    Extending the hydrophobic mismatch concept to amphiphilic membranolytic peptides

    A series of nine amphiphilic, pore-forming α-helical KIA peptides (KIAGKIA repeats) with lengths between 14 and 28 residues were studied by solidstate 15N NMR to determine their alignment in oriented lipid bilayers. In a 2:1 mixture of 1,2-dimyristoyl-sn-glycero-3-phosphatidylcholine (DMPC) with its corresponding 1- myristoyl-2-hydroxy-sn-glycero-3-phosphocholine (lyso-MPC), which has a highly positive spontaneous curvature, the helix tilt angle was found to vary steadily with peptide length. The shortest peptide was aligned transmembrane and upright, while the longer ones successively became tilted away from the membrane normal. This behavior is in agreement with the hydrophobic matching concept, conceived so far only for hydrophobic helices. In 1,2-dioleoyl-sn-glycero-3-phosphatidylcholine, with a negative spontaneous curvature, all KIA peptides remained flat on the bilayer surface, while the cylindrical DMPC lipids permitted a slight tilt. Peptide insertion thus depends critically on the intrinsic lipid curvature, and helix orientation is then fine-tuned by membrane thickness. A refined toroidal pore model is proposed

    Hydrophobic mismatch demonstrated for membranolytic peptides and their use as molecular rulers to measure bilayer thickness in native cells

    Hydrophobic mismatch is a well-recognized principle in the interaction of transmembrane proteins with lipid bilayers. This concept was extended here to amphipathic membranolytic α-helices. Nine peptides with lengths between 14 and 28 amino acids were designed from repeated KIAGKIA motifs, and their helical nature was confirmed by circular dichroism spectroscopy. Biological assays for antimicrobial activity and hemolysis, as well as fluorescence vesicle leakage and solid-state NMR spectroscopy, were used to correlate peptide length with membranolytic activity. These data show that the formation of transmembrane pores is only possible under the condition of hydrophobic matching: the peptides have to be long enough to span the hydrophobic bilayer core to be able to induce vesicle leakage, kill bacteria, and cause hemolysis. By correlating the threshold lengths for biological activity with the biophysical results on model vesicles, the peptides could be utilized as molecular rulers to measure the membrane thickness in different cells

    Measurements of proton induced reaction cross sections on 120Te for the astrophysical p-process

    The total cross sections for the 120Te(p,gamma)121I and 120Te(p,n)120I reactions have been measured by the activation method in the effective center-of-mass energies between 2.47 MeV and 7.93 MeV. The targets were prepared by evaporation of 99.4 % isotopically enriched 120Te on Aluminum and Carbon backing foils, and bombarded with proton beams provided by the FN tandem accelerator at the University of Notre Dame. The cross sections and SS factors were deduced from the observed gamma ray activity, which was detected off-line by two Clover HPGe detectors mounted in close geometry.     Gait and Force Analysis of Provoked Pig Gait on Clean and Fouled Concrete Surfaces

    Gait and force analysis have proven to be useful methods in linking claw injuries to surface material conditions. To determine the relationship between claw disorder and floor properties such as friction and surface abrasiveness, the factors controlling gait must be characterised. The effects of fouled concrete floor conditions on the gait of 10 pigs walking in a curve, using kinematics and kinetics to record gait parameters and slip frequency are described and compared with clean conditions. Pigs adapted to fouled floor conditions by reducing their walking speed and stride length, using a higher number of 3-foot support phases and by lowering diagonality. This adaption produced lower vertical forces, a twofold reduction in propulsion and outward stabilisation force and a threefold increase in braking force, without reducing the peak utilised coefficient of friction (UCOF). The UCOF values for both limbs of the curve walking pigs exceeded the recorded dynamic coefficient of friction and the corresponding UCOF values for pigs walking a straight line in fouled floor condition. As UCOF increased and available friction from the fouled floor surface decreased, this resulted in higher forward and backward slip frequency in both limbs for pigs walking in a curve. Pigs provoked to walk in a curve can adapt to fouled floor condition, but if the floor is heavily fouled this adaption is not sufficient to ensure safe walking

    Measurement of the lifetime of the B_c meson in the semileptonic decay channel

    Using approximately 1.3 fb-1 of data collected by the D0 detector between 2002 and 2006, we measure the lifetime of the B_c meson in the B_c -> J/psi mu nu X final state. A simultaneous unbinned likelihood fit to the J/\psi+mu invariant mass and lifetime distributions yields a signal of 881 +/- 80 (stat) candidates and a lifetime measurement of \tau(B_c) = 0.448 +0.038 -0.036 (stat) +/- 0.032 (syst) ps.Comment: 7 pages, 2 figures, submitted to Phys.     Measurement of the electron charge asymmetry in ppbar->W+X->enu+X events at sqrt{s}=1.96 TeV

    We present a measurement of the electron charge asymmetry in ppbar->W+X->enu+X events at a center of mass energy of 1.96 TeV using 0.75 fb-1 of data collected with the D0 detector at the Fermilab Tevatron Collider. The asymmetry is measured as a function of the electron transverse momentum and pseudorapidity in the interval (-3.2, 3.2) and is compared with expectations from next-to-leading order calculations in perturbative quantum chromodynamics.     A search for the standard model Higgs boson in the missing energy and acoplanar b-jet topology at sqrt(s) = 1.96 TeV

    We report a search for the standard model Higgs boson in the missing energy and acoplanar b-jet topology, using an integrated luminosity of 0.93 inverse femtobarn recorded by the D0 detector at the Fermilab Tevatron Collider. The analysis includes signal contributions from pp->ZH->nu nu b b, as well as from WH production in which the charged lepton from the W boson decay is undetected. Neural networks are used to separate signal from background. In the absence of a signal, we set limits on the cross section of pp->VH times the branching ratio of H->bb at the 95% C.L. of 2.6 - 2.3 pb, for Higgs boson masses in the range 105 - 135 GeV, where V=W,Z. The corresponding expected limits range from 2.8 pb - 2.0 pb.Comment: Submitted to Phys. Rev. Letter
