Community groups often have a problem coming to a decision about projects to undertake. Quite often, resources, both human and financial, are limited; the number of problems seem overwhelming; or there are forceful advocates of a "pet" project. How can a group overcome these difficulties? The four decision-making techniques shown in this guide will make the task easier. Sustainable regeneration : everyday landscapes of food acquisition, Pendleton
The report is structured as follows. Chapter two provides the context to the research, outlining why a study of food acquisition and digital inclusion is necessary in Pendleton at this time, and why both issues are linked to the current regeneration programme. Chapter three sets out the methodology employed along with details of the recruitment of participants. Chapter four provides a detailed analysis of the findings from the digital skills side of the research including: digital usage, confidence and competence, and digital skills and shopping, and chapter five focuses on findings related to the food landscape. Chapter six summarises the key findings by identifying what is going well, making recommendations for changes at a variety of scale, as well as specific recommendations for the on-going regeneration programme, and recommendations for further research
Modelling-based evaluation of the costs, benefits and cost-effectiveness of multipathogen point-of-care tests for sexually transmitted infections in symptomatic genitourinary medicine clinic attendees
Objectives To quantify the costs, benefits and cost-effectiveness of three multipathogen point-of-care (POC) testing strategies for detecting common sexually transmitted infections (STIs) compared with standard laboratory testing.
Design Modelling study.
Setting Genitourinary medicine (GUM) services in England.
Population A hypothetical cohort of 965 988 people, representing the annual number attending GUM services symptomatic of lower genitourinary tract infection.
Interventions The decision tree model considered costs and reimbursement to GUM services associated with diagnosing and managing STIs. Three strategies using hypothetical point-of-care tests (POCTs) were compared with standard care (SC) using laboratory-based testing. The strategies were: A) dual POCT for Chlamydia trachomatis (CT) and Neisseria gonorrhoeae (NG); B) triplex POCT for CT-NG and Mycoplasma genitalium (MG); C) quadruplex POCT for CT-NG-MG and Trichomonas vaginalis (TV). Data came from published literature and unpublished estimates.
Primary and secondary outcome measures Primary outcomes were total costs and benefits (quality-adjusted life years (QALYs)) for each strategy (2016 GB, £) and associated incremental cost-effectiveness ratios (ICERs) between each of the POC strategies and SC. Secondary outcomes were inappropriate treatment of STIs, onward STI transmission, pelvic inflammatory disease in women, time to cure and total attendances.
Results In the base-case analysis, POC strategy C, a quadruplex POCT, was the most cost-effective relative to the other strategies, with an ICER of £36 585 per QALY gained compared with SC when using microcosting, and cost-savings of £26 451 382 when using tariff costing. POC strategy C also generated the most benefits, with 240 467 fewer clinic attendances, 808 fewer onward STI transmissions and 235 135 averted inappropriate treatments compared with SC.
Conclusions Many benefits can be achieved by using multipathogen POCTs to improve STI diagnosis and management. Further evidence is needed on the underlying prevalence of STIs and SC delivery in the UK to reduce uncertainty in economic analyses
How to ask sensitive questions in conservation: A review of specialized questioning techniques
Tools for social research are critical for developing an understanding of conservation problems and assessing the feasibility of conservation actions. Social surveys are an essential tool frequently applied in conservation to assess both people’s behaviour and to understand its drivers. However, little attention has been given to the weaknesses and strengths of different survey tools. When topics of conservation concern are illegal or otherwise sensitive, data collected using direct questions are likely to be affected by non-response and social desirability biases, reducing their validity. These sources of bias associated with using direct questions on sensitive topics have long been recognised in the social sciences but have been poorly considered in conservation and natural resource management.
We reviewed specialized questioning techniques developed in a number of disciplines specifically for investigating sensitive topics. These methods ensure respondent anonymity, increase willingness to answer, and critically, make it impossible to directly link incriminating data to an individual. We describe each method and report their main characteristics, such as data requirements, possible data outputs, availability of evidence that they can be adapted for use in illiterate communities, and summarize their main advantages and disadvantages. Recommendations for their application in conservation are given. We suggest that the conservation toolbox should be expanded by incorporating specialized questioning techniques, developed specifically to increase response accuracy. By considering the limitations of each survey technique, we will ultimately contribute to more effective evaluations of conservation interventions and more robust policy decisions
Structure and function of the Rad9-binding region of the DNA-damage checkpoint adaptor TopBP1
TopBP1 is a scaffold protein that coordinates activation of the DNA-damage-checkpoint response by coupling binding of the 9-1-1 checkpoint clamp at sites of ssDNA, to activation of the ATR-ATRIP checkpoint kinase complex. We have now determined the crystal structure of the N-terminal region of human TopBP1, revealing an unexpected triple-BRCT domain structure. The arrangement of the BRCT domains differs significantly from previously described tandem BRCT domain structures, and presents two distinct sites for binding phosphopeptides in the second and third BRCT domains. We show that the site in the second but not third BRCT domain in the N-terminus of TopBP1, provides specific interaction with a phosphorylated motif at pSer387 in Rad9, which can be generated by CK2
Conductivity and quasinormal modes in holographic theories
We show that in field theories with a holographic dual the retarded Green's
function of a conserved current can be represented as a convergent sum over the
quasinormal modes. We find that the zero-frequency conductivity is related to
the sum over quasinormal modes and their high-frequency asymptotics via a sum
rule. We derive the asymptotics of the quasinormal mode frequencies and their
residues using the phase-integral (WKB) approach and provide analytic insight
into the existing numerical observations concerning the asymptotic behavior of
