Assistência de enfermagem às famílias de indivíduos que tentaram suicídio Asistencia de enfermería a la familia durante la atención de un miembro que intentó suicidio Nursing assistance to a family of a member who attempted suicide
Estudo qualitativo que objetivou conhecer a assistência de enfermagem oferecida aos familiares de indivíduos que tentaram suicídio, durante o atendimento inicial da ocorrência. Os dados foram coletados, em maio e junho de 2008, de 21 enfermeiros atuantes em Unidades de Atenção às Urgências, de três cidades do Sul do Brasil, por meio de entrevista semiestruturada. Na análise emergiram duas categorias: "A prática profissional e o cuidado à família do indivíduo que tentou suicídio" e "Emergindo as emoções no viver profissional", as quais revelam que as famílias normalmente são procuradas para informações sobre o caso, mas raramente constituem foco da assistência. Os profissionais mostram-se descontentes com esta situação, contudo percebem-se limitados, embora acreditem que maior atenção instituiria medidas eficazes para um cuidado humanizado. Estes resultados podem subsidiar a atuação dos enfermeiros com vistas a uma assistência que considere a pessoa e a família de forma holística.<br>Estudio cualitativo que tuvo como objetivo conocer las acciones desempeñadas por los enfermeros junto a los familiares durante la atención inicial de un miembro que intentó suicidio. Los datos fueron recogidos en mayo y en junio de 2008, junto a 21 enfermeros actuantes en Unidades de Atención a las Urgencias, de tres ciudades del Sur de Brasil, por medio de entrevista semiestructurada. En el análisis emergieron dos categorías: "La práctica profesional y la atención a la familia del individuo que intentó suicidio" y "Emergiendo las emociones en el vivir profesional", las cuales revelan que las familias normalmente son buscadas para informaciones sobre el caso, pero raramente constituyen foco de la asistencia. Los profesionales se muestran descontentos con esta situación, con todo se perciben limitados, sin embargo creen que estrategias como mayor atención instituirían medidas eficaces para un cuidado humanizado. Estos resultados pueden subsidiar la actuación de los enfermeros con vistas a una asistencia que considere la persona y la familia de manera holística.<br>This qualitative study aimed to identify nursing care offered to families of individuals who attempted suicide. Data was collected in May and June 2008, from 21 active nurses in Emergency Care Units, of three cities in the South of Brazil, through a semi-structured interview. From the analysis two categories emerged: "The professional practice and care to the individual's family attempt suicide " and "Emotions that emerge in the life of a professional", which reveal that the families are usually sought for information on the case, but they rarely constitute focus of the assistance. Professionals appear to be unhappy with this situation. However, perceive themselves limited, although they believe that more attention would institute effective measures to a humanized care. These results can subsidize the performance of nurses with views to an assistance that considers the person and the family in a holistic manner

A novel phospholipase A2, BJ-PLA2, from the venom of the snake Bothrops jararaca: Purification, primary structure analysis, and its characterization as a platelet-aggregation-inhibiting factor
This paper describes the isolation and primary structure analysis of a new phospholipase A2 with platelet-aggregation-inhibiting activity from the venom of Bothrops jararaca. the protein, named BJ-PLA2, was isolated by means of ammonium sulfate precipitation and anion-exchange and reversed-phase chromatographies and behaved as a homogeneous single-chain protein on SDS-PAGE. Its amino acid sequence was determined by N-terminal sequencing and analysis of overlapped chemical and proteolytic fragments by automated Edman degradation and mass spectometry determination. BJ-PLA2 consists of 124 amino acid residues and has the structural features of snake venom class II phospholipases A2, Chemical modification with p-bromophenacylbromide caused complete loss of enzymatic activity and partially affected the platelet-aggregation-inhibiting activity of BJ-PLA2. A prothrombin activator serine protease from the Lonomia obliqua caterpillar venom (Lopap) biochemical characterization
Lonomia obliqua venom causes a severe consumptive coagulopathy, which can lead to a hemorrhagic syndrome. the crude bristles extract displays a procoagulant activity due to a Factor X and to a prothrombin activating activity. Here, we describe a 69 kDa prothrombin activator serine protease purified from L. obliqua caterpillar bristle extract using gel filtration (Sephadex G 75) and HPLC (C-4 column). the purified protein was able to activate prothrombin in a dose-dependent manner, and calcium ions increased this activity. the prothrombin-derived fluorogenic peptide (Abz-YQTFFNPRTGSQ-EDDnp) had its main cleavage site at the Arg-Thr bond. the kinetic parameters obtained for this substrate were K-mapp. of 4.5 muM, k(cat) of 5.32 s(-1), and a k(cat)/K-mapp of 1.2 x 10(6) M-1 s(-1). the prothrombin fragments generated by the purified enzyme corresponded to the molecular masses of prethrombin 2, fragment 1, fragment 2, and thrombin as seen in SDS-PAGE. the thrombin generated was able to clot purified fibrinogen. the partial amino acid sequence of the purified protein, named Lopap (L. obliqua prothrombin activator protease), showed no similarity to any known prothrombin activator. Unraveling the evolutionary origin of the complex Nuclear Receptor Element (cNRE), a cis-regulatory module required for preferential expression in the atrial chamber
Cardiac function requires appropriate proteins in each chamber. Atria requires slow myosin to act as reservoirs, while ventricles demand fast myosin for swift pumping. Myosins are thus under chamber-biased cis-regulation, with myosin gene expression imbalances leading to congenital heart dysfunction. To identify regulatory inputs leading to cardiac chamber-biased expression, we computationally and molecularly dissected the quail Slow Myosin Heavy Chain III (SMyHC III) promoter that drives preferential expression to the atria. We show that SMyHC III gene states are orchestrated by a complex Nuclear Receptor Element (cNRE) of 32 base pairs. Using transgenesis in zebrafish and mice, we demonstrate that preferential atrial expression is achieved by a combinatorial regulatory input composed of atrial activation motifs and ventricular repression motifs. Using comparative genomics, we show that the cNRE might have emerged from an endogenous viral element through infection of an ancestral host germline, revealing an evolutionary pathway to cardiac chamber-specific expression
