    Synthesis and preliminary assessment of the anticancer and Wnt/β-catenin inhibitory activity of small amide libraries of fenamates and profens

    As part of an ongoing program to study the anticancer activity of non-steroidal anti-inflammatory drugs (NSAIDs) through generating diversity libraries of multiple NSAID scaffolds, we synthesized a series of NSAID amide derivatives and screened these sets against three cancer cell lines (prostate, colon and breast) and Wnt/β-catenin signaling. The evaluated amide analog libraries show significant anticancer activity/cell proliferation inhibition, and specific members of the sets show inhibition of Wnt/β-catenin signaling.</p

    Zero power mismatch islanding detection algorithm for hybrid distributed generating system

    Distributed generation is essential for both keeping up with the rising power demand and reducing the amount of money spent on fossil fuels. There is widespread agreement that the world should prioritize the development of renewable energy systems such as wind and solar energy. This study describes the design and utility-grid integration of a hybrid distributed generating system that utilizes photovoltaic and wind-driven permanent magnet synchronous generators (hybrid PMSG-PV systems). To prevent damage to the grid, hybrid distributed generation systems, consumer devices, and line workers must be protected from islanding. Detection of islanding in hybrid DG systems has been suggested using passive islanding and time-spectral analysis. Measuring and amplifying the ripple content present in voltage at point of common coupling (PCC) about 0.4 seconds after the permissible delay time after the circuit breaker opens on the utility grid side is how islanding is discovered using this method. Compared to other methods, the proposed method has smoother islanding detection waveforms owing to increases in both the window size and threshold limit. The suggested method detects islanding in 40 ms and is verified in a variety of non-islanding scenarios, such as fault occurrence, parallel feeder loss, and load shift. In addition, the cost is reduced, the response time is rapid, and there is no non-detection zone (NDZ) when using these methods. Unlike active islanding detection methods, their function is unaffected by the size, quantity, or type of distributed generators linked to the utility grid; hence, there are no power quality concerns

    Social Determinants and Comorbidities in Non-pharmacological Interventions for Chronic Pain in Community- Dwelling Older Adults: A Scoping Review of Randomized Controlled Trials

    Background: Chronic pain frequently exists with other comorbid conditions in older adults, yet little is known about the effectiveness of non-pharmacologic interventions. A scoping review was conducted to provide an overview of the nature of the published evidence on the nonpharmacological interventions for chronic pain in community dwelling older adults with comorbid conditions. Methods: A literature search of relevant databases was conducted using PRISMA guidelines. Studies were included if they were intervention studies, used a randomized controlled trial design, and met criteria of the critical appraisal. Results: Eleven studies were identified as high quality. Most studies did not quantify a comorbidity index and/or burden, nor reported the specific comorbid conditions of participants with chronic pain. In addition, none of the identified studies measured change in participant comorbidity and/or burden. Conclusion: Future research should address the effects of non-pharmacological interventions on pain, comorbidities, and other health outcomes

    Regulation of rDNA Transcription by Proto-Oncogene PELP1

    Proline-, glutamic acid-, and leucine-rich protein (PELP1) is a novel nuclear receptor coregulator with a multitude of functions. PELP1 serves as a scaffolding protein that couples various signaling complexes with nuclear receptors and participates as a transcriptional coregulator. Recent data suggest that PELP1 expression is deregulated in hormonal cancers, and that PELP1 functions as a proto-oncogene; however, the mechanism by which PELP1 promotes oncogenesis remains elusive.Using pharmacological inhibitors, confocal microscopy and biochemical assays, we demonstrated that PELP1 is localized in the nucleolus and that PELP1 is associated with the active ribosomal RNA transcription. Cell synchronization studies showed that PELP1 nucleolar localization varies and the greatest amount of nucleolar localization was observed during S and G2 phases. Using pharmacological compounds and CDK site mutants of PELP1, we found that CDK's activity plays an important role on PELP1 nucleolar localization. Depletion of PELP1 by siRNA decreased the expression of pre-rRNA. Reporter gene assays using ribosomal DNA (pHrD) luc-reporter revealed that PELP1WT but not PELP1MT enhanced the expression of reporter. Deletion of nucleolar domains abolished PELP1-mediated activation of the pHrD reporter. ChIP analysis revealed that PELP1 is recruited to the promoter regions of rDNA and is needed for optimal transcription of ribosomal RNA.Collectively, our results suggest that proto-oncogene PELP1 plays a vital role in rDNA transcription. PELP1 modulation of rRNA transcription, a key step in ribosomal biogenesis may have implications in PELP1-mediated oncogenic functions

    TS fuzzy control of PV assisted single phase three phase induction motor drive for rural pumping applications

    The motor drives for aqua farms and large-scale irrigation system needs a reliable electric drive, which requires the continuous power supply and efficient control. However, the rural single phase power supply is frequently interrupted. Renewable assistance would improve the availability of supply and heuristic control approach improves robustness in control. This paper presents a three phase induction motor drive fed from single phase electric grid with assistance from PV and battery energy storage. TS- fuzzy based direct torque control is employed for robust control during load changes, and the topology, component modelling, front-end converter control, PV interface DC–DC converter control, and inverter control are presented. MATLAB/Simulink is used to simulate the proposed drive system. The performance of the proposed system is validated using simulation data for both steady-state and transient states

    Sexual dimorphism in cancer.

    The incidence of many types of cancer arising in organs with non-reproductive functions is significantly higher in male populations than in female populations, with associated differences in survival. Occupational and/or behavioural factors are well-known underlying determinants. However, cellular and molecular differences between the two sexes are also likely to be important. In this Opinion article, we focus on the complex interplay that sex hormones and sex chromosomes can have in intrinsic control of cancer-initiating cell populations, the tumour microenvironment and systemic determinants of cancer development, such as the immune system and metabolism. A better appreciation of these differences between the two sexes could be of substantial value for cancer prevention as well as treatment

    The antioxidant and antiproliferative activities of methanolic extracts from Njavara rice bran

    <p>Abstract</p> <p>Background</p> <p>Free radical-induced oxidative stress is the root cause for many human diseases. Naturally occurring antioxidant supplements from plants are vital to counter the oxidative damage in cells. The main objective of the present study was to characterize the antioxidant and antiproliferative potential of rice bran extracted from an important Indian rice variety, Njavara and to compare the same with two commercially available basmati rice varieties: Vasumathi, Yamini and a non medicinal variety, Jyothi.</p> <p>Methods</p> <p>Methanolic extracts of rice bran from four varieties; Vasumathi, Yamini, Jyothi and Njavara were used to study their total phenolic and flavonoid contents, <it>in vitro </it>antioxidant activities including total antioxidant activity, scavenging of nitric oxide and 1,1-Diphenyl-2-picrylhydrazyl (DPPH) radical, reducing power and cytotoxic activity in C6 glioma cells. Correlation coefficient and regression analysis were done by using Sigmastat version 3.1 and Stata statistical package respectively.</p> <p>Results</p> <p>Rice bran methanolic extract from Njavara showed the highest antioxidant and cell cytotoxic properties compared to the other three rice varieties. IC<sub>50 </sub>values for scavenging DPPH and nitric oxide were in the range of 30.85-87.72 μg/ml and 52.25-107.18 μg/ml respectively. Total antioxidant activity and reducing power were increased with increasing amounts of the extract. Total phenolic and flavonoid contents were in the range of 3.2-12.4 mg gallic acid-equivalent (GAE)/g bran and 1.68-8.5 mg quercetin-equivalent (QEE)/g bran respectively. IC<sub>50 </sub>values of cytotoxic assay (MTT assay) were 17.53-57.78 μg/ml. Correlation coefficient and regression analysis of phenolic content with DPPH and NO scavenging, MTT (-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl tetrazolium bromide) assay, total antioxidant assay and reducing power showed a highly significant correlation coefficient values (96-99%) and regression values (91-98%).</p> <p>Conclusion</p> <p>The results of the present study show that the crude methanolic extract from Njavara rice bran contains significantly high polyphenolic compounds with superior antioxidant activity as evidenced by scavenging of free radicals including DPPH and NO. Njavara extracts also showed highest reducing power activity, anti-proliferative property in C6 glioma cells. In conclusion, it is conceivable that the Njavara rice variety could be exploited as one of the potential sources for plant - based pharmaceutical products.</p

    Different behaviour of DVL1, DVL2, DVL3 in astrocytoma malignancy grades and their association to TCF1 and LEF1 upregulation

    Key regulators of the Wnt signalling, DVL1, DVL2 and DVL3, in astrocytomas of different malignancy grades were investigated. Markers for DVL1, DVL2 and DVL3 were used to detect microsatellite instability (MSI) and gross deletions (LOH), while immunohistochemistry and immunoreactivity score were used to determine the signal strengths of the three DVL proteins and transcription factors of the pathway, TCF1 and LEF1. Our findings demonstrated that MSI at all three DVL loci was constantly found across tumour grades with the highest number in grade II (P = 0.008). Collectively, LOHs were more frequent in high-grade tumours than in low grade ones. LOHs of DVL3 gene were significantly associated with grade IV tumours (P = 0.007). The results on protein expressions indicated that high-grade tumours expressed less DVL1 protein as compared with low grade ones. A significant negative correlation was established between DVL1 expression and malignancy grades (P < 0.001). The expression of DVL2 protein was found similar across grades, while DVL3 expression significantly increased with malignancy grades (P < 0.001). The signal strengths of expressed DVL1 and DVL3 were negatively correlated (P = 0.002). However, TCF1 and LEF1 were both significantly upregulated and increasing with astrocytoma grades (P = 0.001). A positive correlation was established between DVL3 and both TCF1 (P = 0.020) and LEF1 (P = 0.006) suggesting their joint involvement in malignant progression. Our findings suggest that DVL1 and DVL2 may be involved during early stages of the disease, while DVL3 may have a role in later phases and together with TCF1 and LEF1 promotes the activation of Wnt signalling
