to detect and localize elements in cells. To derive information useful for biology and medicine, it is essential not only to localize, but also to map quantitatively the element concentration. Here we applied quantitative XRFM to iron in phagocytic cells. Iron, a primary component of living cells, can become toxic when present in excess. In human fluids, free iron is maintained at 10-18 M concentration thanks to iron binding proteins as lactoferrin (Lf). The iron homeostasis, involving the physiological ratio of iron between tissues/secretions and blood, is strictly regulated by ferroportin, the sole protein able to export iron from cells to blood. Inflammatory processes induced by lipopolysaccharide (LPS) or bacterial pathoge inhibit ferroportin synthesis in epithelial and phagocytic cells thus hindering iron export, increasing intracellular iron and bacterial multiplication. In this respect, Lf is emerging as an important regulator of both iron and inflammatory homeostasis. Here we studied phagocytic cells inflamed by bacterial LPS and untreated or treated with milk derived bovine Lf.     On the solar origin of the 200 y Suess wiggles: Evidence from thermoluminescence in sea sediments

    In order to understand the origin of the Suess wiggles, present in the tree ring 14C record, we have studied the thermoluminescence (TL) of a shallow-water Ionian sea core, in an attempt to provide a record of an indicator responding to the solar output more promptly than 14C in tree rings. The spectral content of the TL and radiocarbon records is very similar; neverthless, in addition to the 207 y Suess wave (A43.6% ), the TL record shows the second harmonic (103.5 y; A42.4% ), in phase with the envelope of the 11 y solar cycles. This result, obtained by different spectral methods, in particular by Superposition of Epochs (SE) and by Singular Spectrum Analysis (SSA), strongly supports the solar origin of the 200 y Suess wiggles

    During 2010-2011, the Medicina 32-m dish hosted the 7-feed 18-26.5 GHz receiver built for the Sardinia Radio Telescope, with the goal to perform its commissioning. This opportunity was exploited to carry out a pilot survey at 20 GHz over the area for {\delta} > + 72.3{\deg}. This paper describes all the phases of the observations, as they were performed using new hardware and software facilities. The map-making and source extraction procedures are illustrated. A customised data reduction tool was used during the follow-up phase, which produced a list of 73 confirmed sources down to a flux density of 115 mJy. The resulting catalogue, here presented, is complete above 200 mJy. Source counts are in agreement with those provided by the AT20G survey. This pilot activity paves the way to a larger project, the K-band Northern Wide Survey (KNoWS), whose final aim is to survey the whole Northern Hemisphere down to a flux limit of 50 mJy (5{\sigma}).Comment: 10 pages, 10 figures.     Uso de leguminosas na fitorremediação de solo contaminado com sulfentrazone.

    RESUMO: Resíduos de sulfentrazone podem permanecer no solo por cerca de dois anos, oferecendo risco de contaminação a lençóis freáticos e possibilitando o cultivo de espécies susceptíveis. Diante disto, a fitorremediação pode representar uma opção para a descontaminação de áreas que sofreram com intensas aplicações deste herbicida. O presente trabalho avaliou o potencial remediador das espécies Crotalaria juncea, Canavalia ensiformis, Cajanus cajan e Cajanus cajan (anão), com relação ao herbicida sulfentrazone, utilizando-se o milheto (Pennisetum glaucum) como planta bioindicadora. O experimento foi instalado em casa-de-vegetação e os tratamentos compostos pela combinação entre as quatro espécies citadas, além da testemunha, e quatro doses de sulfentrazone (0 g i.a. ha-1, 200 g i.a. ha-1, 400 g i.a. ha-1 e 800 g i.a. ha-1). Foram avaliadas a altura de plantas, fitotoxicidade ao sulfentrazone e biomassa fresca e seca da parte aérea. Quando P. glaucum foi cultivado após C. juncea, apresentou maior ganho em biomassa e maior altura, e os sintomas de fitotoxicidade foram menos acentuados. C. juncea apresentou a maior capacidade de fitorremediar solos contaminados com sulfentrazone.ABSTRACT: Use of leguminous plants for phytoremediation of soil contaminated with sulfentrazone. Residual sulfentrazone may remain in the soil for around two years, being a source of groundwater contamination and preventing the growth of usceptible plants. So, phytoremediation may represent an option for decontaminating areas which were highly affected by applications of this herbicide. This study aimed at evaluating the potential of Crotalaria juncea, Canavalia ensiformis, Cajanus cajan and Cajanus cajan (dwarf) for remediation of soil contaminated with sulfentrazone, using millet (Pennisetum glaucum) as a bioindicator plant. The experiment was installed in a greenhouse and treatments consisted of the four species already mentioned, plus a control, and four sulfentrazone doses (0 g a.i. ha-1, 200 g a.i. ha-1, 400 g a.i. ha-1 and 800 g a.i. ha-1). Plants height, phytotoxicity to sulfentrazone and fresh and dry matter were evaluated. When P. glaucum was grown after C. juncea, the fresh and dry matter, as well as plant height, were higher, while the phytotoxicity symptoms were less evident. C. juncea showed the best results for phytoremediation of soils contaminated with sulfentrazone

    O sistema denominado de Plantio Cruzado vem sendo reportado na mídia relacionada ao setor agrícola como uma tecnologia capaz de proporcionar incrementos significativos na produtividade da cultura da soja. A motivação para essas divulgações tem como pano de fundo os altos rendimentos obtidos por alguns agricultores que adotaram tal sistema em parte de suas propriedades. A partir desse cenário, objetivou-se com o trabalho avaliar o crescimento e a produtividade da cultivar de soja de hábito indeterminado BRS 359 RR, cultivada no sistema de plantio cruzado e não cruzado. O experimento foi realizado na Fazenda Experimental da Embrapa Soja em Londrina-PR, durante o período de outubro de 2011 a março de 2012. Utilizou-se o delineamento de blocos ao acaso em esquema fatorial 2 x 2 x 2, com três repetições. Os tratamentos foram formados pela combinação de dois espaçamentos entre fileiras (0,4 e 0,6 m), duas densidades de semeadura (375.000 e 562.500 sementes ha-1) e dois sistemas de plantio (plantio cruzado e não cruzado). O crescimento e a produtividade de grãos da cultivar de soja de hábito indeterminado BRS 359 RR não foi afetada pelo plantio cruzado, o qual reduziu a densidade de plantas na colheita

    Costs of managing adverse events in the treatment of first-line metastatic renal cell carcinoma: Bevacizumab in combination with interferon-α2a compared with sunitinib

    Background: Bevacizumab plus interferon-α2a (IFN) prolongs progression-free survival to>10 months, which is comparable with sunitinib as first-line treatment of metastatic renal cell carcinoma (RCC). The two regimens have different tolerability profiles; therefore, costs for managing adverse events may be an important factor in selecting therapy.Methods: Costs of managing adverse events affecting patients with metastatic RCC eligible for treatment with bevacizumab plus IFN or sunitinib were evaluated using a linear decision analytical model. Management costs were calculated from the published incidence of adverse events and health-care costs for treating adverse events in the United Kingdom, Germany, France and Italy.Results: Adverse event management costs were higher for sunitinib than for bevacizumab plus IFN. The average cost per patient for the management of grade 3-4 adverse events was markedly lower with bevacizumab plus IFN compared with sunitinib in the United Kingdom (\[euro]1475 vs \[euro]804), Germany (\[euro]1785 vs \[euro]1367), France (\[euro]2590 vs \[euro]1618) and Italy (\[euro]891 vs \[euro]402). The main cost drivers were lymphopaenia, neutropaenia, thrombocytopaenia, leucopaenia and fatigue/asthaenia for sunitinib; and proteinuria, fatigue/asthaenia, bleeding, anaemia and gastrointestinal perforation for bevacizumab plus IFN.Conclusion: The costs of managing adverse events are lower for bevacizumab plus IFN than for sunitinib. The potential for cost savings should be considered when selecting treatments for RCC
