Protective Efficacy of BCG Overexpressing an L,D-Transpeptidase against M. tuberculosis Infection
Background: M. bovis Bacille Calmette-Guérin (BCG), currently the only available vaccine against tuberculosis (TB), fails to adequately protect individuals from active and latent TB infection. New vaccines are desperately needed to decrease the worldwide burden of TB. Methods and Findings: We created a recombinant strain of BCG that overproduces an L,D-transpeptidase in order to alter the bacterial peptidoglycan layer and consequently increase the ability of this immunogen to protect against virulent M. tuberculosis (Mtb). We demonstrate that this novel recombinant BCG protects mice against virulent Mtb at least as well as control BCG, as measured by its ability to reduce bacterial burden in lungs and spleen, reduce lung histopathology, and prolong survival. A nutrient starved recombinant BCG preparation, while offering comparable protection, elicited a response characterized by elevated levels of select Th1 cytokines. Conclusions: Recombinant BCG overexpressing a L,D-transpeptidase that is nutrient starved elicits a stronger Th1 type response and is at least as protective as parent BCG. Results from this study suggest that nutrient starvation treatment of live BCG vaccines should be further investigated as a way to increase host induction of Th-1 related cytokines in the development of experimental anti-TB vaccines
An epidemiological study of respiratory syncytial virus associated hospitalizations in Denmark
Respiratory syncytial virus (RSV) is the most common viral pathogen that causes lower respiratory tract infections in infants. Studies have implicated severe RSV infections early in life as a risk factor for subsequent development of reactive airway disease. We are conducting a study to validate RSV-associated diagnoses in the Danish National Patient Registry, to assess whether the incidence of severe RSV infection is increasing in Denmark, to identify predisposing and protective factors for RSV-associated hospitalization in Denmark, and to examine the association of severe RSV infection with reactive airway disease. The influence of various biological, social and environmental factors on hospitalization for RSV infection will be studied through several population-based registers, including the Danish National Birth Cohort: 'Better health for mothers and children'. The RSV hospitalization cases will be compared with control individuals selected within the same population groups on a case–control or a cohort basis in order to produce estimates of age-adjusted and sex-adjusted relative risks (odds ratio and relative risk) for hospitalization associated with various risk factors. Using register linkage and unique registration of exposures collected through interviews and blood samples from the Danish National Birth Cohort, we will be able to resolve the issues referred to above in a very large sample of Danish children
The hidden architecture of higher education:Building a big data infrastructure for the ‘smarter university’
Universities are increasingly organized and managed through digital data. The collection, processing and dissemination of Higher Education data is enabled by complex new data infrastructures that include both human and nonhuman actors, all framed by political, economic and social contingencies. HE data infrastructures need to be seen not just as technical programs but as practical relays of political objectives to reform the sector. This article focuses on a major active data infrastructure project in Higher Education in the United Kingdom. It examines the sociotechnical networks of organizations, software programs, standards, dashboards and visual analytics technologies that constitute the infrastructure, and how these technologies are fused to governmental imperatives of market reform. The analysis foregrounds how HE is being reimagined through the utopian ideal of the ‘smarter university’ while simultaneously being reformed through the political project of marketization

Looking for love in the student experience
This chapter represents an early attempt to engage and think with the ethos that underpins the Manifesto for a Post-Critical Pedagogy (Hodgson et al. 2017), specifically from a sociology of education perspective. The Manifesto is an exhortation to move beyond the critical by also celebrating what we may love and therefore wish to preserve in education. With this in mind, the author undertakes a re-examination of the literature on the university student experience in the UK. They argue that, outside pedagogical research on students, there are three overlapping but somewhat distinct literatures, each of which focuses primarily on social inequalities, aspects of marketization, or geographies, in relation to higher education. It appears that there are gaps in each of these bodies of scholarship, with some dimensions of identity underrepresented in the first, surprisingly little empirical work in the second, while the third has attracted minimal attention to date. Furthermore, it seems that there is little to love in our current understanding of the student experience as the overwhelming focus is on dysfunctions in and around UK higher education. Applying the tenets of a Post-Critical Pedagogy does suggest that widening our scope would allow for an extension of the literature and a more positive appreciation of the UK student experience. At the same time, though, there may be some aspects of the Manifesto that require minor revisions
