Creating ex situ protected bioreservoirs as a powerful strategy for the reproductive biotechnology-mediated rescue of threatened Polish livestock breeds

The current article presents the state of the art of the creation of bioreservoirs of cryopreserved somatic and stem cell lines and cryopreserved or lyophilized germplasm-based resources of selected farm animal species. It also presents the agricultural, biological and biotechnological determinants, and showcases the importance of the National Research Institute of Animal Production (NRIAP) in Poland in this process. The aforementioned bioreservoirs serve as an innovative research tool used for cryogenically or lyophilizogenically assisted and species-specific ex situ conservation. The latter ensures, in the long term, not only restitution, but also perpetuation of sustainable biodiversity that underlies genotypic and phenotypic, intra- and inter-population variability within pure-breeding herds of the national conserved livestock breeds

Praktická část se zabývá vytvořením samotného školení. Začíná průzkumem spokojenosti klien-tů společnosti MKMA s.r.o. se stávající formou produktového školení, následuje vyhodnocení dotazníku a návrh na změnu průběhu. Poslední kapitola bakalářské práce zahrnuje vytvoření sportovního kempu s přednáškami. Porovnává tři lokality, pro které je vytvořena kalkulace, denní rozpis a doporučuje nejvýhodnější z nich.The goal of this thesis is to create interesting and entertaining training for retailers in order to introduce them outcomming products. The thesis deals with theory of correct communication with employees, procedure how to pro-perly prepare a presentation and how to present it to the audience and theory of customer ser-vice. Práce v první kapitole obsahuje popis různých standardů sítí včetně ukázky jejích hardwarové konfigurace. Druhá kapitola pojednává o jednotlivých programovacích jazycích obsažených v prostředí STEP7 a demonstruje jejich užití na cvičných úlohách.This thesis deals with software development kit STEP7 and its posibilities in using different networking and programing options. Design and Optimization of Tool Production for Shaping Specific Internal Gearing

Tato bakalářská práce se zabývá problematikou návrhu a výroby nástrojů. Hlavním cílem této práce je navrhnout nástroj pro vnitřní spirálové ozubení, navrhnout technologický postup výroby nože a optimalizovat jeho výrobu. Práce obsahuje čtyři hlavní části. První část je rešeršní a zabývá se problematikou návrhu a výrobou nástrojů. Tato část je zaměřena na jednotlivé části konstrukce nástroje a možnosti jeho výroby. V druhé části je návrh a technologický postup výroby monolitního obrážecího nože. V třetí části je optimalizace nástroje, pojata z hlediska konstrukce. Je zde navrhnuto těleso nástroje pro vyměnitelnou břitovou destičku a taktéž vytvořen technologický postup. Poslední část obsahuje technicko-ekonomické zhodnocení, ve kterém jsou porovnány oba typy nástrojů.This Bachelor Thesis deals with the design and production of tools. The main goal of this work is to design a tool for internal spiral gearing, to design the technological process of tool production and to optimize production. The work contains four main parts. The first part is a research and deals with the issue of design and production of tools. This part is focused on individual parts of the tool construction and the possibilities of its production. Diplomová práce analyzuje dopad osobnosti vedoucího pracovníka na podnikovou kulturu pomocí dotazníkového šetření a na základě těchto výsledků navrhuje komunikační plán. Tento plán byl navrhnut zejména pro problematické oblasti, kterými jsou nedostatečná komunikace firemní vize a slabá informovanost zaměstnanců o aktuálním dění. Během průzkumu bylo zjištěno, že zaměstnanci zkoumané společnosti mají vysokou úroveň oddanosti i angažovanosti a že na hodnocení schopností generálního ředitele má vliv typ osobnosti jeho zaměstnanců.The diploma thesis analyzes the impact of a manager's personality on corporate culture by using a questionnaire survey and based on these results proposes a communication plan. This plan was designed especially for problematic areas, which are insufficient communication of the company's vision and week information knowledge of employees about current situation. During the survey, was found that the employees of the surveyed company have a high level of devotion and commitment and that the evaluation of the CEO's abilities is influenced by the personality type of his employees

Anabolic steroids activate the NF-KB pathway in porcine ovarian putative stem cells independently of the ZIP-9 receptor

Boldenone (Bdn) and nandrolone (Ndn) are anabolic androgenic steroids (AASs) that, as our previous studies have shown, may increase the risk of neoplastic transformation of porcine ovarian putative stem cells (poPSCs). The NF-κB pathway may be important in the processes of carcinogenesis and tumour progression. Therefore, in this work, we decided to test the hypothesis of whether Bdn and Ndn can activate the NF-κB pathway by acting through the membrane androgen receptor ZIP-9. For this purpose, the expression profiles of both genes involved in the NF-κB pathway and the gene coding for the ZIP-9 receptor were checked. The expression and localization of proteins of this pathway in poPSCs were also examined. Additionally, the expression of the ZIP-9 receptor and the concentration of the NF-κB1 and 2 protein complex were determined. Activation of the NF-κB pathway was primarily confirmed by an increase in the relative abundances of phosphorylated forms of RelA protein and IκBα inhibitor. Reduced quantitative profiles pinpointed not only for genes representing this pathway but also for unphosphorylated proteins, and, simultaneously, decreased concentration of the NF-κB1 and 2 complex may indicate post-activation silencing by negative feedback. However, the remarkably and sustainably diminished expression levels noticed for the SLC39A9 gene and ZIP-9 protein suggest that this receptor does not play an important role in the regulation of the NF-κB pathway
