Developing a strawberry yogurt fortified with marine fish oil \u3csup\u3e1\u3c/sup\u3e
Fortified dairy products appeal to a wide variety of consumers and have the potential to increase sales in the yogurt industry and help increase intake of long-chain n-3 fatty acids. The objectives of this study were to develop a strawberry yogurt containing microencapsulated salmon oil (MSO; 2% wt/vol) and evaluate its characteristics during 1 mo of storage. Unpurified salmon oil (USO) was purified (PSO) and both USO and PSO were analyzed for peroxide value (PV), anisidine value (AV), total oxidation, free fatty acids (FFA), and moisture content. A stable emulsion was prepared with 7% PSO, 22% gum arabic, 11% maltodextrin, and 60% water. The emulsion was spray-dried to produce MSO. The MSO was added to strawberry-flavored yogurt (SYMSO) before pasteurization and homogenization, and a control (SY) without MSO was produced. Both yogurts were stored for 1 mo at 4°C and we determined the quality characteristics including acidity (pH), syneresis, thiobarbituric acid (TBA), fatty acid methyl ester composition, color, and lactic acid bacteria (LAB) count. The entire experiment was replicated 3 times. Total oxidation (unitless) of USO, PSO, and MSO was calculated to be 20.7 ± 1.26, 10.9 ± 0.1, and 13.4 ± 0.25, respectively. Free fatty acid contents were 1.61 ± 0.19%, 0.59 ± 0.02%, and 0.77 ± 0.02% for USO, PSO, and MSO, respectively. Eicosapentaenoic acid and docosahexaenoic acid were the predominant polyunsaturated fatty acids in MSO and in SYMSO, but neither was detected in SY. Fortification of SY with MSO had no significant effect on yogurt pH or syneresis. A decrease in concentration of lactic acid bacteria was observed during the storage of all yogurts. Thiobarbituric acid values significantly increased as storage time increased and SY had a significantly lighter (higher L *) and less yellow (lower b *) color than SYMSO. Although some slight differences were observed in the color and oxidation of SYMSO compared with SY, the study demonstrated that SY could be fortified with salmon oil. © 2011 American Dairy Science Association
Discovery of SMAD4 promoters, transcription factor binding sites and deletions in juvenile polyposis patients
Inactivation of SMAD4 has been linked to several cancers and germline mutations cause juvenile polyposis (JP). We set out to identify the promoter(s) of SMAD4, evaluate their activity in cell lines and define possible transcription factor binding sites (TFBS). 5′-rapid amplification of cDNA ends (5′-RACE) and computational analyses were used to identify candidate promoters and corresponding TFBS and the activity of each was assessed by luciferase vectors in different cell lines. TFBS were disrupted by site-directed mutagenesis (SDM) to evaluate the effect on promoter activity. Four promoters were identified, two of which had significant activity in several cell lines, while two others had minimal activity. In silico analysis revealed multiple potentially important TFBS for each promoter. One promoter was deleted in the germline of two JP patients and SDM of several sites led to significant reduction in promoter activity. No mutations were found by sequencing this promoter in 65 JP probands. The predicted TFBS profiles for each of the four promoters shared few transcription factors in common, but were conserved across several species. The elucidation of these promoters and identification of TFBS has important implications for future studies in sporadic tumors from multiple sites, and in JP patients

Antiulcer, wound healing and hepatoprotective activities of the seaweeds Gracilaria crassa, Turbinaria ornata and Laurencia papillosa from the southeast coast of India
Seaweeds have bioactive compounds of interest in the pharmaceutical industry. In India, seaweeds are used exclusively for phycocolloids production and have not yet received consideration as a dietary supplement. So, it has become imperative to explore the biomedical potential of seaweeds and promote their utilization as a functional food. The seaweeds Turbinaria ornata, Gracillaria crassa and Laurencia papillosa, collected from the Tuticorin coast of the Southeast coast of India and selected based on preliminary screening, were extracted with acetone and evaluated for antiulcer, wound healing and hepatoprotective activities. L. papillosa showed the highest level of gastric protection activity (81%) at 200 mg/kg, comparable to the standard drug ranitidine (90%). G. crassa followed with 76%. G. crassa and L. papillosa, showed marked wound-healing activity. G. crassa at 200 mg/kg, showed a marked effect on the serum marker enzymes indicating prominent hepatoprotective activity. The noteworthy wound-healing and hepato-protective properties of G. crassa besides anti-ulcer activity next to L. papillosa were indicative of its potential for further consideration

Robust estimation of bacterial cell count from optical density
Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data

peer reviewedThis study presents data from an in-depth proximate compositional analysis of three marine fish species: blue
whiting (Micromesistius poutassou), boarfish (Capros aper) and Atlantic herring (Clupea harengus). These fish
contained significant amounts of protein (16–17%), lipids (4–11%) and minerals (2–6% ash). The proteins, particularly
from boarfish, had close to optimum amino acid profiles for human and fish nutrition. They compared favourably
with other fish species in terms of total lipids and relative concentration of the omega-3 fatty acids docosahexaenoic
acid and eicosapentaenoic acid (11.8–13.3% and 5.9–8.1% in triacylglycerols [TG] and 24.6–35.4% and 5.8–12.0% in
phospholipids [PL]). Atlantic herring had the highest lipid content among the three fish and was found to contain
high levels of PL poly-unsaturated fatty acids, including omega-3 fatty acids. Minerals detected in the fish included
calcium (272–1,520 mg/100 g), phosphorus (363–789 mg/100 g), iron (1.07–2.83 mg/100 g), magnesium (40.70–62.10
mg/100 g), potassium (112.00–267.00 mg/100 g), selenium (0.04–0.06 mg/100 g), sodium (218.00–282.00 mg/100 g)
and zinc (1.29–5.57 mg/100 g). Boarfish had the highest ash fraction and also the highest levels of all the minerals,
except potassium. Atlantic herring had considerably lower mineral content compared with the other two species
and, levels detected were also lower than those reported in previously published studies. Heavy metals contents
were quantified, and levels were significantly below the maximum allowable limits for all elements except arsenic,
which ranged from 1.34 to 2.44 mg/kg in the three fish species. Data outlined here will be useful for guiding product
development. Future studies would benefit from considering catch season, sex and developmental stage of the fish
